Deciphering genetic basis of phenylethanoid glycoside production through integrative multiomics analysis in Callicarpa nudiflora.

Callicarpa nudiflora is a medicinal plant rich in phenylethanoid glycosides (PhGs), particularly verbascoside, but the genetic basis of PhG biosynthesis and natural variation in its content remain largely unexplored. Here, we assembled a chromosome-level genome of C. nudiflora (511 Mb, contig N50 = 12.78 Mb) with 39 317 annotated protein-coding genes and 99.5% Benchmarking Universal Single-Copy Orthologs completeness and quantified its bioactive compounds and gene expression across multiple tissues. Using the dataset, we elucidated the complete biosynthetic pathway of verbascoside and demonstr

Cotton Classical Extensin GhCEXT1-A Regulates Fiber Elongation by Modulating Cell Wall Structure and Composition.

Classical extensins (CEXTs) are hydroxyproline-rich glycoproteins that belong to the extensin superfamily and constitute critical structural components of the plant cell wall. However, the genome-wide characterization and functional role of CEXTs in cotton fiber development remain very limited. Here, we conducted a comprehensive comparative genomic analysis across four Gossypium species and identified a total of 30 CEXT genes, which were systematically classified into four evolutionary lineages (Groups A-D) based on analyses of phylogenetic relationship and gene structural organization. Sequen

Transcriptomic insights into the molecular regulatory mechanism of staminody in Chinese narcissus (Narcissus tazetta subsp. chinensis).

Staminody (petaloid stamen) contributes to floral diversity in horticultural plants, yet its molecular regulation remains poorly understood. Using two Chinese narcissus varieties, 'Jinzhanyintai' (single-flowered) and 'Yulinglong' (double-flowered), we conducted anatomical and histological analyses, which confirmed that the petaloid stamens of 'Yulinglong' arise from a homeotic transformation of normal stamens of 'Jinzhanyintai', as evidenced by conserved internal locule-like structures and gradual morphological intermediates. Comparative transcriptomics across five key developmental stages re

Enhanced Rice Yellow Mottle Virus Resistance via CRISPR/Cas9-Targeted Mutagenesis of the Rice eIF(iso)4G Gene.

Rice is a staple crop primarily recognised for its high content of carbohydrates and proteins. Rice yellow mottle disease (RYMD) is a destructive disease affecting rice fields in sub-Saharan Africa and is caused by the rice yellow mottle virus (RYMV). Development of virus-resistant genotypes is a highly recommended and effective approach to controlling RYMV. A genetic approach that exploits recessive mutations in susceptibility (S) genes may enhance resistance to the virus. Reports indicate that most rice genotypes grown in Kenya are vulnerable to RYMV infection. Genome editing has shown promi

Cold-induced SlPAL3 confers cold tolerance in tomato through interacting with putative transcription factor SlMYB58.

Cold stress severely restricts tomato growth, yield, and geographical distribution. However, the molecular mechanisms governing phenylpropanoid-mediated cold tolerance remain incompletely understood, especially the upstream regulators of specific phenylalanine ammonia-lyase (PAL) isoforms. In this study, we functionally characterized SlPAL3 and identified its putative upstream transcriptional activator SlMYB58 in tomato. Using phylogenetic analysis, subcellular localization, and the generation of CRISPR-Cas9 knockout (CR) and overexpression (OE) transgenic lines, we demonstrated that cytoplasm

Genome-wide characterization of BiP gene family and functional analysis of VvBiP3 in Vitis vinifera.

Drought stress exacerbated by climate change severely impairs the growth and quality of horticultural crops. Grapevine (Vitis vinifera L.) is an economically vital fruit species, and exploring stress-tolerant genes lays a foundation for its genetic improvement and sustainable cultivation. BiP proteins serve as pivotal endoplasmic reticulum (ER) molecular chaperones and participate widely in plant abiotic stress acclimation, yet the genomic features and drought-associated functions of grapevine BiP family genes remain largely unclear. In this study, three VvBiP members were identified genome-wi

Integrating the tomato chlorosis virus with the tobacco rattle virus facilitates the direct infection of tomato.

