The alterations of the Cyclic Alternating Pattern (CAP) recently found in narcoleptic adult patients suggest the presence of an impaired modulation of the fluctuations of the arousal level during their non-rapid eye movement (NREM) sleep, possibly because of the persistence of neurophysiological mechanisms typical of rapid eye movement (REM) sleep. The same mechanism might play a role in the occurrence of leg movement (LM) activity during sleep characterized by low levels of periodicity. The aim of this study was to evaluate CAP and sleep LM activity in a group of children and adolescents with

INTRODUCTION: Sleep disorders are common in individuals with kidney failure. Whether kidney impairment is the direct cause of sleep abnormalities is unclear, however, partly due to confounding factors including comorbidities, dialysis, and drugs.
METHODS: Here, we used a mouse model of acute kidney injury (AKI) transitioning to chronic kidney disease (CKD) induced by aristolochic acid to examine the effects of kidney impairment on sleep architecture. Each group, comprising 8~10 male mice, underwent cortical electroencephalogram (EEG) and electroencephalogram (EMG) recordings to measure sleep

BACKGROUND: Neuropathic pain is commonly associated with disturbances in sleep architecture and circadian rhythms, leading to fragmented sleep, body temperature fluctuations, and altered locomotion. While pregabalin and morphine are frequently prescribed for neuropathic pain management, their effects on sleep and circadian regulation are poorly understood.
METHODS: To identify the effects of spared nerve injury (SNI) on sleep architecture and circadian rhythms, male and female C57BL/6JRJ mice were implanted with wireless transmitters for continuous monitoring of electroencephalogram, electrom

Sleep has traditionally been conceptualized as a state of cognitive disconnection, yet emerging evidence indicates that decision-making capacities persist across sleep stages. Here, we elucidate the computational mechanisms underlying real-time lexical decision-making during polysomnographically-verified sleep, using facial electromyography and hierarchical drift diffusion modeling in both healthy individuals and participants with narcolepsy. We found that lexical decision-making was preserved during N1 and lucid REM sleep, but relied on distinct computational strategies: in N1 sleep, both enh

Orexin (Orx) and melanin-concentrating hormone (MCH) neurons in the lateral hypothalamic area are central to regulating the sleep-wake cycle and coordinating memory consolidation and forgetting through dynamic homeostatic mechanisms. This review systematically examines the functional characteristics of these two neural systems and their interactions: first, MCH neurons facilitate the transition from non-rapid eye movement (NREM) to rapid eye movement (REM) sleep at night via a dual regulatory mechanism and regulate the active forgetting of memories via hippocampal neural circuits; second, orex

BACKGROUND: Altitude training is used by elite athletes to improve sports performance, but it may also disrupt sleep. The aim of this study was to examine the effects of 2 weeks at high altitude on the sleep of young elite athletes.
METHODS: Participants (n=10) were members of the Australian under-17 soccer team on an 18-day (19-night) training camp in Bolivia, with six nights at near sea level in Santa Cruz (430 m) and 13 nights at high altitude in La Paz (3600 m). Sleep was monitored using polysomnography during a baseline night at 430 m and three nights at 3600 m (immediately after ascent,

Modulation of TASK-3 (Kcnk9) potassium channels affect neurotransmitter release in thalamocortical centers and other sleep-related nuclei having the capacity to regulate arousal cycles and REM sleep changes associated with mood disorders and antidepressant action. Circumstantial evidence from this and previous studies suggest the potential for TASK-3 to be a novel antidepressant therapeutic target; TASK-3 knock-out mice display augmented circadian amplitude and exhibit sleep architecture characterized by suppressed REM activity. Detailed analysis of locomotor activity indicates that the amplit

Electrodermal Activity (EDA) is a measure of skin conductance that reflects autonomic nervous system activity, offering insight into stress, emotional regulation, and autonomic arousals. This study investigates EDA fluctuations and sleep storms in older patients (≥50 years) with epilepsy, a population that has been underrepresented in previous research. We analyzed concurrent EEG and EDA recordings from 18 patients with 31 recording samples to characterize EDA events, particularly their distribution across sleep stages and quadrants of the night. Our findings reveal that Electrodermal Response

