Hidradenitis Suppurativa: Comprehensive Review of Predisposing Genetic Mutations and Changes.

Hidradenitis suppurativa (HS) is a chronic inflammatory skin disorder. A genetic component in the pathogenesis is highly likely considering that ~30% to 40% of patients with HS report a family history of the disease. The genetic mutations related to HS that have been reported to date suggest HS can be inherited as a monogenic trait because of a defect in either the Notch signaling pathway or inflammasome function, or as a polygenic disorder resulting from defects in genes regulating epidermal proliferation, ceramide production, or in immune system function. This review provides a summary of ge

Bioactive Sphingolipids of Cordyceps sinensis Modulate Tumor Immunity Through the SphK1-Mediated Cer/S1P Axis.

Cordyceps sinensis is a traditional medicinal fungus known for its immunomodulatory properties. Its bioactive sphingolipids (SPLs) exhibit antitumor potential, though their mechanisms remain poorly understood. This study aimed to identify the active SPLs from C. sinensis and investigate their synergistic effects with αPD-1 (anti-programmed death-1 antibody) therapy. SPLs were isolated from C. sinensis via UPLC-MS/MS-guided purification. The antitumor efficacy and immunomodulatory synergy with αPD-1 were assessed using a Lewis lung carcinoma (LLC) tumor-bearing mouse model and in vitro co-cultu

Role of mitochondria-associated ER membranes in lipid metabolism: implications for renal lipotoxicity.

Dysregulated lipid metabolism is implicated in renal injury associated with diabetic nephropathy, acute kidney injury, chronic kidney diseas … suggesting that MAM dysregulation may contribute to the abnormal metabolism of phospholipids (PLs), ceramides, cholesterol, fatty acids, and triglycerides in renal cells, thereby addressing a gap between previous reviews on renal lipotoxicity and those focusing on MAM-dependent calcium signaling in kidney diseases. Key mechanisms include impaired PL transfe …

Disruption of sphingolipid and glycosphingolipid profiles in human bronchial epithelial cells and extracellular vesicles during gradual benzo[a]pyrene-induced epithelial-to-mesenchymal transition.

In this study, effects of environmental carcinogen benzo[a]pyrene (BaP) on deregulation of sphingolipid (SL) and glycosphingolipid (GSL) metabolism were studied during BaP-induced transformation of normal human bronchial epithelial HBEC-12KT cells. After 2-weeks of exposure, BaP altered their morphology, while it downregulated sphingosine-1-phosphate (S1P) and upregulated sphingosine, gangliosides, GM3 and Lc3 GSLs. A longer, 8-week exposure to BaP, further increased cell migratory capacity, induced epithelial-to-mesenchymal transition (EMT) markers and EMT-related transcriptional regulators (

Miglustat ameliorates isoproterenol-induced cardiac fibrosis via targeting UGCG.

BACKGROUND: Cardiac fibrosis is significant global health problem, which is associated with numerous cardiovascular diseases, and ultimately leads to the progression to heart failure. β-adrenergic receptor (β-AR) overactivation play a role in the development of cardiac fibrosis. Miglustat (Mig) has shown anti-fibrosis effects in multiple fibrotic diseases. However, it is unclear whether and how Mig can ameliorate cardiac fibrosis induced by β-AR overactivation. METHODS: In vivo, mice were injected with isoproterenol (ISO) to induce cardiac fibrosis and treated with Mig. In vitro, primary card

The GLP-1R agonist liraglutide limits hepatic lipotoxicity and inflammatory response in mice fed a methionine-choline deficient diet.

Nonalcoholic fatty liver disease (NAFLD) is the most common hepatic disorder related to type 2 diabetes (T2D). The disease can evolve toward … llobaculum and Turicibacter genera. In conclusion, liraglutide prevents accumulation of C16 and C24-ceramides/sphingomyelins species, inflammation and initiation of fibrosis in MCD-diet-fed mice liver, suggesting beneficial hepatic actions independent of weight loss and global hepatic steatosis.

Altered expression of epidermal lipid bio-synthesis enzymes in atopic dermatitis skin is accompanied by changes in stratum corneum lipid composition.

BACKGROUND: The barrier dysfunction in atopic dermatitis (AD) skin correlates with stratum corneum (SC) lipid abnormalities including reduction of global lipid content, shorter ceramide (CER) as well as free fatty acid (FFA) chain length and altered CER subclass levels. However, the underlying cause of these changes in lipid composition has not been fully investigated. AIM: We investigated whether the expression of CER and FFA biosynthesis enzymes are altered in AD skin compared with control skin and determine whether changes in enzyme expression can be related with changes in lipid compositi

Acid Sphingomyelinase (ASM) is a Negative Regulator of Regulatory T Cell (Treg) Development.

BACKGROUND/AIMS: Regulatory T cell (Treg) is required for the maintenance of tolerance to various tissue antigens and to protect the host from autoimmune disorders. However, Treg may, indirectly, support cancer progression and bacterial infections. Therefore, a balance of Treg function is pivotal for adequate immune responses. Acid sphingomyelinase (ASM) is a rate limiting enzyme involved in the production of ceramide by breaking down sphingomyelin. Previous studies in T-cells have suggested that ASM is involved in CD28 signalling, T lymphocyte granule secretion, degranulation, and vesicle she

Defining a role for acid sphingomyelinase in the p38/interleukin-6 pathway.

Acid sphingomyelinase (ASM) is one of the key enzymes involved in regulating the metabolism of the bioactive sphingolipid ceramide in the sphingolipid salvage pathway, yet defining signaling pathways by which ASM exerts its effects has proven difficult. Previous literature has implicated sphingolipids in the regulation of cytokines such as interleukin-6 (IL-6), but the specific sphingolipid pathways and mechanisms involved in inflammatory signaling need to be further elucidated. In this work, we sought to define the role of ASM in IL-6 production because our previous work showed that a paralle

Development of an isoform-discriminating RT-qPCR assay for transcript-level profiling of human ASAH2 in colorectal cancer cell models.

BACKGROUND: N-Acylsphingosine Amidohydrolase 2 (ASAH2) encodes neutral ceramidase (nCDase), one of several ceramidases involved in sphingolipid metabolism. nCDase hydrolyzes ceramide into sphingosine and contributes to the regulation of the ceramide-sphingosine-1-phosphate signaling axis. Compared with acid and alkaline ceramidases, nCDase remains less well characterized, and the expression and functional relevance of alternatively spliced ASAH2 isoforms are poorly understood. Dysregulated ASAH2 expression has been implicated in colorectal cancer (CRC), suggesting that transcript-level analysi

Integrated assessment of the effects of PFOA exposure on hepatic transcriptome and lipid profiles in mice expressing human PPARα.

Per- and polyfluoroalkyl substances (PFAS) are a family of persistent chemicals that continue to be released pervasively into the environment, leading to widespread human exposure. Emerging epidemiological evidence shows adverse effects on liver lipids; however, past toxicological studies have been limited by a focus on peroxisome proliferator activated receptor α (PPARα) driven effects on triglycerides in rodent systems. Here, we use a more agonostic approach incorporating lipidomics and transcriptomics to test the hypothesis that activation of human PPARα by perfluorooctanoic acid (PFOA), di
