Comprehensive analysis of the ubiquitome in rabies virus-infected brain tissue of Mus musculus.

Ubiquitination is an important post-translational modification (PTM) that plays a key role in almost every aspect of cellular processes and many signaling pathways in eukaryotes. In this study, we performed a quantitative ubiquitome study to identify the global change of ubiquitination induced by rabies virus (RABV) infection in the mouse brain tissue. 4,243 ubiquitinated sites, mapping to 1,626 proteins were identified; using a cutoff of fold change >2, 644 and 70 ubiquitinated proteins were up- and down-regulated, respectively. GO analysis indicated that the differentially ubiquitinated prot

RBM protein family: Focusing on the occurrence, progression and treatment of cancer.

The RNA-binding motif (RBM) protein family plays a crucial role in regulating RNA metabolism, including transcription, splicing, stability, and translation. Dysregulation of RBM proteins has been increasingly implicated in the initiation, progression, and therapeutic resistance of various cancers. This review summarizes current knowledge on the RBM protein family, with a focus on their involvement in alternative splicing and m6A RNA methylation-two key post-transcriptional mechanisms that influence cancer biology. Members such as RBM5 and RBM10 function as tumor suppressors by modulating apopt

Medically important differences in snake venom composition are dictated by distinct postgenomic mechanisms.

Variation in venom composition is a ubiquitous phenomenon in snakes and occurs both interspecifically and intraspecifically. Venom variation can have severe outcomes for snakebite victims by rendering the specific antibodies found in antivenoms ineffective against heterologous toxins found in different venoms. The rapid evolutionary expansion of different toxin-encoding gene families in different snake lineages is widely perceived as the main cause of venom variation. However, this view is simplistic and disregards the understudied influence that processes acting on gene transcription and tran

Targeted protein degradation: mechanistic diversity, therapeutic expansion, and clinical translation.

Targeted protein degradation (TPD) has transitioned from a paradigm-shifting concept to a clinically validated strategy, with multiple degraders achieving proof-of-concept in oncology and a rapidly expanding toolbox. By co-opting the ubiquitin-proteasome system to eliminate pathogenic proteins in an event-driven manner, TPD addresses targets long deemed undruggable. To capture this momentum, we present a comprehensive overview of the field. We dissect the mechanisms of heterobifunctional PROTACs and their clinical translation, covering late-stage programs targeting canonical oncogenic drivers

A druggable redox switch on SHP1 controls macrophage inflammation.

Immunological proteins are major disease targets, yet most remain undrugged. Post-translational redox modification of cysteine residues has emerged as an important mode of immune cell regulation, particularly in macrophage cytokine responses. Here, we develop a strategy for systematic discovery and small-molecule functionalization of redox-regulated cysteines on immunological proteins. Using deep redox proteomics, we annotate 788 in vivo redox-regulated cysteines across diverse immune-relevant protein domains. We demonstrate how these sites enable cysteine-directed pharmacology through discove

HDAC-mediated non-histone deacetylation as a central regulatory network integrating crop growth and stress adaptation.

Non-histone deacetylation is a widespread and reversible post-translational modification (PTM) dynamically regulated by histone acetyltransferases (HATs) and histone deacetylases (HDACs). Although initially characterized as a chromatin-associated mechanism controlling transcription, recent discoveries demonstrate that HDAC-mediated non-histone deacetylation extends far beyond gene regulation, targeting diverse transcription factors and functional proteins across cellular pathways. Here, we summarize advances in Arabidopsis, rice, and wheat recently, highlighting how non-histone deacetylation m

Aluminum chloride in Alzheimer's disease: A dual focus on molecular mechanisms and rat experimental models.

Alzheimer's disease (AD) is a leading cause of dementia among middle-aged and elderly individuals globally. Animal models of AD are widely used to investigate disease mechanisms and evaluate potential treatments for disease modification. Among non-genetically modified models, aluminum (Al3+) induced neurotoxicity has been widely employed to mimic key features of AD, including neuroinflammation and cognitive decline. This review comprehensively elucidates current evidence on the molecular and cellular mechanisms underlying Al3+-induced AD-like pathology, including amyloid-β accumulation, tau pr

Salicylic acid engages central metabolic regulators SnRK1 and TOR to govern immunity by differential phosphorylation of NPR1.

