Ammonia downregulates GLAST mRNA glutamate transporter in rat astrocyte cultures.

Abnormalities in glutamate metabolism and glutamatergic neurotransmission appear to play an important role in the pathogenesis of hyperammonemia and hepatic encephalopathy. Previous studies from our laboratory have shown that treatment of cultured astrocytes with NH4Cl result in decreased glutamate uptake. To extend these observations, we performed a Northern blot analysis of the astroglial glutamate transporter GLAST (EAAT1) in NH4Cl-treated primary rat astrocyte cultures. Following treatment with 2, 5, 10 mM NH4Cl for 3 days, cortical astrocytes showed a 22, 29, 36% decrease in GLAST mRNA, r…

Regulatory volume decrease in primary astrocyte cultures: relevance to methylmercury neurotoxicity.

Because of the multiple and varied roles of astrocytes in brain homeostasis, primary cultures of astrocytes from neonatal rat brains have proven to be an excellent model for the study of in vitro cell functions and control mechanisms. In addition, their ability to preferentially sequester a number of heavy metals, such as methylmercury, has lead to intense research on their potential to modulate heavy metal-induced dysfunction. In the present review we briefly discuss the mechanisms associated with astrocytic swelling, an early and prominent event in brain injury, followed by a description on …

Occupational exposure to chlorinated aliphatic hydrocarbons and risk of astrocytic brain cancer.

Chlorinated aliphatic hydrocarbons (CAHs) were evaluated as potential risk factors for astrocytic brain tumors. Job-exposure matrices for six individual CAHs and for the general class of organic solvents were applied to data from a case-control study of brain cancer among white men. The matrices indicated whether the CAHs were likely to have been used in each industry and occupation by decade (1920-1980), and provided estimates of probability and intensity of exposure for "exposed" industries and occupations. Cumulative exposure indices were calculated for each subject. Associations of astrocy

Extracellular guanosine increases astrocyte cAMP: inhibition by adenosine A2 antagonists.

Both extracellular guanosine and adenosine stimulated astrocyte proliferation in vitro and increased intracellular cAMP 6-fold within 2 min. The effects of both guanosine and adenosine on proliferation and cAMP levels were inhibited by antagonists of adenosine A2 receptors but augmented by A1 receptor antagonists. The correlation between cAMP accumulation and stimulation of cell proliferation by adenosine and guanosine indicates that increased intracellular cAMP may be one of the second messengers involved in these effects. Guanosine is not an adenosine A2 receptor agonist and does not activat

Congenital cerebral primitive neuroectodermal tumor with astrocytic differentiation and extracranial metastases.

A cerebral primitive neuroectodermal tumor with astrocytic differentiation and extracranial metastases in a 28-day-old infant is reported. The infant presented with a progressively enlarged head, cutaneous lesions in the neck, and enlarged cervical lymph nodes. A computed tomography brain scan demonstrated a giant thalamic tumor with subarachnoid dissemination and hydrocephalus. Biopsy material from the cervical lesions showed a picture of glioma with anaplastic astrocytes. The patient received a ventriculoperitoneal shunt operation and palliative chemotherapy, but died at 3 months of age. Aut…

[Clinical aspects and follow-up in astrocytic hamartoma of the retina and optic papilla].

The clinical picture and the results of follow-up examinations in 21 patients with an astrocytic hamartoma of the retina and/or disk are described. Fifty-one tumors were found; 7 of them (in 5 patients) initially appeared as homogeneous, whitish tumors, but later assumed a mulberry appearance. Follow-up in these patients was between 9 and 19 years. Depigmented lesions of the skin ("White Spots") were frequently seen at an early age and were especially helpful when a retinoblastoma had to be excluded in the differential diagnosis.

Astrocyte maturation creates a stage-specific niche that instructs hippocampal neurogenesis.

