Comparative effects of chronic administration of some psychotropic drugs on rat brain cortex acetylcholinesterase activity.

The response of central cholinergic neurotransmission to the chronic administration of some psychotropic drugs to rats was investigated using brain acetylcholinesterase activity as a neurochemical marker for cholinergic neurons. Rats were divided into four groups. Three experimental groups were given chlorpromazine, amitriptyline, or diazepam respectively, for a period of 30 days; and the control group received physiological saline only. Long-term treatment of chlorpromazine and amitriptyline resulted in significant increases in rat brain cortex enzyme activity, whereas only a slight increase

[The morphological characteristics of the parasympathetic innervation of the pyloric sphincter in the cat].

The method of axonal transport of horse-radish peroxidase was used to detect the localization of neurons in the dorsal motor nucleus of the vagus nerve sending the axons to the pyloric sphincter. The investigation was carried out in cats. Under study were also morphological features of the nodular ganglion responsible for afferent innervation of the sphincter. The maximum amount of the corresponding cells are found in the dorsomedial part of the dorsal motor nucleus in the area from +1.0 to +2.0 mm (with respect to obex). The afferent neurons to which information comes from interoceptors of th

Xanthine derivatives IBMX and S-9977-2 potentiate transmission at an Aplysia central cholinergic synapse.

In an attempt to investigate the role of cAMP-dependent phosphorylations on synaptic transmission at an Aplysia cholinergic buccal ganglion synapse, the effects of xanthine derivatives such as 3-isobutyl-1-methylxanthine (IBMX), which is well known to inhibit phosphodiesterase activity thereby promoting cAMP accumulation, and a novel xanthine derivative, S-9977-2 were evaluated. They were found to potentiate cholinergic transmission by significantly increasing the time constant of decay (Tc) of inhibitory postsynaptic currents (IPSCs). The postsynaptic origin of the phenomenon was supported by

Atropine-resistant secretory responses of the ovine parotid gland to reflex and direct parasympathetic stimulation.

A study was made of mechanisms involved in the parasympathetic control, especially of protein concentration in, and also flow, of ovine parotid secretion. The effects of distension of the caudal thoracic oesophagus were characterized in four anaesthetized sheep each with a unilateral chronic superior cervical sympathetic ganglionectomy. Oesophageal distension produced increases in both the flow and protein concentration of saliva from the control glands similar to that from the sympathectomized glands indicating the response was mediated by the parasympathetic innervation. Atropine, in doses w

An attempt to produce cholinergic hypofunction in rat brain using choline mustard aziridinium ion: neurochemical and histological parameters.

Deficiencies in cholinergic nerve function have been clearly documented in patients with Alzheimer's disease. The lack of this neurotransmitter may be responsible for early memory loss in patients with the disease. Choline mustard Az ion has been used in our laboratory to produce cholinergic hypofunction in rat brain. Injections of the compound into the medial septal nucleus and dorsal hippocampus produced tissue lesions. The lesions were dose and time-dependent. Lesions produced in the medial septum resulted in transmitter depression in the hippocampus. These results suggest non-specific tiss

Parasympathetic control of coronary blood flow.

Active parasympathetic coronary vasodilation in excess of any changes in myocardial metabolism has been observed in a number of circumstances. Electrical stimulation of the cardiac end of the cut vagus nerve produces a cholinergic coronary vasodilation that is blocked by atropine. Activation of carotid body chemoreceptors, carotid sinus baroreceptors, or left ventricular receptors elicits reflex parasympathetic coronary vasodilation. The coronary vasodilation produced by these reflexes can be prevented by vagotomy or atropine. The relative importance of parasympathetic coronary control in rela

The site of action of Ptychodiscus brevis toxin within the parasympathetic axonal sodium channel h gate in airway smooth muscle.

The red tide toxin produced by Ptychodiscus brevis ( PBTX ) may cause cough, sneezing, and asthma. Previous in vitro studies with isolated canine tracheal smooth muscle demonstrated that PBTX stimulates sodium channels of parasympathetic nerve endings and thus causes a contractile response. The present study investigated the mechanism of the PBTX effect on canine tracheal smooth muscle. Repeated exposure of the muscle strip to PBTX (final concentration 46 micrograms/ml) followed by washout of the toxin resulted in reestablishment of baseline tension but a failure of contraction on further addi

Cholinergic control of canine antral immunoreactive gastrin release and motility.

We used acute anesthetized dogs to investigate the role of cholinergic receptors in the relationship between antral immunoreactive (I) gastrin release and antral motility. Electrical stimulation of extrinsic nerves via the cervical vagus or the nerve of Latarjet appeared to increase I gastrin release and antral motility by separate pathways as blockade of muscarinic receptors, i.e., atropinization inhibited motility but did not alter I gastrin release. On the other hand, blockade of nicotinic receptors by hexamethonium treatment obliterated I gastrin release induced by stimulation of the extri

The distribution of visceral primary afferents from the pelvic nerve to Lissauer's tract and the spinal gray matter and its relationship to the sacral parasympathetic nucleus.