Immunity requires a delicate balance between combating infection and preserving metabolic functions. However, the mechanisms by which immune responses are coordinated with cellular metabolism remain largely unknown. Here, we show that NONEXPRESSER OF PR GENES 1 (NPR1), the central plant immune regulator of salicylic acid (SA)-mediated defense responses, is controlled by a cascade of post-translational modifications (PTMs) involving two master nutrient-sensing kinases. In the absence of pathogen challenge, TARGET OF RAPAMYCIN (TOR) inhibits NPR1 through phosphorylation at Ser-55/59. During defe

Dual regulatory roles of CPT1C in chronic stress-induced depression-related outcomes.

Regulation of α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid receptor (AMPAR) function has emerged as a novel strategy for major depressive disorder (MDD); however, the underlying molecular mechanisms remain unclear. Here, we demonstrate that enhanced GluA1 depalmitoylation in the nucleus accumbens mediates depressive-like behaviors following chronic stress, and identify that the dysfunction of carnitine palmitoyltransferase 1 C (CPT1C), a depalmitoylating enzyme that specifically depalmitoylates GluA1, mediated depression-like behaviors in mice. Furthermore, dopamine D2 receptor-express

The role of H2S and its derivatives in plant responses to low O2.

Hydrogen sulfide (H2S) is a gasotransmitter molecule that regulates essential plant biological processes. In the context of climate change and increasing flooding events, its role in plant responses to oxygen (O2) deficiency has gained significant attention. Accumulating evidence indicates that H2S exerts beneficial effects under hypoxia by modulating antioxidant defenses, mitochondrial function and hormonal signaling, mainly through its ability to refine protein activity via the post-translational modification (PTM) of persulfidation. Here, we discuss how this PTM is linked to the gasotransmi

Lactylation: A central metabolic-epigenetic driver of sepsis-associated acute kidney injury.

Sepsis-associated acute kidney injury is a lethal complication with limited therapeutic options. The discovery of protein lactylation, a novel modification where lactate modifies lysine residues, has redefined lactate from a metabolic waste product to a key epigenetic regulator. This review summarizes the role of lactylation as a central metabolic-epigenetic hub in SA-AKI pathogenesis. We detail its molecular basis, governed by a dynamic "writer-reader-eraser" network. In SA-AKI, lactylation orchestrates a multifaceted pathological cascade: it reprograms immune responses (e.g., altering macrop

NLRP3 inflammasome promotes PAD2/PAD4 release and protein citrullination in rheumatoid arthritis.

Anti-citrullinated protein antibodies (ACPAs), a hallmark of rheumatoid arthritis (RA), arise from citrullinated proteins presenting immunogenic neoepitopes that break self-tolerance. Citrullination is a post-translational modification catalyzed by peptidylarginine deiminases (PADs). Among five isoforms, PAD2 and PAD4 expressed in immune cells are most closely associated with RA pathogenesis. Although typically intracellular, PAD2 and PAD4 are detectable extracellularly in RA, yet the mechanisms governing their release remain unclear. Here, we show that NLRP3 inflammasome activation triggers P

Lysine l-Lactylation: Bridging Metabolism, Chromatin and Disease.

Lysine L-lactylation (KL-la) is a newly identified metabolite-derived post-translational modification that directly bridges cellular metabolic states to chromatin regulation and protein function. Mounting evidence shows that KL-la has pivotal roles in transcription regulation and diverse cellular processes and is implicated in multiple pathophysiological conditions. This review comprehensively examines KL-la across both histone and non-histone substrates in biology and disease. We first illustrate the historical development of KL-la and distinguish it from its isomers. We then delineate the en

The Enigma of Protein Succinylation in Diabetes.

Lysine succinylation is a dynamic post-translational modification that alters protein structure and function by adding a succinyl group to lysine residues. This review first presents current evidence on the discovery of lysine succinylation and its regulatory enzymes, and then focuses on its roles in diabetes and its complications. We summarize that succinylation is potentially governed by writers (HAT1), erasers (SIRT5), and readers (GAS41), linking metabolic state to signaling and epigenetic regulation. Dysregulation of succinylation is associated with mitochondrial dysfunction, oxidative st

The signaling upstream of DOT1L-mediated H3K79 methylation.