Astrocytes are a major part of the hippocampal stem cell niche and have been shown to control several steps of neurogenesis, from neural stem and precursor cell (NSPC) proliferation to the differentiation and maturation of newborn neurons. With the aim to identify new factors by which astrocytes promote neurogenesis, we isolated hippocampal astrocytes across key postnatal stages (p3, p10, and p14) and collected astrocyte conditioned medium (ACM) while keeping them for 1 week in culture. Depending on the developmental stage of the astrocytes, we uncovered a shift in the astrocyte secretome. Spe…

A Review of the Molecular Evidence for Near-Infrared Light in Astrocyte-Mediated Neurorehabilitation.

Near-infrared (NIR) light represents a promising, noninvasive therapeutic for neurological disorders, predicated on its capacity to modulate astrocyte function. As the primary glial population in the central nervous system, astrocytes are indispensable for maintaining neuronal homeostasis, regulating glutamate metabolism, and supporting metabolic energetics. This review synthesizes recent advancements in NIR-driven neurorehabilitation, elucidating the molecular signaling cascades and transduction pathways activated within the astrocytic network. We specifically examine translational applicatio…

Astrocytic facilitation of somatic NMDARs underpins hippocampal E-S potentiation.

EPSP-spike (E-S) potentiation is a form of plasticity defined by a persistent increase in neuronal spike output for a given synaptic input, reflecting enhanced E-S coupling independent of synaptic strength. However, its cellular mechanisms remain poorly understood. Here, we show that brief induction of E-S potentiation triggers a sustained calcium activity increase in astrocytes adjacent to the hippocampal CA1 pyramidal layer. Mechanistically, E-S potentiation critically requires GluN2B-containing N-methyl-D-aspartate receptors (NMDARs) localized to the neuronal soma. Sustained astrocytic acti…

Astrocytic frataxin deficiency drives neurocognitive impairment in sickle cell mice.

Individuals with sickle cell disease (SCD) suffer from a high burden of neurocognitive impairment. Cerebrovascular lesions, characterized by diffusion tensor imaging identified neuroaxonal damage, are associated with learning and memory deficits. The molecular and cellular intermediates driving microstructural neuroaxonal damage and cognitive impairment in SCD remain largely unexplored. Herein, we report that sickle cell mice (SS) have reduced expression of frataxin (FXN), a mitochondrial protein, in their astrocytes compared with normal control (AA) mice. A newly generated sickle bone marrow …

Context-Dependent modulation of astrocytic Ca2+ signals by mitochondria: a computational study.

Mitochondria regulate intracellular Ca2+ by uptake through the mitochondrial Ca2+ uniporter (MCU) and release via the mitochondrial permeability transition pore (mPTP). In astrocytes, neurotransmitter stimulation evokes Ca2+ signaling, yet the role of mitochondria in shaping these responses remains unclear. We extended a compartmental astrocyte model developed by our group to include MCU- and mPTP-mediated dynamics, and simulated glutamatergic and dopaminergic inputs modeled as Poisson processes driving IP3 synthesis through the phospholipase-C (PLC) pathway. Both unipolar and bifurcated-termi…

A fractional order model for amyloid beta driven astrocyte calcium signaling.

BACKGROUND: Idiopathic membranous nephropathy (MN) is a major cause of nephrotic syndrome. Conventional treatment strategies induce remission but the relapse rates are high. Different doses of rituximab (RTX) appeared effective in reducing proteinuria in MN but long-term follow-up data are rare. METHODS: Since 2006, a total of 14 patients (median age 51 (26 - 69) years, 4 women, 10 men) with biopsy-proven MN (1 - 4 relapses, MN since 4 (1 - 13) years) were treated with RTX (4 doses of RTX 375 mg/m2 on Days 0, 30, 60, 90). All patients had prior immunosuppressive therapy with Cyclosporin A, 7

Astrocytic D1 Dopamine-Signaling Regulates Synaptic Remodeling and Cocaine Seeking.

