Measurement of circulating immunoreactive alpha-atrial natriuretic peptide in human plasma: validation and clinical application.

Recent evidence suggests that plasma alpha-atrial natriuretic peptide (alpha-ANP) may serve as a useful biochemical marker of severe heart failure, as circulating levels become ighly elevated. In the present study, a specific radioimmunoassay (RIA) protocol was adapted and optimised for rapid quantitative measurement of circulating alpha-ANP levels in plasma taken from clinically normal subjects and heart transplant recipients with no evidence of heart failure or heart transplant rejection. The plasma concentration of immunoreactive alpha-ANP in heart transplant recipients (115 +/- 10 pg/ml, n

Integration of Multiomics Datasets to Identify Allergy Biomarkers and Protective Signatures.

The identification of biomarkers for diseases such as allergies requires analytical strategies capable of handling high-dimensional and heterogeneous biological data. Advances in omics technologies have enabled the systematic exploration of molecular features across multiple biological levels. However, their effective integration and interpretation remain challenging. In this chapter, we present a data-driven bioinformatic framework for biomarker discovery based on single-omics and multi-omics datasets, with a particular focus on allergy-related applications. We describe complementary strategi

From exposure to embedding: how interconnected social determinants drive health inequalities-a complex adaptive system dynamics approach.

The social determinants of health (SDOH)-including socioeconomic position, education, housing, and social networks-are fundamental drivers of health inequalities worldwide. These inequalities persist because social determinants interact dynamically across time and context, with health emerging from the interplay of social, political, environmental, and biological processes in which determinants act simultaneously as causes and consequences. Early-life exposures and historical conditions produce enduring physiological vulnerability through biological embedding, encompassing stress dysregulation

GD2TIF cells as a platform for single-dose and long-term delivery of biologics.

The efficacy of biologics, such as peptide and protein drugs, is often limited by their short half-lives in vivo, necessitating repeated infusions to maintain therapeutic effects. Here, we demonstrate that long-lived CAR T cells can be leveraged as a versatile platform for long-term delivery of biologics. Our recent findings show that the depletion of BCOR and ZC3H12A induces GD2 CAR T cells into an immortal-like and functional state, which we have termed GD2TIF cells. These GD2TIF cells expanded in immunocompetent mice without the need for chemotherapeutic conditioning and persisted as a poly

Treating temporomandibular disorders with permanent mandibular repositioning: is it medically necessary?

In this paper, the authors review the rationale and history of mandibular repositioning procedures in relation to temporomandibular disorders (TMDs) as these procedures have evolved over time. A large body of clinical research evidence shows that most TMDs can and should be managed with conservative treatment protocols that do not include any mandibular repositioning procedures. Although this provides a strong clinical argument for avoiding such procedures, very few reports have discussed the biologic reasons for either accepting or rejecting them. This scientific information could provide a b

Generation and biological activities of oxidized phospholipids.

Glycerophospholipids represent a common class of lipids critically important for integrity of cellular membranes. Oxidation of esterified unsaturated fatty acids dramatically changes biological activities of phospholipids. Apart from impairment of their structural function, oxidation makes oxidized phospholipids (OxPLs) markers of "modified-self" type that are recognized by soluble and cell-associated receptors of innate immunity, including scavenger receptors, natural (germ line-encoded) antibodies, and C-reactive protein, thus directing removal of senescent and apoptotic cells or oxidized li

Synthesis of functionalized copillar[4+1]arenes and rotaxane as heteromultivalent scaffolds.

In this study, novel copillar[4+1]arenes were used as central heteromultivalent scaffolds via orthogonal couplings with a series of biologically relevant molecules such as carbohydrates, α-amino acids, biotin and phenylboronic acid. Further modifications by introducing maleimides or cyclooctyne groups provided molecular probes adapted to copper-free click chemistry. An octa-azidated fluorescent rotaxane bearing two distinct ligands was also generated in a fully controlled manner.

Dynamic changes in the metabolic network and signalling pathways of cancer stem cells in therapy resistance.

Cancer stem cells (CSCs) play a pivotal role in therapeutic resistance, metastasis, and tumour recurrence. During treatment, these cells undergo dynamic changes that enable survival and metabolic adaptation, leading to poor clinical outcomes. CSCs are capable of developing resistance to drugs by alterations in the metabolic and signalling pathways. They exhibit metabolic adaptations that enable therapy resistance, tumour recurrence, and metastasis. These adaptations involve shifts in glycolysis, oxidative phosphorylation (OXPHOS), lipid and amino acid metabolism, mitochondrial dynamics, and in

Genetic characterization of highly similar H9N2 viruses isolated from swine and poultry in China.

H9N2 avian influenza viruses (AIVs) are widely distributed in poultry and have attracted continued attention because of their high reassortment potential and ability to cross species barriers. In March 2019, two H9N2 viruses were isolated from a pig and a chicken, respectively, in Shandong Province, China. In this study, we characterized the whole-genome features, phylogenetic relationships, molecular characteristics, and pathogenicity of the two viruses in mice. Whole-genome sequence analysis showed that the eight gene segments of the two isolates shared 99.9%-100% nucleotide sequence identit

Population Genomics of Almond (Prunus dulcis) Reveals Region-Specific Selection and a Complex History of Domestication.

