[Mitochondrial beta-oxidation defects].

BACKGROUND: Mitochondrial beta-oxidation of fatty acids is an important source of energy for the cells, especially during fasting. Since 1973 several inherited defects in beta-oxidation have been described. Defects in mitochondrial beta-oxidation are one of the largest groups of inborn errors of metabolism. MATERIAL AND METHODS: This review article is based on the experience of the authors and on literature studies. The authors' experience is from laboratory diagnostics and clinical experience in the departments of medical biochemistry and peadiatrics at our hospital. RESULTS AND INTERPRETAT

Nitric oxide in cardiac transplantation.

The endothelium of coronary arteries has been identified as an important organ locally regulating coronary perfusion and cardiac function by paracrine secretion of nitric oxide (NO) and other vasoactive mediators. Therefore, the established organ procurement in cardiac transplantation using hypothermic storage solutions designed to preserve myocytes but not endothelial cells has to be critically discussed. Heart transplantation is a prestigious high-end treatment for end-stage heart failure patients with promising survival rates: 84% one-year and 65% five-year survival. However, these survival

Oxidative status in rheumatoid arthritis.

The insufficiency of antioxidant defense systems and the acceleration of the oxidative reactions can be results of the pro-oxidant/antioxidant imbalance in rheumatoid arthritis (RA). The aim of our study was to investigate the changes in oxidant status by measuring two different parameters; one was the level of malondialdehyde (MDA) as an index of lipid peroxidation and the other was total oxidative status; we could then compare our results with the antioxidant status, superoxide dismutase (SOD) enyzme activities. All were assessed in 22 patients with active RA and 18 age- and gender-matched c

Oxidants in chronic kidney disease.

Chronic kidney disease is a worldwide public health problem that affects approximately 10% of the US adult population and is associated with a high prevalence of cardiovascular disease and high economic cost. Chronic renal insufficiency, once established, tends to progress to end-stage kidney disease, suggesting some common mechanisms for ultimately causing scarring and further nephron loss. This review defines the term reactive oxygen metabolites (ROM), or oxidants, and presents the available experimental evidence in support of the role of oxidants in diabetic and nondiabetic glomerular disea

Oxidative DNA damage in pterygium.

PURPOSE: Epidemiological evidence suggests that UV irradiation plays the most important role in pterygial formation. The noxious effects of UV irradiation are either directly by a UV phototoxic effect or indirectly by formation of radical oxygen species (ROS). ROS are very harmful to cells, because they injure cellular DNA, proteins, and lipids (called oxidative stress). Among numerous types of oxidative DNA damage, the formation of 8-hydroxydeoxyguanosine (8-OHdG) presents only a minor fraction of UV induced DNA damage, but it is a ubiquitous marker of oxidative stress. If pterygium is relate

Oxidative stress in patients with phenylketonuria.

Phenylketonuria (PKU) is an autossomal recessive disease caused by phenylalanine-4-hydroxylase deficiency, which is a liver-specific enzyme that catalyzes the hydroxylation of l-phenylalanine (Phe) to l-tyrosine (Tyr). The deficiency of this enzyme leads to the accumulation of Phe in the tissues and plasma of patients. The clinical characterization of this disease is mental retardation and other neurological features. The mechanisms of brain damage are poorly understood. Oxidative stress is observed in some inborn errors of intermediary metabolism owing to the accumulation of toxic metabolites

Oxidative stress in the pathogenesis of asthma.

Asthma affects 5% to 10% of the population of the United States. In asthmatics, oxidative stress occurs not only as a result of inflammation but also from environmental exposure to air pollution. The specific localization of antioxidants in the lung and the adaptive changes during asthma underscore the importance of oxidative stress, and therapeutic interventions that decrease exposure to environmental reactive oxygen species or augment endogenous antioxidant defenses might be beneficial as adjunctive therapies in asthmatic patients.

Nitric oxide and tubulointerstitial nephritides.

As part of the exponential growth in our understanding of nitric oxide (NO) in health and disease over the past 2 decades, the kidney has become appreciated as a major site where NO may play a number of important roles. Although earlier work on the kidney focused more on effects of NO at the level of larger blood vessels and glomeruli, there has been a rapidly growing body of work showing critical roles for NO in tubulointerstitial disease. In this review we discuss some of the recent contributions to this important field.

Nitric oxide-still no consensus.