The central distribution of visceral primary afferent fibers from the pelvic nerve of the cast and the relationship of these fibers to preganglionic neurons of the sacral parasympathetic neurons (SPN) have been studied. Horseradish peroxidase (HRP) applied to the cut pelvic nerve was detected ipsilaterally in preganglionic neurons and dorsal root ganglion cells (segments S1-S3), and in central afferent projections to Lissauer's tract (LT), the dorsal columns, the dorsolateral funiculus, and spinal gray matter. The afferent projections were strongest in the region of the SPN (S1-S3) but extende

The effect of hemicholinium-3 on choline and acetylcholine levels in a sympathetic ganglion.

Pregangliaaonic stimulation of the cat's superior cervical ganglion in the presence of hemicholinium-3 (HC-3) produced the expected depletion of acetylcholine (ACh) stores, but failed to cause a corresponding reduction in the choline content. These results suggest that either HC-3 possesses an intracellular site of action or that in lower doses it selectively inhibits a specialized choline transport system in cholinergic nerves. At a dose of 2 mg/kg, HC-3 probably blocked ACh synthesis completely in ganglia stimulated at 20 Hz. Under these conditions, there was a rapid depletion of ACh to abou

Nutritional factors in the pathobiology of human essential hypertension.

Endothelial cells produce vasodilator and vasoconstrictor substances. Dietary factors such as sodium, potassium, calcium, magnesium, zinc, selenium, vitamins A, C, and E, and essential fatty acids and their products such as eicosanoids can influence blood pressure, cardio- and cerebrovascular diseases, and concentrations of blood lipids and atherosclerosis. There might be a close interaction between these dietary factors, sympathetic and parasympathetic nervous systems, the metabolism of essential fatty acids, nitric oxide, prostacyclin, and endothelium in human essential hypertension. A defic…

[Shy-Drager syndrome: its autonomic function, alpha 2 receptor density and mechanism of postprandial hypotension].

The autonomic function, platelet alpha 2 receptor (alpha 2R) density, and mechanism of postprandial hypotension were examined in a 58-year-old man with Shy-Drager syndrome (SDS). His chief complaints were orthostatic syncope and impotence. His blood pressure was kept within normal limits in a supine position, but severe hypotension and fainting occurred when he assumed an upright posture. There were diminished sweating response to warm stress, abnormal pupil reactions to drugs, lack of blood pressure elevation at phase IV during the Valsalva maneuver, and a lack of pressor response to hyperven

Autonomic mechanisms in oral sensations.

Sympathetic fibres reach the oro-facial region by several routes, the main one being via the carotid nerve and the trigeminal or facial nerv … these effects are probably not due to specific modulatory mechanisms. There is no evidence that the parasympathetic nervous system affects the properties of oral receptors. Patients with Horner's syndrome appear to have no sensory abnormalities, apart from pain which is sometimes associated with the onset of the condition. Reflex sympathetic dystrophy seems to be rare in the or …

Recognition of cholinergic agonists by the muscarinic receptor. 1. Acetylcholine and other agonists with the NCCOCC backbone.

A theoretical model is used to deduce the pharmacologically active conformation of acetylcholine and other agonists interacting with the muscarinic receptor of the parasympathetic and central nervous systems. This is accomplished by replacing the usual dihedral angles tau 1 and tau 2, which define the conformations of cholinergic drugs, with two new geometric parameters more suitable for describing the muscarinic pharmacophore: a characteristic distance, [PQ], and a dihedral angle, PNOQ. Values for these parameters are determined by conformational analysis on semirigid muscarinic agonists usin

The autonomic nervous system regulates pancreatic β-cell proliferation in adult male rats.

The pancreatic β-cell responds to changes in the nutrient environment to maintain glucose homeostasis by adapting its function and mass. Nutrients can act directly on the β-cell and also indirectly through the brain via autonomic nerves innervating islets. Despite the importance of the brain-islet axis in insulin secretion, relatively little is known regarding its involvement in β-cell proliferation. We previously demonstrated that prolonged infusions of nutrients in rats provoke a dramatic increase in β-cell proliferation in part because of the direct action of nutrients. Here, we addressed t

Pubertal developmental, body mass index, and cardiovascular autonomic function in children and adolescents with and without autism spectrum disorder: a four-time point accelerated longitudinal study.

BACKGROUND: The Autonomic Nervous System (ANS) regulates 'automatic' functions such as heart rate, and alterations may have significant impacts on health outcomes. Cardiovascular measures of autonomic function such as heart rate variability are of interest as biological markers in autism spectrum disorder (ASD). The interplay between the ANS and physical health establishes a need to examine cardiovascular autonomic functioning in youth with and without ASD over development. The current study aimed to identify change in autonomic function and balance across the parasympathetic and sympathetic b

Long noncoding RNA H19 promotes the apoptosis of corpus cavernsum smooth muscle cells after cavernosal nerve injury via JNK signalling pathway.

JNK/ Bcl-2/ Bax pathway participates in corpus cavernosal smooth muscle cells apoptosis during early period after cavernosal nerve (CN) crush injury (CNCI). Nevertheless, the regulation mechanisms of long noncoding RNA H19 in apoptosis during early stage after CN injury are still poorly understood. The rats in sham group were not direct injury to the CNs. The rats in CNCI group were performed to bilateral CN crush injury. The ICP/MAP rate and smooth muscle content were significantly lower than that in the sham group. Primary CCSMCs were prepared from the tissues samples after completing erecti