The domestication of perennial crops in the Mediterranean Basin remains unclear, particularly regarding the genomic consequences of human-mediated demographic shifts and selection. We analysed 8.1 million single nucleotide polymorphisms from 96 cultivated almond (Prunus dulcis) accessions from Europe, North America, Central Asia, and New Zealand, alongside four wild relatives. Population structure analyses revealed four geographically differentiated cultivated groups (Central Asian, North American, and two European) and three wild populations (P. spinosissima, P. orientalis, and P. fenzliana).

Base excision repair in Staphylococcus aureus: a defense against oxidative DNA damage and a potential target for novel antibiotics.

Staphylococcus aureus (S. aureus) remains a major clinical challenge due to its high virulence, persistence, and remarkable capacity to develop antibiotic resistance. Survival within the host requires adaptation to oxidative and nitrosative stress generated by immune cells, which inflict extensive damage on bacterial DNA, including oxidized bases, abasic sites, and strand breaks. Under these conditions, the base excision repair (BER) pathway plays a central role in maintaining genome integrity and supporting bacterial survival under immune and antibiotic-induced stress. This review examines th

Genome-wide identification and functional characterization of the Magnesium Transporter (MGT) gene family and its expression patterns to different anionic magnesium stresses in Yinshania henryi.

BACKGROUND: Magnesium (Mg) is an essential macronutrient required for numerous physiological and biochemical processes in plants. Both Mg deficiency and excess can disrupt plant growth and development, affecting a wide range of biological processes. Mg uptake and distribution are primarily mediated by the magnesium transporter (MGT) family. Karst landscape soils are typically enriched in calcium and magnesium, often leading to greater Mg uptake by plants than in non-karst environments. Yinshania henryi, a member of the Brassicaceae family endemic to the magnesium-enriched karst areas of southw

Influences of Ruminococcus bromii and Peptostreptococcaceae on voluntary exercise behavior in a rodent model.

INTRODUCTION: This study investigates the relationship between the gut microbiome and voluntary exercise, focusing on wheel running activity in a rat model. The gut microbiome plays a crucial role in host physiology, homeostasis, and behavior. Alterations in the gut microbiome have been linked to various pathological states and health conditions, including obesity. METHODS: Given the strong association between physical inactivity and obesity development, our study aimed to identify microbiome factors associated with elevated levels of voluntary exercise. Male Sprague Dawley rats were used in

Allicin improves the function of cardiac microvascular endothelial cells by increasing PECAM-1 in rats with cardiac hypertrophy.

OBJECTIVE: Cardiac microvascular damage is significantly associated with the development of cardiac hypertrophy (CH). Researchers found that allicin could inhibit CH, but the relationship between cardiac microvessel and the inhibition of allicin on CH has not been reported. We aimed to investigate the effect of allicin on the function of cardiac microvascular endothelial cells (CMECs) in CH rat. MATERIALS AND METHODS: The hemodynamic parameters were measured by BL-420F biological function experimental system and the indicators of the ventricular structure and function were measured by echocar

Silicon central pattern generators for cardiac diseases.

Cardiac rhythm management devices provide therapies for both arrhythmias and resynchronisation but not heart failure, which affects millions of patients worldwide. This paper reviews recent advances in biophysics and mathematical engineering that provide a novel technological platform for addressing heart disease and enabling beat-to-beat adaptation of cardiac pacing in response to physiological feedback. The technology consists of silicon hardware central pattern generators (hCPGs) that may be trained to emulate accurately the dynamical response of biological central pattern generators (bCPGs

Evolutionary concepts in ecotoxicology: tracing the genetic background of differential cadmium sensitivities in invertebrate lineages.

In many toxicological and ecotoxicological studies and experimental setups, the investigator is mainly interested in traditional parameters such as toxicity data and effects of toxicants on molecular, cellular or physiological functions of individuals, species or statistical populations. It is clear, however, that such approaches focus on the phenotype level of animal species, whilst the genetic and evolutionary background of reactions to environmental toxicants may remain untold. In ecotoxicological risk assessment, moreover, species sensitivities towards pollutants are often regarded as rand

Metabolic and functional connectivity changes in mal de debarquement syndrome.

BACKGROUND: Individuals with mal de debarquement syndrome (MdDS) experience a chronic illusion of self-motion triggered by prolonged exposure to passive motion, such as from sea or air travel. The experience is one of rocking dizziness similar to when the individual was originally on the motion trigger such as a boat or airplane. MdDS represents a prolonged version of a normal phenomenon familiar to most individuals but which persists for months or years in others. It represents a natural example of the neuroplasticity of motion adaptation. However, the localization of where that motion adapta

Resolution of the species problem in African trypanosomes.

There is a general assumption that eukaryote species are demarcated by morphological or genetic discontinuities. This stems from the idea that species are defined by the ability of individuals to mate and produce viable progeny. At the microscopic level, where organisms often proliferate more by asexual than sexual reproduction, this tidy classification system breaks down and species definition becomes messy and problematic. The dearth of morphological characters to distinguish microbial species has led to the widespread application of molecular methods for identification. As well as providing

Why does digitally predicted tooth movement fail? the biology-digital mismatch in orthodontics: a critical narrative review.

Digital orthodontics has advanced through intraoral scanning, CAD/CAM, virtual planning, and clear aligners; however, discrepancies remain between predicted and actual tooth movements. This narrative review critically examined the biological and biomechanical factors underlying the limited predictability of digitally planned orthodontic treatment. A structured literature search of PubMed, Scopus, and Google Scholar was conducted and peer-reviewed articles were critically analyzed. Current digital systems primarily operate on geometric and algorithm-based models that assume linear and standardi
