Recent progress in the development of anthrax vaccines.

Bacillus anthracis is the etiological agent of anthrax. Although anthrax is primarily an epizootic disease; humans are at risk for contracting anthrax. The potential use of B. anthracis spores as biowarfare agent has led to immense attention. Prolonged vaccination schedule of current anthrax vaccine and variable protection conferred; often leading to failure of therapy. This highlights the need for alternative anthrax countermeasures. A number of approaches are being investigated to substitute or supplement the existing anthrax vaccines. These relied on expression of Protective antigen (PA), t

CRM197-conjugated serogroup C meningococcal capsular polysaccharide, but not the native polysaccharide, induces persistent antigen-specific memory B cells.

Neisseria meningitidis is one of the leading causes of bacterial meningitis and septicemia in children. Vaccines containing the purified polysaccharide capsule from the organism, a T cell-independent antigen, have been available for decades but do not appear to provide protection in infancy or immunologic memory as measured by antibody responses. By contrast, T cell-dependent serogroup C protein-polysaccharide conjugate vaccines protect against serogroup C meningococcal disease from infancy onward and prime for immunologic memory. We compared the magnitude and kinetics of plasma cell and memor

Immunization with meningococcal outer-membrane protein vesicles containing lipooligosaccharide protects mice against lethal experimental group B Neisseria meningitidis infection and septic shock.

Detergent-treated group B Neisseria meningitidis outer membrane vesicles (D-OMVs) from wild-type M986 and from nonencapsulated mutant M986-non-capsule variant (NCV) were compared as immunogens. Eight weeks after 3 consecutive immunizations with the immunogens, mice were challenged with a lethal dose of purified endotoxin or heat-killed or living N. meningitidis, plus d-galactosamine (400 mg/kg). D-OMVs from M986 induced bactericidal antibodies to both M986 (B : 2a : P1.5,2 : L3,7) and 6275 (B : 2a : P1.2,5 : L3) and protected the animals against both strains, whereas D-OMVs from M986-NCV did n

Impact of host factors and invasive meningococci on bacterial adhesion, proliferation, primary nasal epithelial barrier function, and immune response.

Neisseria meningitidis colonizes the human upper airway mucosa, which can progress into invasive meningococcal disease (IMD) upon breaching the epithelial barrier. Some serogroups and lineages are associated with IMD, whereas others rarely infect patients. Although multiple virulence factors have been described, it is unclear what makes some lineages hyperinvasive. Here, we examined meningococcal infection of air-to-liquid interface (ALI)-differentiated primary epithelial cells to assess host- and strain-dependent differences in colonization that could explain hyperinvasiveness. Nasal epitheli

Klebsiella pneumoniae and Disseminated Intravascular Coagulation: A Comprehensive Review of Pathogenesis, Clinical Manifestations, and Management Strategies.

K. pneumoniae represents a significant opportunistic pathogen capable of causing life-threatening infections, particularly in immunocompromised and critically ill patients. Despite increasing reports of severe coagulopathy in K. pneumoniae sepsis, pathogen-specific mechanisms and tailored management strategies remain insufficiently characterized. This review aims to systematically examine the intricate relationship between K. pneumoniae infections and the development of DIC, a critical complication with high mortality rates, by integrating microbial virulence factors with the host's thrombo-in

Decoding Virulence Mechanisms of Bacillus anthracis Using a Galleria mellonella Infection Model: Differential Host Response Profiles Elicited by AtxA and PlcR.

A thorough understanding of the functions of virulence regulators in Bacillus anthracis evolution and host adaptation, particularly the systematic host responses they trigger, requires an efficient infection model capable of resolving subtle mechanisms. This study constructed a high-resolution host immune response decoder based on Galleria mellonella to analyze the specific response profiles elicited by different virulence regulators in a capsule-deficient background. By integrating transcriptomic, histopathological, and bacterial colonization analyses, the research delineated distinct host im

Characterization and therapeutic efficacy of phage p9676 against epidemic ST11-KL64 Klebsiella pneumoniae: Insights from genomic analysis and in vivo studies.

INTRODUCTION: Klebsiella pneumoniae presents a formidable challenge in clinical settings due to its increasing resistance to last-line antibiotics like carbapenem, tigecycline, and colistin, raising global concerns. An emerging alternative strategy involves harnessing bacteriophages to combat infections caused by multidrug-resistant bacteria. METHODS AND RESULTS: In this study, we isolated the bacteriophage p9676 from hospital sewage and demonstrated its ability to lyse K. pneumoniae strains with ST11-KL64 capsular type. Transmission electron microscopy and genomic analysis identified p9676 a

Investigation into the efficiency of diverse N-linking oligosaccharyltransferases for glycoengineering using a standardised cell-free assay.

The application of bacterial oligosaccharyltransferases (OSTs) such as the Campylobacter jejuni PglB for glycoengineering has attracted considerable interest in glycoengineering and glycoconjugate vaccine development. However, PglB has limited specificity for glycans that can be transferred to candidate proteins, which along with other factors is dependent on the reducing end sugar of glycans. In this study, we developed a cell-free glycosylation assay that offers the speed and simplicity of a 'yes' or 'no' determination. Using the assay, we tested the activity of eleven PglBs from Campylobact

Probiotics Supplements Improve the Sarcopenia-Related Quality of Life in Older Adults with Age-Related Muscle Decline.