PURPOSE OF REVIEW: Sleep and cluster headache (CH) are believed to be interconnected but the precise relation to the other trigeminal autonomic cephalalgias (TACs) is uncertain and complex. A better understanding of these relations may eventually lead to a clarification of the underlying mechanisms and eventually to more effective therapeutic regimens. This review aims to evaluate the existing literature on the subject of TACs and sleep. An association between episodic CH and distinct macrostructural sleep phases, especially the relation to rapid eye movement (REM) sleep, has been described in

BACKGROUND: Lewy body disease (LBD), including Parkinson's disease (PD) and dementia with Lewy bodies (DLB), is defined pathologically as degeneration in the central and peripheral nervous system associated with Lewy bodies. Somatic symptom disorder often predates the clinical diagnosis of PD and DLB. It is crucial to make an initial diagnosis of LBD in patients with psychiatric symptoms because administering psychotropic drugs often causes or exacerbates extrapyramidal signs. Given the close association between rapid eye movement (REM) sleep behaviour disorder and LBD, REM sleep without atoni

Chronic opioid use is associated with profound sleep disturbances that may contribute to relapse in opioid use disorder (OUD). However, how opioid self-administration and abstinence alter sleep architecture in a sex-dependent manner remains poorly understood. Here, we examined the effects of volitional fentanyl intravenous self-administration (IVSA) on sleep-wake architecture in male and female mice across drug-taking and abstinence phases. Continuous home-cage monitoring revealed sex differences in baseline sleep, with females exhibiting increased wakefulness and reduced NREM sleep relative t

Sleep is regulated by complex biological systems and environmental influences, neither of which is fully clarified. This study demonstrates differential effects of partial sleep deprivation (SD) on sleep architecture and psychomotor vigilance task (PVT) performance using two different protocols (sequentially) that each restricted daily sleep to 3 hours in healthy adult men. The protocols differed only in the period of sleep restriction; in one, sleep was restricted to a 3-hour block from 12:00 AM to 3:00 AM, and in the other, sleep was restricted to a block from 3:00 AM to 6:00 AM. The present article is meant to suggest an approach to the guidelines for the therapy of sleep disturbances in Parkinson's Disease (PD) patients. The factors affecting the quality of life in PD patients are depression, sleep disturbances and dependence. A large review of the literature on sleep disturbances in PD patients, provided the basis for the following classification of the sleep-arousal disturbances in PD patients. We suggest a model based on 3 steps in the treatment of sleep disturbances in PD patients. This model allowing the patient, the spouse or the caregiver a quiet sleep at night

The review article on posttraumatic stress disorder (PTSD) and sleep provides a comprehensive overview of circadian genes in PTSD-related sleep disturbances, drawing insights from patient studies. PTSD is often accompanied by significant sleep-related disturbances, which can manifest as nightmares, disrupted sleep continuity, and alterations in sleep architecture, particularly changes in Rapid Eye Movement (REM) sleep-regulation. Despite the well-documented symptomatic complaints of poor sleep among PTSD patients, the underlying mechanism remains poorly understood. This review underscores the

We report the case of a 46-year-old male with myotonic dystrophy who developed daytime hypersomnia and dyspnoea. After a therapeutic tracheostomy, overnight polysomnographic studies were performed under three different ventilatory conditions. When the patient breathed spontaneously through a tracheal cannula, abnormal cyclical sleep increased and rapid eye movement (REM) sleep decreased markedly. The apnoea and hypopnoea index (AH index) was 35.1. When breathing spontaneously through his normal airway, there were many instances of cyclical sleep, but few instances of deep sleep and no episodes

Functional imaging of human sleep has been performed with nuclear medicine methods, but MRI has been difficult to implement, in part because of the noise associated with echo-planar imaging as well as the difficulty in reading physiologic signals in the MRI environment. We describe a silent MR sequence that can record brain activation over many hours with simultaneous acquisition of an EEG. This shows activation of occipital cortex and deactivation of frontal cortex during REM sleep, in agreement with previous studies using other techniques. MRI-Sleep-REM sleep.

OSA is known to increase the risk for SUDEP in persons with epilepsy, but the relationship between these two factors is not clear. Also, there is no study showing the acute responses to obstructive apnea in a chronic epilepsy model. Therefore, this study aimed to characterize cardiorespiratory responses to obstructive apnea and chemoreceptor stimulation in rats. In addition, we analyzed respiratory centers in the brain stem by immunohistochemistry. Epilepsy was induced with pilocarpine. About 30-60 days after the first spontaneous seizure, tracheal and thoracic balloons, and electrodes for rec
