Radioligand therapy in combination with CAR T cells overcomes the heterogeneous immunosuppressive prostate tumor microenvironment.

UNLABELLED: 177 Lu-PSMA-617 (Pluvicto TM , Lu-177 RLT) is an FDA-approved targeted radioligand therapy (RLT) for metastatic castration-resistant prostate cancer (mCRPC), but its durability of response to this singular approach poses a challenge to the field. Chimeric antigen receptor (CAR) T cell therapy has revolutionized clinical practice for hematological malignancies, but its clinical development for solid tumors, including mCRPC, has been encumbered by antigen heterogeneity and the immunosuppressive tumor microenvironment (TME). Here, we evaluate the therapeutic combination of Lu-177 RLT …

Activating mutations in ESR1 contribute to an immunosuppressive breast tumor microenvironment by dampening cytokine secretion.

Patients with estrogen receptor+ (ER+, ESR1+) breast cancer are most at risk of relapse, where activating mutations in ESR1 promote metastas … and IL-1β. Collectively, our findings reveal that ESR1 mutations contribute to an immunosuppressive tumor microenvironment by dampening cytokine secretion and immune cell activity.

Pulsed photobiomodulation reprograms the tumor immune microenvironment to restore local T cell-mediated antitumor immunity.

Developing strategies to precisely modulate the tumor immune microenvironment (TME) is crucial for advancing immunotherapy. Here, we reported that 980-nm pulsed photobiomodulation (PBM) inhibits early tumor growth by locally reinvigorating antitumor immunity, without direct cancer cell cytotoxicity. In syngeneic LLC and MC38 models, daily PBM delayed tumor growth and conferred durable protection upon rechallenge without systemic toxicity, while efficacy was dependent on CD8+ T cells. Single-cell RNA sequencing and flow cytometry revealed that PBM directly alleviates CD8+ T cell exhaustion and

Tumor microenvironment transcriptional activity enables robust stratification of chemotherapy response in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) exhibits heterogeneous treatment responses, yet molecular subtypes based on predefined biological pathways show limited prognostic value. We introduce tumor-specific total mRNA expression (TmS), a pathway-agnostic deconvolution metric derived from matched RNA/DNA sequencing, as a robust stratification tool. Analyzing 575 TNBC patients across Western and East Asian populations, TmS outperforms established subtypes in predicting chemotherapy outcomes, stratifying patients into high TmS with favorable prognosis and low TmS with poor prognosis. Stromal enrichme

Recent advances in dissolving microneedles for breast cancer immunotherapy: local delivery and tumor microenvironment remodeling.

Breast cancer is generally characterized by limited immunogenicity. Although immune checkpoint inhibitors (ICIs) provide survival benefits in a subset of patients with triple-negative breast cancer (TNBC), overall response rates remain limited and are accompanied by pronounced therapeutic heterogeneity and systemic immune-related adverse events. The immunosuppressive tumor immune microenvironment (TIME), characterized by restricted antigen presentation, limited effector T-cell infiltration, and enrichment of immunosuppressive cell populations, represents a key barrier to durable immunotherapy

VG161 oncolytic virus remodels the tumor microenvironment by expanding CD3+ macrophages and enhancing antitumor immunity.

Oncolytic virotherapy has emerged as a promising strategy for cancer immunotherapy; however, its immunomodulatory mechanisms in colorectal cancer remain incompletely understood. Here, we evaluated the therapeutic effects of the oncolytic virus VG161 in murine colorectal cancer models (CT26.WT and MC38) and systematically characterized the associated tumor microenvironment (TME) remodeling. Using single-cell RNA sequencing, flow cytometry, and multiplex immunohistochemistry, we identified a distinct population of CD3+ macrophages that was markedly expanded following VG161 treatment. Functional …

NIR-II Imaging-Guided Phototherapy-Induced Senescence Reprograms Immunosuppressive Tumor Microenvironment for Potent Cancer Immunotherapy.

Cellular senescence, characterized by irreversible cell-cycle arrest, is increasingly recognized as a pivotal mechanism in tumor suppression through regulating the tumor microenvironment (TME). Nevertheless, strategies that exploit senescence inducers to promote senescence-associated secretory phenotypes (SASPs) expression and thereby enhance immune infiltration remain underexplored. In this work, we present a phototherapy-based senescence induction approach to overcome the immunosuppressive TME in triple-negative breast cancer. We developed a reactive oxygen species-responsive nanoplatform th…

Highly CD81-expressing stromal cells characterize a tumor-suppressive immune microenvironment in invasive breast cancer.

BACKGROUND: CD81, a member of the tetraspanin family, is implicated in tumor progression, and its elevated expression in tumor cells has been associated with poor prognosis across multiple cancers. However, the functional significance of CD81 within the stromal compartments of the tumor microenvironment (TME) remains unclear. This study investigated the clinical relevance and mechanistic role of stromal CD81 (sCD81) in invasive breast cancer. METHODS: A total of 462 human primary invasive breast cancer samples, along with 13 normal breast tissues and 10 ductal carcinoma in situ samples collec…

Tumor microenvironment-activated nanoplatform via Fe3+-mediated self-assembly for synergistic chemodynamic/gene/chemo therapy.