When NO became available for clinical use, it seemed to hold enormous promise for newborn infants with lung disease. With regard to the term and near-term infant, the role of NO seems satisfactorily defined: Near-term and term infants with hypoxic respiratory failure unresponsive to current therapy, excluding infants with diaphragmatic hernia, should have a trial of inhaled nitric oxide. In the preterm infant, the situation is less clear with, as yet, no specific indication identified.

[Nitric oxide and lung diseases].

INTRODUCTION: All three isoforms of nitric oxide synthase (NOS) are identified within various tissues of the respiratory system. ISOFORMS OF NOS: Under physiological conditions, small amounts of NO, produced by constitutive isoforms of NOS, appear to be important in regulation of basal pulmonary vascular tone and in mediating transition from fetal to neonatal circulation; also, NO exerts antiinflammatory actions, and modulates the respiratory smooth muscle tone. NO AND RESPIRATORY TRACT DISEASES: Under pathological conditions, inducible NOS-derived NO may produce lung damage. In addition, inc

Nitric oxide: an antioxidant and neuroprotector.

Indirect evidence, including neuroprotection against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-neurotoxicity by nitric oxide synthase (NOS) inhibitors and resistance of transgenic animals deficient in NOS, is controversial. We have reviewed evidence in favor of oxidative stress during the development of MPTP-neurotoxicity and the influence of antioxidants, including nitric oxide (NO) and NO donors, on MPTP-induced dopaminergic neurotoxicity. Systemic administration of MPTP causes dose-dependent generation of hydroxyl radicals (OH) in vivo in the striatum in mice; OH scavengers protec

Ethanol and oxidative mechanisms in the brain.

There is strong evidence showing that chronic and excessive ethanol consumption may enhance oxidative damage to neurons and result in cell death. Although not yet well understood, ethanol may enhance ROS production in brain through a number of pathways including increased generation of hydroxyethyl radicals, induction of CYP2E1, alteration of the cytokine signaling pathways for induction of iNOS and sPLA(2), and production of prostanoids through the PLA(2)/COX pathways. Since many neurodegenerative diseases are also associated with oxidative and inflammatory mechanisms in the brain, it would b

Nitric oxide in parasitic infections.

Nitric oxide (NO) is implicated as an integral component of the host armament against invading parasites. Strongest evidence has come from laboratory models of protozoan infections. During malaria, toxoplasmosis and leishmaniasis, to name just a few, the preferential production of pro-inflammatory cytokines predisposes to the increased synthesis of NO, which mediates host protection through either direct parasite killing or by limiting parasite growth. More recently, evidence has been put forward for a beneficial role of NO during helminthic infections. In the case of Schistosomiasis mansoni,

Mineralization of CCl4 with copper oxide.

Experimentally, CCl4 was effectively mineralized by CuO to yield stable inorganic species of CO2 and CuCl2 (CCl4 + 2CuO --> 2CuCl2 + CO2). High CCl4 conversions (63-83%) were found in the mineralization process performed at 513-603 K for 10-30 min. Using X-ray-absorption near edge structure (XANES) and X-ray photoelectron spectroscopies, we found that most CuCl2 was encapsulated in the CCl4-mineralized product solid (mineralization at 513 K for 30 min). At higher mineralization temperatures (563-603 K), CuCl2 was found to be predominant on the surfaces of the mineralization product. Speciation

Micronutrients: oxidant/antioxidant status.

Potentially damaging species (reactive oxygen, nitrogen and chlorine species) arise as by-products of metabolism and as physiological mediators and signalling molecules. Levels of these species are controlled by the antioxidant defence system. Several components of this system are micronutrients (e.g. vitamins C and E) or are dependent upon dietary micronutrients (e.g. CuZn and Mn superoxide dismutase). The antioxidant defences act as a coordinated system where deficiencies in one component may affect the efficiency of the others. Oxidative stress may be an important factor in infection if mic

Nitric oxide and asthma: a review.

Nitric oxide (NO) is synthesized from the amino acid arginine by enzymes called nitric oxide synthases. NO has an important physiologic role in the regulation of vascular tone, response to vascular injury, and hemostasis. It also acts as a neurotransmitter for the nonadrenergic noncholinergic nerves and has important antimicrobial, immunologic, and proinflammatory activities. The lung is rich in nitric oxide synthases, and NO is normally present in the exhaled air. Use of NO in the treatment of asthma has not withstood the test of time and is not recommended. With the advent of analyzers capab
