Dietary Protein Reduction in Lean Pigs From Pietrain Paternal Line Under Commercial Setting: An Interplay Between Nitrogen Efficiency and Meat Attributes.

A trial was conducted to research the impact of reducing dietary crude protein (CP) levels from 14.8% to 12.6% in the finishing diet (90-120 kg of body weight) of lean pigs, with an essential AA supply (8.8 g Lys, 2.1 g Met, 6.7 g Thr, 5.5 g Ile, and 6.7 g Val, per kilogram of feed) in intact males and females under commercial setting. The productive performances and nitrogen (N) balance, blood markers related to protein metabolism, apparent digestibility of nutrients, carcass, and meat quality were evaluated. This dietary CP restriction did not impair the growth performances, decreased the N

Paternal preconception alcohol consumption increased Angiotensin II-mediated vasoconstriction in male offspring cerebral arteries via oxidative stress-AT1R pathway.

Alcohol consumption is popular worldwidely and closely associated with cardiovascular diseases. Influences of paternal preconception alcohol consumption on offspring cerebral arteries are largely unknown. Male rats were randomly given alcohol or water before being mated with alcohol-naive females to produce alcohol- and control-sired offspring. Middle cerebral artery (MCA) was tested with a Danish Myo Technology wire myograph, patch-clamp, IONOPTIX, immunofluorescence and quantitative PCR. Alcohol consumption enhanced angiotensin II (AngII)-mediated constriction in male offspring MCA mainly vi

Paternal morphine exposure enhances morphine self-administration and induces region-specific neural adaptations in reward-related brain regions of male offspring.

BACKGROUND: A growing body of preclinical studies report that preconceptional experiences can have a profound and long-lasting impact on adult offspring behavior and physiology. However, less is known about paternal drug exposure and its effects on reward sensitivity in the next generation. METHODS: Adult male rats self-administered morphine for 65 days; controls received saline. Sires were bred to drug-naïve dams to produce first-generation (F1) offspring. Morphine, cocaine, and nicotine self-administration were measured in adult F1 progeny. Molecular correlates of addiction-like behaviors w

Case Report: A Novel Homozygous Mutation in MYF5 Due to Paternal Uniparental Isodisomy of Chromosome 12 in a Case of External Ophthalmoplegia With Rib and Vertebral Anomalies.

External ophthalmoplegia with rib and vertebral anomalies (EORVA) is characterized by congenital nonprogressive external ophthalmoplegia, ptosis, scoliosis, torticollis, vertebral, and rib anomalies, caused by homozygous mutations in the myogenic factor 5 gene (MYF5) located on chromosome 12q21.31. Uniparental disomy (UPD) is a rare inheritance of a pair of chromosomes originating from only one parent. This study describes a case of an 8-year-old boy with ptosis, scoliosis, and dysmorphic hypoplastic ribs with fusion anomalies. Trio-based exome sequencing (trio-ES) identified a novel homozygou

Paternal Exposure to Non-essential Heavy Metal Affects Embryo Cleavage and Implantation in Intracytoplasmic Sperm Injection (ICSI) Cycles: Evidence for a Paradoxical Effect.

Although the adverse effects of non-essential heavy metals on semen quality have been demonstrated in experimental animal models and occupational human exposure studies, little is known about the reproductive efficiency of exposed sperm during the process of intracytoplasmic sperm injection (ICSI). Our study aims to evaluate the effect of paternal exposure to non-essential heavy metals on embryo efficiency outcomes (embryo cleavage, fragmentation, implantation, and live birth) in ICSI cycle. Ninety-five heterosexual couples who underwent 95 ICSI cycles and 78 fresh embryo transfers between Jan

The human homologue (PEG3) of the mouse paternally expressed gene 3 (Peg3) is maternally imprinted but not mutated in women with familial recurrent hydatidiform molar pregnancies.

OBJECTIVES: We mapped a locus for autosomal recessive molar pregnancies with biparental genomic contribution to chromosome 19q13.4 between D19S924 and D19S890. This 5-Mb region is homologous to proximal mouse chromosome 7 and contains a cluster of Krüppel-type zinc finger genes, including the human homologue of the mouse imprinted genes: the paternally expressed gene 3 (PEG3) and the maternally expressed Zim1 genes. We analyzed the PEG3 gene for mutations in women with familial recurrent hydatidiform moles and to determine its imprinting status in humans. METHODS: We used database searches an

Father-to-daughter transmission of focal dermal hypoplasia associated with nonrandom X-inactivation: support for X-linked inheritance and paternal X chromosome mosaicism.