Nucleic acid-based nanotherapeutics hold great promise for tumor treatment, yet face challenges in drug loading capacity, stability during delivery, and controlled intracellular release. Herein, we propose a carrier-free, tumor microenvironment (TME)-responsive nanoplatform (FAM NPs-FA) constructed via coordination-driven self-assembly of Survivin antisense oligodeoxynucleotide (ASO), methotrexate (MTX), and Fe3+ ions. The resulting nanocomposites demonstrate high drug loading, enhanced serum stability, and intrinsic synergistic activity by integrating chemodynamic, gene, and chemotherapeutic …

A 5-Fluorouracil-Constituted DNA Hydrogel Embedded with Quercetin Remodels Tumor Microenvironment for Robust Chemoimmunotherapy.

Chemoimmunotherapy has emerged as a promising treatment modality for triple-negative breast cancer (TNBC). However, its clinical utility is often hindered by the severe toxicity of chemotherapeutic agents and the immunosuppressive nature of the tumor microenvironment (TME). Herein, we engineer an injectable 5-fluorouracil-constituted DNA hydrogel embedded with quercetin (Q-5FDHG) through a novel DNA amplification reaction to navigate these impediments in a dual-pronged manner. Q-5FDHG ensures progressive enzymatic degradation, which continuously releases 5-fluorouracil (5FU) and quercetin (Que…

Multi-omics reveals tumor microenvironment heterogeneity and therapeutic vulnerabilities in peritoneal metastasis of gastric cancer.

Peritoneal metastasis of gastric cancer (PMGC) is associated with a poor prognosis, with approximately one-third of patients presenting with peritoneal metastasis at diagnosis. Current systemic therapies have limited efficacy, underscoring the need for precise therapeutic targets in PMGC. We employed single-cell RNA sequencing (scRNA-seq) to characterize the cellular composition of the PMGC microenvironment, reconstruct epithelial cell developmental trajectories, and analyze intercellular communication networks. Spatial transcriptomics and multiplex immunohistochemistry were integrated to vali

Targeting glioblastoma mitochondrial metabolism with S-Gboxin induces cytotoxicity under conditions of the tumor microenvironment.

Glioblastoma (GB) is the most common primary malignant brain tumor in adults. Gboxin, a novel compound that targets oxidative phosphorylation via complex V inhibition, has shown promise in preclinical models of GB. We examined the efficacy of the pharmacokinetically optimized S-Gboxin under conditions replicating the GB microenvironment, including nutrient deprivation and hypoxia. We assessed cytotoxicity and growth-inhibitory effects of S-Gboxin in human GB cell lines, primary GB cultures, as well as immortalized and primary human astrocytes under different nutrient and oxygen deprivation sce

Tumor-Regional Immune Microenvironment: A Critical Factor in the Design of Radiotherapy-Immunotherapy Combination Trials.

Clinical trials combining radiotherapy (RT) with immune checkpoint blockade (ICB) have shown improved outcomes in only a fraction of patients, and optimal strategies for integrating these modalities remain under intense investigation. With a few exceptions, phase III combination trials have yielded disappointing results. This may be due to detrimental effects of RT on the tumor-regional immune microenvironment (TRIME), including tumor-draining lymph nodes (TDLNs) and intratumoral immune aggregates such as tertiary lymphoid structures. TDLNs are crucial for generating tumor-specific T cells, in

Multifaceted roles of chemokines in gastric cancer: From tumor microenvironment modulation to therapeutic targeting (Review).

Gastric cancer (GC) is a malignant solid tumor with limited treatment options, particularly for advanced tumor stages. Chimeric antigen receptor (CAR)-T therapy has been successful in treating hematological malignancies; however, its efficacy in solid tumors remains suboptimal due to the complex tumor microenvironment (TME). The TME in GC comprises diverse signaling molecules and cellular components, including fibroblasts, immune cells and stromal proteins; these components can support tumor growth, invasion and immune evasion. In the GC TME, the dynamic interactions between cytokines, chemoki…

Understanding immune checkpoint inhibitor efficacy through spatial decoding of the lung cancer tumor immune microenvironment.

Intervertebral disc degeneration (IDD) is closely associated with aging. Although the Runt-related transcription factor 1 (RUNX1) is well known for its role in skeletal development and other musculoskeletal related diseases such as osteoarthritis, its involvement in IDD pathogenesis remains elusive. In this study, we examined the function of Runx1 specifically in the nucleus pulposus (NP) in vivo. To achieve NP-specific postnatal overexpression of Runx1, we crossed Krt19CreERT mice with Rosa26-Runx1 transgenic mice previously generated in our laboratory. Mice with NP specific Runx1 overexpress

Osteocyte-Derived GDF15 Promotes Paclitaxel Resistance, Tumor Growth, and Bone Microenvironment Regulation in Prostate Cancer.

With changes in lifestyle and dietary patterns, the incidence of prostate cancer in China has been increasing steadily, and it has become one of the most common malignancies in men. Chemotherapy remains a primary treatment for prostate cancer, but the subsequent development of drug resistance by tumor cells markedly compromises its therapeutic efficacy. Growth differentiation factor 15 (GDF15) has been shown to be closely associated with tumor cell proliferation and metastasis; however, its contribution to the drug resistance of prostate cancer cells within the tumor microenvironment has not b…