D1-type receptor (D1R)-mediated dopaminergic signaling within the nucleus accumbens shell (NAcSh) is essential for forming adaptive circuit changes that embed persistent memories associated with drug seeking and craving. While D1R is expressed in both NAcSh neurons and astrocytes, the specific contribution of astrocytic D1R to drug-related circuit plasticity remains poorly understood. Here, we demonstrate in mouse NAcSh slices that D1R agonists increase astrocytic Ca 2+ activity, an effect that is attenuated by astrocyte-specific D1R knockdown. Furthermore, selective knockdown of astrocytic D1…

Cinnamaldehyde improves the pathology of vascular cognitive impairment via astrocytic TRPA1.

Vascular cognitive impairment (VCI) comprises cognitive dysfunction caused by vascular pathology and is associated with various forms of dementia. VCI is typically caused by chronic cerebral hypoperfusion (CCH), which is accompanied by oxidative stress, central nervous system inflammation, microglial activation, and white matter injury. Therapeutic targets for VCI remain largely unknown. Transient receptor potential ankyrin 1 (TRPA1) is functionally expressed in the brain, where it senses changes in oxygen levels and is activated by hypoxia and hyperoxia. We investigated the pathophysiological

Focal astrocyte loss reveals nuclear translocation during lesion repopulation.

Astrocyte loss occurs in various neurological conditions and can disrupt local tissue homeostasis. While astrocytes surrounding border-forming lesions adopt reactive states without restoring astrocyte networks, how astrocytes respond to spatially confined astrocyte loss remains poorly understood. Here we used longitudinal in vivo two-photon microscopy, combined with spatiotemporal transcriptional profiling, to examine astrocyte responses following focal aquaporin-4 antibody-mediated ablation in the somatosensory cortex of adult mouse brain, a model of astrocytopathy relevant to neuromyelitis o…

An astrocytic cellular model of Lafora disease to study polyglucosan accumulation and inflammation.

Lafora disease (LD) is a devastating form of progressive myoclonus epilepsy characterized by the accumulation of insoluble forms of glycogen [polyglucosan bodies (PGBs)] in the brain and peripheral tissues. It has been proposed that the accumulation of PGBs is pathogenic. Several mouse models of LD have been generated to study the relationship between PGBs and the pathophysiology of LD. However, the use of LD mice is difficult and time consuming; thus, more amenable cellular systems would be desirable. We recently described a cellular model based on the culture of primary postnatal astrocytes …

When lactate speaks: Rewiring astrocyte-neuron metabolism in Alzheimer's disease.

Metabolic dysfunction is a defining but poorly understood feature of Alzheimer's disease. Du et al. show that a brain-penetrant GLP-1 receptor agonist rewires astrocyte-neuron metabolic coupling through lactate-driven histone lactylation, linking astrocytic glycolysis to neuronal lipid homeostasis and positioning metabolite signaling as a therapeutic axis in neurodegeneration.

APP/Aβ Signaling Orchestrates Reactive Astrocyte Networks in Alzheimer's Disease.

Alzheimer's disease (AD) is characterized by amyloid-β (Aβ) accumulation, neurofibrillary pathology, synaptic dysfunction, and chronic neuroinflammation, yet the mechanisms driving early, localized pathology remain elusive. While traditionally viewed through a neuron-centric lens, astrocytes express abundant amyloid precursor protein (APP)-predominantly Kunitz-type protease inhibitor (KPI)-containing isoforms-and possess the complete enzymatic machinery for APP processing and Aβ clearance. Astrocytic APP is a stress-responsive signaling molecule regulated by inflammatory, metabolic, excitotoxi…

Identification of a Small-Molecule Modulator of Astrocyte Reactivity for Optic Nerve Protection.

PURPOSE: Injury of the optic nerve leads to retinal ganglion cells (RGCs) apoptosis and irreversible vision loss, in which reactive astrocytes play a central role. The aim of this study is to modulate pathological reactive astrocytes to reduce the progression of optic nerve degeneration. METHODS: Given the therapeutic potential of small molecules to modulate astrocyte reactivity, we used a drug-screening platform to identify small molecules, and evaluated their capacity to regulate astrocyte phenotypes and preserve RGCs after optic nerve crush (ONC). The primary astrocytes from neonatal C57BL…