Focal dermal hypoplasia (FDH) is a rare syndrome of severe developmental anomalies of the tissues and organs derived from ectoderm and mesoderm. Though data have suggested that FDH is an X-linked dominant trait associated with male hemizygote lethality, a hypothesis supported by the observation of three unrelated infants with FDH manifestations and de novo chromosome rearrangements involving Xp22, observations of father-to-daughter transmission have suggested possible genetic heterogeneity and autosomal dominant inheritance with sex limitation. We hypothesize that, if FDH is an X-linked disord

Immune reactivity of allogeneically pregnant mice to paternal MHC antigens on fetal and placental cells assessed by second set rejection of ascites tumor cells.

In vivo immunogenicity of fetus- and placenta-derived cells as well as the immune reactivity of pregnant mice to fetal cells were examined for graft rejecting response (GRR). Systemic administration of small numbers of fetal cells but not placental cells from allogeneically pregnant mice (10(6) cells per mouse) or adult allogeneic spleen cells (10(4) cells) sensitized mice for second-set rejection of an ascitic tumor bearing paternal major histocompatibility complex (MHC) antigens. Despite this fact and the known positive humoral response, pregnant and parous mice are not even minimally sensit

Fatherhood in testicular cancer patients with carcinoma in situ in the contralateral testicle.

We report paternity by assisted fertilization in 3 highly oligospermic patients with stage-I unilateral testicular germ cell tumor and contralateral carcinoma in situ. If such patients plan future paternity, surveillance is the optimal treatment allowing maximal recovery of spermatogenesis after orchiectomy. Multiple semen cryopreservations should be done during the first post-orchiectomy year.

No relationship of testicular size at orchiopexy with fertility in men who previously had unilateral cryptorchidism.

PURPOSE: We determined whether testicular size at orchiopexy is predictive of fertility potential and whether size correlates with sperm parameters, hormone levels or testicular volume in adulthood. MATERIALS AND METHODS: Testicular size obtained from the operative notes of patients who underwent unilateral orchiopexy was classified as normal, small or large for age. These data were compared with outcome data. Paternity data were included on 166 men who reported achieving or unsuccessfully attempting paternity for 12 or more months. Of the men 49 provided blood for hormone level measurement, …

Paternal BPA and BPS exposure induce testicular dysfunction in pubertal male offspring: roles of OCTN2 carnitine transporter disruption and exacerbated oxidative stress/apoptosis.

Although numerous studies have reported the male reproductive toxicity of bisphenol A (BPA) and its substitute bisphenol S (BPS), the impacts of paternal exposure to these chemicals on the male reproductive system of offspring and the underlying molecular mechanism remain inadequately explored. In this study, we investigated male reproductive toxicity in pubertal offspring resulting from paternal exposure to environmentally relevant doses of BPA (0.45 μg/kg body weight [bw]/day) or BPS (0.15 μg/kg bw/day). Our results showed that paternal exposure to BPA or BPS reduced pubertal offspring testo

Paternal Transmission of Angelman Syndrome via a Mosaic Small Supernumerary Marker Chromosome 15: A Rare Case of Germline Epigenetic Reset Failure and Somatic Rescue.

INTRODUCTION: Angelman syndrome (AS) is an imprinting disorder that arises almost exclusively through maternal mechanisms; most commonly maternal 15q11.2-q13 deletion; less often, paternal uniparental disomy (UPD) of chromosome 15. Paternal transmission of a 15q11.2-q13 deletion typically results in Prader-Willi syndrome, making familial paternal AS transmission exceptionally rare. Herein, we report a pedigree in which a neurotypical father transmitted AS to multiple offspring via a structural mechanism distinct from UPD. CASE PRESENTATION: The progenitor exhibited 47,XY,+mar[24]/46,XY [6] mo

Paternal genomic history and Y-chromosome phylogeny from the Chengdu Plain reveal multiple Indigenous East Asian farmer contributions to ancient Shu and their descendants.

