InstructPLM-mu: protein fitness prediction from structure instructions.

Multimodal protein language models (PLMs) deliver strong performance on mutation-effect prediction, but training such models from scratch demands substantial computational resources. In this paper, we propose a fine-tuning framework called InstructPLM-mu and investigate the question: Can a pretrained sequence-only PLM, augmented with structural information during fine-tuning, match the performance of end-to-end trained multimodal models? Surprisingly, our experiments show that fine-tuning ESM2 with structural inputs can reach performance comparable to ESM3. To understand how this is achieved,

Self-Initialized Locomotion Mode Prediction With GPU-Free Terrain Reconstruction.

Lower-limb wearable robots require accurate Locomotion Mode Prediction (LMP) to provide appropriate assistance across diverse terrains. Recent reconstruction-based LMP methods fuse high-dimensional multimodal sensor data to model the relationship between human motion and terrains, improving prediction accuracy and cross-terrain adaptability. However, they typically incur high computational cost, including GPU dependency, and often require initialization procedures involving wearer participation or professional supervision. This paper proposes a self-initialized, GPU-free LMP method to overcome

DrugDL: dual-modal deep learning framework for multi-property drug prediction and targeted therapy discovery.

MOTIVATION: The accurate and robust representation of drug molecule features, the prediction of drug-target biomacromolecule interactions, and the determination of physicochemical properties are crucial in drug development. However, these tasks remain challenging due to issues such as the limited generalizability of single-modal representations, the absence of multitask prediction frameworks, and weak adaptability in cold-start scenarios. RESULTS: In this study, we present DrugDL, a framework for comprehensive drug molecule representation and the prediction of multiple downstream tasks, inclu

Adaptive Interaction Network for Human Motion Prediction During Human-Robot Collaboration.

Human motion prediction during human-robot collaboration is critical for achieving safe and efficient interactions in shared environments. Unlike previous studies that have primarily focused on humans and objects, we focus on the robot-aware human motion prediction task, which explicitly models the influence of robots on human motion. This task presents new challenges, such as capturing human-robot heterogeneity and modeling complex spatial interactions. To address these issues, we develop an adaptive interaction network (AINet) model that consists of two branches: a primary branch that predic

Causally-informed deep learning towards explainable and generalizable outcome prediction in critical care.

The rapid evolution of deep learning is revolutionizing critical care medicine by enabling highly accurate early warning systems. These systems are capable of predicting diverse clinical deteriorations, including acute kidney injury, myocardial infarction, and circulatory failure. While demonstrating strong predictive capabilities, conventional deep learning models face two fundamental limitations in clinical implementation: their opaque decision-making processes and limited generalizability across diverse patient populations and care environments. To address these challenges, we present a cau

Predictive divergence in machine learning models for clinical mortality risk: A multicohort study of covid-19 patients.

BACKGROUND: Machine learning (ML) algorithms are increasingly used in healthcare to support clinical decision-making. While models with similar overall performance are often considered interchangeable for deployment, they may produce divergent predictions, a phenomenon known as algorithmic multiplicity. In such cases, the choice of algorithm may introduce bias. This study investigates the impacts of algorithmic multiplicity in mortality prediction and assesses the influence of patient characteristics on model decisions. METHODS: A cohort of 4,337 adult patients (≥18 years) with RT-PCR-confirm

Retrospective Forecasting Using Provincial Grid Data and a Quota-Based Hybrid Physical-AI Model.

The global energy transition and ongoing electricity market reforms require power grid enterprises to balance reliable power supply with increasingly stringent transmission and distribution tariff regulations. Traditional budgeting methods based on historical extrapolation often fail to reflect the physical basis of asset operations, whereas data-driven machine learning models achieve high predictive accuracy but lack the transparency required for regulatory cost verification. To address the trade-off between forecasting accuracy and interpretability, this study proposes a hybrid cost-forecast

Reservoir computing for system identification and model predictive control.

Model predictive control (MPC), widely used for real-time control of complex dynamical systems, operates by repeatedly solving an optimization problem over a receding time horizon. Its success hinges on dynamical models that are accurate yet efficient enough for rapid online computation. Frequently, the governing models of complex systems are either unknown or computationally inefficient, forcing MPC to rely on data-driven surrogate models. Echo state networks (ESNs), a class of recurrent neural networks trained through computationally efficient ridge regression, are well-suited for this role

Systematic review and meta-analysis of machine learning models predicting massive hemorrhage protocol in trauma.

Timely activation of massive hemorrhage protocols (MHP) is critical to prevent exsanguination and improve survival in trauma patients. Current clinical tools for predicting MHP need have limited sensitivity and moderate specificity. Machine learning (ML) models may enhance early recognition of life-threatening hemorrhage. We conducted a systematic review and meta-analysis of studies that developed or validated ML models to predict MHP need in patients with traumatic injuries (PROSPERO CRD42024574891). Databases searched included MEDLINE, EMBASE, Web of Science, the Cochrane Central Register of

Prediction of Postoperative Complications Following Mandibular Fractures Using Machine Learning.

Postoperative complications in oral and maxillofacial surgery (OMFS), pose a significant challenge to patient recovery. Accurate prediction of such complications could enable earlier interventions and improve clinical outcomes. Advances in machine learning (ML) demonstrated potential in analysing complex data within healthcare, yet their application in OMFS remains limited. Here, the use of ML models, particularly random forest is explored, to predict postoperative complications for mandibular fractures, utilising demographic, clinical, and surgical data. With nested cross validation an accura

A data-driven approach to hepatitis C forecasting using machine learning and epidemiological models.

The hepatitis C virus is a significant global health concern and a major cause of chronic liver disease. Therefore, developing a mathematical model is essential for understanding, controlling and managing its transmission dynamics. In this paper, we developed a mathematical model and simulated the dynamics of transmission of the hepatitis C virus, considering alcohol use, a key factor contributing to increased damage to the liver of infected individuals. Computational investigation of nonlinear biological models by traditional numerical solvers is challenging due to their nonlinearity and inhe

Machine learning based prediction of gestational diabetes mellitus using early pregnancy biomarkers and clinical data.

Gestational diabetes mellitus (GDM) is a common pregnancy-related condition that can lead to significant maternal and neonatal complications, but conventional screening methods often delay diagnosis. Therefore, it is of interest to develop a machine learning model for early prediction of GDM using first-trimester biomarkers and clinical data. Hence, a prospective study of 100 pregnant women was conducted and various machine learning algorithms were trained to predict GDM. The random forest model showed the best performance with an accuracy of 86% and an AUC of 0.90. Thus, we show the potential

Mechanisms underlying dopamine-mediated reward bias in compulsive behaviors.

Pathological behaviors such as problem gambling or shopping are characterized by compulsive choice despite alternative options and negative costs. Reinforcement learning algorithms allow a computation of prediction error, a comparison of actual and expected outcomes, which updates our predictions and influences our subsequent choices. Using a reinforcement learning model, we show data consistent with the idea that dopamine agonists in susceptible individuals with Parkinson's disease increase the rate of learning from gain outcomes. Dopamine agonists also increase striatal prediction error acti
