Activity of anti-cancer protein kinase inhibitors against Leishmania spp.

OBJECTIVES: There is an urgent need to develop new and effective treatments for poverty-related neglected diseases. In light of the time required to bring a new drug to market and the cost involved (10-15 years, >1 billion US$), one approach to identifying new treatments for diseases like leishmaniasis is to evaluate drugs that are already registered for the treatment of other diseases. This paper describes the anti-leishmanial activities of 10 FDA-approved protein kinase inhibitors already available for the treatment of human cancers. METHODS: In vitro and in vivo models of Leishmania infect

Recent advances in cytogenetics and molecular biology of adult hepatocellular tumors: implications for imaging and management.

Focal nodular hyperplasia (FNH), hepatocellular adenoma (HCA), and hepatocellular carcinoma (HCC) compose hepatocellular neoplasms that occur in adults. These tumors demonstrate characteristic epidemiologic and histopathologic features and clinical and imaging manifestations. HCAs are monoclonal neoplasms characterized by increased predilection to hemorrhage or rupture and occasional transformation to HCC. On the other hand, FNH is a polyclonal tumorlike lesion that occurs in response to increased perfusion and has an indolent clinical course. Up to 90% of HCCs occur in the setting of cirrhosi

Sorafenib improves the survival of patients with advanced hepatocellular carcinoma: a meta-analysis of randomized trials.

There is no effective systemic therapy for patients with advanced hepatocellular carcinoma (HCC) except liver transplantation. Sorafenib, a multikinase inhibitor, has been shown to significantly increase overall survival (OS) in a randomized, placebo-controlled, phase III trial of patients with HCC (SHARP). The aim of this study was to evaluate the effectiveness of sorafenib for advanced HCC by carrying out a meta-analysis of randomized controlled trials that compared sorafenib-based therapy with other agent-based therapy. Randomized controlled trials comparing sorafenib or combined chemothera

Long lasting response to the multikinase inhibitor bay 43-9006 (Sorafenib) in a heavily pretreated metastatic thymic carcinoma.

Metastatic thymic carcinoma is an aggressive neoplasm for which multimodal therapies are often ineffective. We describe here a heavily pretreated patient with advanced thymic carcinoma responsive to multikinases inhibitor BAY 43-9006 (Sorafenib). Of note, a hitherto unreported c-kit missense mutation on exon 17 (D820E) identified in tumor cells seems to explain the clinical response and highlight the key role of molecular analysis in predicting efficacy of targeted therapies even in thymic neoplasms.

Targeted therapies for metastatic renal cell carcinoma: an overview of toxicity and dosing strategies.

The targeted therapies sunitinib, sorafenib, temsirolimus, and bevacizumab (when used in combination with interferon-alpha2a) have dramatically improved outcomes for patients with advanced renal cell carcinoma (RCC). Clinical application of these novel agents outside the trial setting, however, may present some challenges for treating individual patients with unique needs. In some patients, dose modifications may be considered for potential drug interactions and for management of severe cases of hematologic or nonhematologic toxicities. The more common grade 3 or 4 side effects with sunitinib

Phase 2 study of the combination of merimepodib with peginterferon-alpha2b, and ribavirin in nonresponders to previous therapy for chronic hepatitis C.

BACKGROUND/AIMS: While combination of peginterferon-alpha (PEG-IFN) and ribavirin (RBV) therapy is the current standard of care for chronic hepatitis C (CHC), only 44-51% of genotype-1 patients achieve a sustained virological response (SVR), and both agents produce treatment-limiting toxicities. In the hepatitis C virus (HCV) replicon system, merimepodib (MMPD), a novel, selective inhibitor of inosine monophosphate dehydrogenase, has shown potent antiviral effects. METHODS: This randomized, placebo-controlled, double-blind study evaluated the safety and antiviral activity of PEG-IFN-alpha2b a

Gastroprotective effects of pantoprazole against experimental mucosal damage.