BACKGROUND: The Chengdu Plain and surrounding regions (CPS) host a diverse mosaic of archaeological cultures and ethnolinguistic groups, yet the origins of these populations and the extent of Western Eurasian and East Asian influences remain unclear and underrepresented in genomic studies. As a molecular archaeological archive, Y-chromosome data provide vital insights into the paternal genetic history of ancient Shu populations and their descendants. METHODS: Here, we combine our newly generated genomic data with publicly available large-scale whole-Y-chromosome sequences from East Asia to re

Paternal Sertoli cell-derived extracellular vesicles transfer mtDNA and microRNA cargo to trigger germ cell oxidative stress and induce metabolic risks in offspring.

While obesity causes male infertility through testicular lipid stress, the mechanisms by which this stress alters Sertoli cell-derived extracellular vesicles (EVs) to induce paternal reproductive defects and offspring metabolic dysfunction remain unclear. Here, we report that lipid-overloaded Sertoli cells release modified EVs that impair paternal reproductive health and predispose offspring to metabolic disorders. Specifically, we found that palmitic acid treatment alters the lipid composition of Sertoli-derived EVs, resulting in an elevated phosphatidylethanolamine (PE)/phosphatidylcholine (

Chronic paternal Wi-Fi RF-EMR exposure impairs placental efficiency and fetal growth: Association with sex-dependent histone modification patterns in mice.

The widespread use of Wi-Fi-derived radiofrequency electromagnetic radiation (RF-EMR) has raised concerns regarding male reproductive health; however, whether paternal exposure exerts transgenerational effects on offspring development remains unclear. This study investigated the effects of chronic paternal 2.4 GHz Wi-Fi RF-EMR exposure (whole-body SAR 0.125-0.5 W/kg, 4 h/day, 6 days/week for 20 weeks) on sperm quality in F0 male mice and placental development in F1 offspring, with a focus on epigenetic regulation. Paternal RF-EMR exposure significantly reduced sperm concentration and increased

Effect of paternal exposure to glyphosate and microcystin-LR on zebrafish (Danio rerio) fathers and progeny: An integrated investigation based on behavioral, histopathological, biochemical, and transcriptomic analysis.

The coexistence of glyphosate (GLY) and microcystin-LR (MC-LR) in the environment has become a problem that cannot be ignored. However, the combined toxic effects of these compounds on aquatic organisms remain underexplored. The purpose of this study was to evaluate the individual and combined effects of GLY and MC-LR on the neurobehavior of exposed adult zebrafish and their untreated offspring (F1 larvae), as well as to explore the underlying mechanisms. In this study, six month old male zebrafish were exposed to 3.5 mg/L GLY and 35 μg/L MC-LR, alone and in combination, for 21 d. The results

Non-Invasive Determination of the Paternal Inheritance in Pregnancies at Risk for β-Thalassaemia by Analyzing Cell-Free Fetal DNA Using Targeted Next-Generation Sequencing.

Non-invasive prenatal testing (NIPT) has been widely adopted for the screening of chromosomal abnormalities; however, its adoption for monogenic disorders, such as β-thalassaemia, has proven challenging. Haemoglobinopathies are the most common monogenic disorders globally, with β-thalassaemia being particularly prevalent in Cyprus. This study introduces a non-invasive prenatal haplotyping (NIPH) assay for β-thalassaemia, utilizing cell-free DNA (cfDNA) from maternal plasma. The assay determines paternal inheritance by analyzing highly heterozygous single-nucleotide variants (SNVs) in the β-glo

Case report: Long term response to growth hormone in a child with Silver-Russell syndrome-like phenotype due to a novel paternally inherited IGF2 variant.

Silver-Russell syndrome (SRS, OMIM, 180860) is a rare genetic disorder with a wide spectrum of symptoms. The most common features are intrauterine growth retardation (IUGR), poor postnatal development, macrocephaly, triangular face, prominent forehead, body asymmetry, and feeding problems. The diagnosis of SRS is based on a combination of clinical features. Up to 60% of SRS patients have chromosome 7 or 11 abnormalities, and <1% show abnormalities in IGF2 signaling pathway genes (IGF2, HMGA2, PLAG1 and CDKN1C). The underlying genetic cause remains unknown in about 40% of cases (idiopathic SRS)