A pathological increase in intestinal leak is implicated in age-associated muscle loss, termed sarcopenia, and reduced sarcopenia-related quality-of-life (SarQoL). However, the potential therapies remain elusive. We investigated the effects of probiotic supplementation on sarcopenia and SarQoL in geriatric older adults. We randomized sarcopenic men into placebo (age = 71.4 ± 3.9 years, n = 63) and probiotic (age = 73 ± 4.1 years, n = 60) groups for 16 weeks. The probiotic used was one capsule daily of Vivomix 112 billion for 16 weeks. We measured sarcopenia parameters of handgrip strength (HGS

Adult Onset Acute Disseminated Encephalomyelitis: A Case Report.

Acute disseminated encephalomyelitis (ADEM) is a rare autoimmune demyelinating disorder of the central nervous system that can mimic other neurological diseases, such as multiple sclerosis. ADEM is thought to manifest in the presence of environmental triggers, namely viral or bacterial infections, with multiple simultaneous neurological deficits, frequently accompanied by encephalopathy. Here, we report the case of a 49-year-old female patient who presented in the emergency department with encephalopathy, right-side muscle weakness, dizziness, vertigo, ataxia, and postural imbalance, preceded

Stability and antigenicity of Chlamydia muridarum major outer membrane protein antigen at body temperature.

Chlamydia is an obligate intracellular bacterial pathogen responsible for disease and infertility across multiple species. Currently vaccines are being studied to help reduce the prevalence of this disease. The main advantage of protein subunit vaccines is their high degree of safety although this is traded off with the requirement for multiple booster doses to achieve complete protection. Although in certain populations the booster dose can be difficult and costly to administer, development of delayed vaccine delivery techniques, such as a vaccine capsule, could be the solution to this proble

Get to Know Your Neighbors: Characterization of Close Bacillus anthracis Isolates and Toxin Profile Diversity in the Bacillus cereus Group.

Unexpected atypical isolates of Bacillus cereus s.l. occasionally challenge conventional microbiology and even the most advanced techniques for anthrax detection. For anticipating and gaining trust, 65 isolates of Bacillus cereus s.l. of diverse origin were sequenced and characterized. The BTyper3 tool was used for assignation to genomospecies B. mosaicus (34), B. cereus s.s (29) and B. toyonensis (2), as well as virulence factors and toxin profiling. None of them carried any capsule or anthrax-toxin genes. All harbored the non-hemolytic toxin nheABC and sphygomyelinase spH genes, whereas 41 (

Human neutrophil peptide 1 promotes immune sterilization in vivo by reducing the virulence of multidrug-resistant Klebsiella pneumoniae and increasing the ability of macrophages.

By studying the expression in patients and cell modeling in vitro, antimicrobial peptides for Klebsiella were screened. Killing curve and membrane permeability experiments are used to study the antibacterial effect of antimicrobial peptides in vitro. Cytotoxicity-related indicators including lipopolysaccharide (LPS), capsule polysaccharide (CPS), and outer membrane protein expression were measured. Intranasal inoculation of pneumoconiosis was used to construct a mouse infection model, and the survival rate and cytokine expression level were tested. Human neutrophil peptide 1 (HNP-1) showed a s

The glycointeractome of serogroup B Neisseria meningitidis strain MC58.

Neisseria meningitidis express numerous virulence factors that enable it to interact with diverse microenvironments within the host, during both asymptomatic nasopharyngeal colonization and invasive disease. Many of these interactions involve bacterial or host glycans. In order to characterise the meningococcal glycointeractome, glycan arrays representative of structures found on human cells, were used as a screening tool to investigate host glycans bound by N. meningitidis. Arrays probed with fluorescently labelled wild-type MC58 revealed binding to 223 glycans, including blood group antigens

Streptococcus agalactiae in elephants - A comparative study with isolates from human and zoo animal and livestock origin.

Streptococcus (S.) agalactiae represents a significant pathogen for humans and animals. However, there are only a few elderly reports on S. agalactiae infections in wild and zoo elephants even though this pathogen has been isolated comparatively frequently in these endangered animal species. Consequently, between 2004 and 2015, we collected S. agalactiae isolates from African and Asian elephants (n=23) living in four different zoos in Germany. These isolates were characterised and compared with isolates from other animal species (n=20 isolates) and humans (n=3). We found that the isolates from

Changes in the Population Structure of Invasive Neisseria meningitidis in the United States After Quadrivalent Meningococcal Conjugate Vaccine Licensure.

BACKGROUND: Meningococcal conjugate vaccines against serogroups A, C, W, and Y (MenACWY) are recommended for routine use in adolescents aged 11-18 years. The impact of these vaccines on the meningococcal population structure in the United States have yet to be evaluated. METHODS: Meningococcal isolates recovered during 2006-2010 (ie, after introduction of MenACWY) collected through Active Bacterial Core surveillance (ABCs) were characterized; serogroup distribution and molecular features of these isolates were compared to previously published data on ABCs isolates recovered from 2000 to 2005

A snap-shot of Mannheimia hemolytica A1 gene expression during infection in the bovine host.

It is expected that Mannheimia hemolytica A1 expresses a particular collection of genes during infection in the host. The bacterial gene products are produced in the in vivo environment to facilitate growth and survival. Here, we examined gene expression by M. hemolytica A1 in the bovine host after 6 days of infection. Total RNA from M. hemolytica A1 recovered from pneumonic lungs of two animals was used to produce cDNA to screen a custom M. hemolytica A1 microarray. The expression profile was compared to a RNA sample from an in vitro grown culture. The data showed that 44 genes were different