The present study investigated the gastroprotective effects of the proton pump inhibitor pantoprazole on gastric mucosal damage induced by ethanol-HCl in rats. Omeprazole was used as reference drug. The morphometric analysis of gastric histological sections revealed that pantoprazole and omeprazole dose-dependently prevented the necrotic mucosal injury evoked by ethanol-HCl (ED50 = 14.1 and 21.6 micromol/kg, respectively). These effects were associated with a marked increment of Alcian blue recovery from gastric bound mucus (ED50 = 18.8 and 29.3 micromol/kg, respectively). In addition, both pa

Single-Cell Lineage Tracing Uncovers Resistance Signatures and Sensitizing Strategies to FLT3 Inhibitors in Acute Myeloid Leukemia.

UNLABELLED: Whereas FMS-like tyrosine kinase-3 (FLT3) inhibitors have significantly improved the treatment of aggressive FLT3-mutated acute myeloid leukemia (AML), the emergence of resistance remains as a major challenge. In this study, we applied our recently developed single-cell lineage-tracing method ReSisTrace to identify cells that are intrinsically resistant or sensitive to the FLT3 inhibitors midostaurin and quizartinib in AML with FLT3-ITD mutations. Comparison of the gene expression profiles of these cells revealed transcriptional resistance signatures, including upregulation of GSPT

Comparative efficacy of different therapeutic approaches in treatment naïve FLT3-mutated AML eligible for intensive chemotherapy: a Bayesian network meta-analysis of randomized trials.

FLT3-mutated acute myeloid leukemia (FLT3mut AML) is associated with poor outcomes. Although FLT3 inhibitors (FLT3is) combined with chemotherapy improve responses, long-term survival remains limited, and the optimal first-line strategy is unclear. We conducted a Bayesian network meta-analysis of eight randomized trials, including 1,793 patients, to compare treatment used in patietne eligible for intensive chemoteherapy for overall survival (OS). Treatments studied were 3 + 7 with midostaurin, quizartinib, sorafenib, gemtuzumab ozogamicin (GO), glasdegib, CPX-351, and decitabine. FLT3i-based re

Nintedanib is a potent FLT3 inhibitor with activity against FLT3-ITD and overcomes the gatekeeper F691L resistance mutation in acute myeloid leukemia.

Acute myeloid leukemia (AML) is a molecularly heterogeneous malignancy in which FMS-like tyrosine kinase 3 (FLT3) mutations, particularly internal tandem duplications (FLT3-ITD), occur in approximately 30% of cases and are associated with poor prognosis and high relapse risk. While FLT3 inhibitors have improved clinical outcomes, acquired resistance, often mediated by secondary tyrosine kinase domain (TKD) mutations or other mechanisms, remains a major therapeutic challenge. Nintedanib is an oral multi-kinase inhibitor approved for the treatment of fibrotic lung diseases; however, its potentia

EPHX1 enhances drug resistance to regorafenib by activating the JAK/STAT signaling pathway in hepatocellular carcinoma cell lines.

BACKGROUND: Regorafenib serves as a second-line treatment for patients with advanced hepatocellular carcinoma (HCC). Microsomal epoxide hydrolase 1 (EPHX1) is closely associated with tumorigenesis and drug resistance. However, the relationship between EPHX1 and regorafenib resistance, as well as the underlying mechanisms in HCC, remains unclear. OBJECTIVE: To investigate the role and mechanisms of EPHX1 in mediating regorafenib resistance in HCC. METHODS: We assessed the protein expression levels of EPHX1 in human HCC tissues and adjacent non-tumor tissues. Subsequently, we constructed HCC c

DDX1 facilitates lenvatinib resistance in hepatocellular carcinoma through regulating ephrin-A3 and activating the Wnt/β-catenin signaling pathway.

Background Hepatocellular carcinoma (HCC) is one of the leading causes of cancer-related death worldwide. Lenvatinib is a common first-line treatment for advanced HCC. However, resistance to lenvatinib is the greatest challenge limiting its clinical application. Currently, the molecular mechanisms of resistance remain poorly understood. Methods The expression of DDX1 and Ephrin-A3 in lenvatinib-resistant HCC cells was identified via RNA-seq and Western blotting. Bioinformatic analyses were applied to explore its expression and prognostic role. The biological role of DDX1 was evaluated via CCK8