Tomato chlorosis virus (ToCV) threatens tomato production, resulting in substantial yield losses. An efficient inoculation system for ToCV can accelerate the isolation and utilization of tomato-resistant genetic resources. However, infectious ToCV clones cannot directly infect tomato plants. In this study, we cloned the full-length ToCV genome, including RNA1 and RNA2 molecules, and integrated their corresponding DNA fragments separately into the less virulent tobacco rattle virus-based pTRV2GW vector. Agrobacterium suspensions carrying pTRV1, pTRV2GW-RNA1, and pTRV2GW-RNA2 plasmids were mixed

Spatiotemporal Dissection of the Infection Response to Bacterial Soft Rot in Non-heading Chinese cabbage.

Bacterial soft rot caused by Pectobacterium brasiliense (Pbr) leads to substantial yield losses in cruciferous vegetables, including non-heading Chinese cabbage (pak choi, Brassica rapa subsp. chinensis). This study employed an integrated physiological and multi-tissue transcriptomic approach to decipher the infection response strategies of pak choi against Pbr. Age-dependent resistance was detected, with susceptibility decreasing as plants matured. In susceptible four-week-old plants, Pbr infection induced oxidative stress, characterized by the accumulation of hydrogen peroxide (H₂O₂) and mal

Identification of a Glycosyltransferase-Encoding Gene (EuGT8) from Eucommia ulmoides That Catalyzes the Glycosylation of Pinoresinol to Pinoresinol Diglucoside.

Pinoresinol diglucoside (PDG), one of the major lignans isolated from E. ulmoides Oliver bark, has various pharmacological functions, including antihypertension and prevention of osteoporosis. However, the glycosyltransferase-encoding gene (GT) involved in regulating the glycosylation of pinoresinol to form PDG has not been reported in E. ulmoides. In this study, we screened and cloned the EuGT8 gene from E. ulmoides based on our transcriptome data. The expression pattern of the EuGT8 gene exhibited a strong positive correlation with dynamic changes in the PDG contents in three different organ

Efficient In Planta Induction of Transgenic Hairy Roots in Macadamia Seedlings and Mature Trees Using Visual Reporters.

Macadamia (Macadamia spp.) is an economically important nut crop whose severe recalcitrance to genetic transformation substantially hinders progress in functional genomics and molecular breeding. To overcome this critical technical bottleneck, this study established a highly efficient and broadly applicable in planta hairy root genetic transformation system with integrated visual screening. This system utilizes an Agrobacterium rhizogenes-mediated transformation method, employing multiple visual reporter gene systems (DsRed2, eGFP, RUBY, and AtPAP2) to achieve antibiotic-independent and non-de

Development of Resistance to Damping-Off in Rice, Oryza sativa L., Using CRISPR/Cas9.

Damping-off disease hinders rice seedling growth and reduces yield. Current control methods, such as seed or soil sterilization, rely on chemicals that cause environmental pollution and promote pathogen resistance. As a sustainable alternative, we targeted the damping-off resistance-related gene OsDGTq1 using CRISPR/Cas9. Field experiments first verified OsDGTq1's significance in resistance. The CRISPR/Cas9 system, delivered via Agrobacterium-mediated transformation, was used to edit OsDGTq1 in rice cultivar Ilmi. Lesions from major damping-off pathogens, Rhizoctonia solani and Pythium gramini

Comparative mitochondrial genome analysis of Panax ginseng reveals structural variation and genetic diversity.