Histone lysine methylation is a post-translational modification system that has a profound impact on gene expression. The methylation of H3K79 plays an important role in gene transcription, repair of DNA damage, and cell cycle. In recent years, it has been discovered that Disruptor of telomeric silencing 1-like (DOT1L) can methylate H3K79 and is the only known H3K79 methyltransferase. DOT1L activity in leukemia cells interacts with ubiquitinated H2B to promote nucleosome binding. AF family proteins bind DNA to ENL to recruit DOT1L to the promoters of KMT2A (formerly MLL1)-rearranged leukemia t

HIV-1 infection induces Vif-mediated SUMOylation of host RNA splicing factors important for proper viral RNA splicing.

SUMOylation is a dynamically regulated post-translational modification involving covalent attachment of small ubiquitin-like modifiers (SUMOs) to lysine residues of target proteins. SUMOylation modulates multiple fundamental host cellular pathways, including pathways hijacked by HIV-1 to enable replication, but has not been explored by large-scale proteomics in the context of HIV-1 infection. Here, we performed a proteome-wide, mass spectrometry-based screen to identify proteins that are SUMOylated in response to HIV-1 infection. We show that infection with HIV-1 leads to the widespread increa

Targeting BRD4 bromodomains and beyond: exploring new therapeutic frontiers.

Bromodomain-containing protein 4 (BRD4) is a key transcriptional regulator in the bromodomain and extraterminal (BET) family. It promotes cancer and inflammation by binding to chromatin through its bromodomains. Although bromodomain inhibitors (e.g., BETi) show preclinical efficacy, their clinical application has been limited primarily by dose-limiting toxicities (DLTs), with resistance emerging as a secondary challenge. This motivates the development of strategies beyond conventional bromodomain inhibition. Here, we review emerging evidence that BRD4 sustains oncogenic programs through bromod

The xCT/CD98 complex suppresses ferroptosis in pan-cancer via a non-canonical RACK1-mediated iron homeostasis pathway.

Evasion of cell death drives tumor recurrence and metastasis, yet pan-cancer mechanisms of ferroptosis remain limited. We treated multiple tumor cell lines with ferroptosis inducers or anticancer agents and utilized post-translational modification proteomics including ubiquitination, acetylation, and phosphorylation during ferroptosis. We identify a significant increase in K280 ubiquitination of the ribosomal protein RACK1. Further pan-cancer cellular experiments suggest that RACK1 may suppress ferroptosis by regulating iron export via FPN1. Immunoprecipitation coupled with liquid chromatograp

Synthesis and anti-acute ischemic stroke effect of quinazoline-benzothiazole RIPK1 inhibitors.

Receptor-interacting protein kinase 1 (RIPK1) inhibitors show considerable translational potential for the treatment of neurodegenerative diseases; however, limited literature exists regarding their application in acute ischemic stroke (AIS). In this study, through structural modification, we designed and synthesized a series of RIPK1 inhibitors based on a benzothiazole-quinazoline core scaffold. Twenty compounds exhibited potent RIPK1 kinase inhibitory activity (IC50 < 100 nM), and five of these demonstrated remarkable anti-necroptotic efficacy at the cellular level (EC50 < 20 nM). Among them

Critical impact of lysine 136 in TDP-43 phase separation, compartmentalization, and aggregation in living vertebrates.

TDP-43 is a nuclear RNA-binding protein that undergoes liquid-liquid phase separation (LLPS) and forms insoluble aggregates in neurodegenerative diseases. By studying TDP-43 in living vertebrates, we confirmed that TDP-43 undergoes LLPS and forms dynamic biomolecular condensates in spinal motor neurons. We validated in vivo that interfering with the lysine residue at position 136 altered the phase separation behavior of TDP-43 by reducing cytoplasmic mislocalization and aggregation. These alterations were post-translational modification (PTM) independent, highlighting that residue 136 is a key