Ginseng (Panax ginseng) is a medicinal plant of considerable medical and economic interest. However, preserving its genetic stability and targeted selection pose challenges that limit the effective conservation and use of its genetic resources. This study aims to systematically elucidate the structural characteristics, evolutionary patterns, and phylogenetic relationships of the ginseng mitochondrial genome, in order to create a theoretical basis for the conservation of ginseng genetic resources and the acceleration of molecular selection processes. Using PacBio HiFi and DNBSEQ-T7 short-read s

The origins and spread of the opium poppy (Papaver somniferum L.) revealed by genomics and seed morphometrics.

The opium poppy (Papaver somniferum L.) is one of the most important plants in human history. It is the main source of opiates used as analgesic medicines or psychotropic drugs, the latter related to addiction problems, illegal trafficking and geopolitical issues. Poppyseed is also used in cooking. The prehistoric origins, domestication and cultivation spread of the opium poppy remain unresolved. Traditionally, Papaver setigerum has been considered the wild ancestor with early cultivation presumed to have occurred in the Western Mediterranean region, where setigerum is autochthonous. Other the

Engineering Climate-Resilient Rice Using a Nanobiostimulant-Based "Stress Training" Strategy.

Under a changing climate, cultivating climate-resilient crops will be critical to maintaining food security. Here, we propose the application of reactive oxygen species (ROS)-generating nanoparticles as nanobiostimulants to trigger stress/immune responses and subsequently increase the stress resilience of plants. We established three regimens of silver nanoparticles (AgNPs)-based "stress training": seed training (ST), leaf training (LT), and combined seed and leaf training (SLT). Trained rice seedlings were then exposed to either rice blast fungus (Magnaporthe oryzae) or chilling stress (10 °C

Identification of genes related to tipburn resistance in Chinese cabbage and preliminary exploration of its molecular mechanism.

BACKGROUND: Tipburn, also known as leaf tip necrosis, is a severe issue in Chinese cabbage production. One known cause is that plants are unable to provide adequate Ca2+ to rapidly expanding leaves. Bacterial infection is also a contributing factor. Different cultivars have varying degrees of tolerance to tipburn. Two inbred lines of Chinese cabbage were employed as resources in this research. RESULTS: We determined that the inbred line 'J39290' was the tipburn resistant material and the inbred line 'J95822' was the tipburn sensitive material based on the severity of tipburn, and the integrit

The bioavailability of (pro) vitamin A carotenoids and maximizing the contribution of homestead food production to combating vitamin A deficiency.

An estimated 100-140 million children worldwide suffer vitamin A deficiency disorders (VADD). Strategies for combating VADD are best used in combination because they serve particular target groups and none has full coverage. Homestead food production (HFP) can contribute to combating vitamin A deficiency directly, by increasing intake of vitamin A-rich foods, and indirectly through improving health and increasing income. By the late 1990s, conversion factors for estimating vitamin A obtained from plant foods were revised from 6:1 to 12:1 (microg beta-carotene:retinol activity equivalent) by th

Impact of cryopreservation on pollen viability and morphological integrity of five Indian Crinum species.

Crinum species are ornamental plants of high horticultural value and medicinal importance, known for their bioactive alkaloids with neurological and anticancer properties. Sustainable use and genetic conservation of Crinum require reliable long-term germplasm conservation strategies, and pollen cryopreservation offers an effective approach by enabling year-round breeding and conservation independent of seasonal and geographic constraints. This study presents the first species-comparative evaluation of pollen germination behaviour and cryopreservation responses in five ecologically contrasting

Pasture-based fattening does not cause severe nonperforating lesions in veal calves of dairy breeds.

Abomasal damage is a known health issue in intensive veal production and is associated with improper housing and feeding. Grass-based veal production could be an improvement, as access to pasture allows the expression of natural behaviors, such as species-specific foraging, and thus can contribute to calf welfare. However, data on the abomasal health of grazed calves have not yet been reported. As feed composition can affect the severity of mucosal damage, we compared the abomasa of calves that grazed on 2 swards with different plant composition. From 2018 to 2020, 111 calves of 2 dairy breeds
