Mobile genetic elements drive a plasmid fusion and deletion lifecycle shaping evolution and antimicrobial resistance.

Plasmids are key drivers of bacterial adaptation, yet the mechanisms that generate their diversity remain poorly understood. Here, we show that mobile genetic elements (MGEs) orchestrate a fusion-deletion life cycle that repeatedly remodels plasmids in Staphylococcus aureus. Large-scale genomic analyses reveal that multireplicon plasmids are widespread and strongly enriched in transposases. Using experimental assays, we demonstrate that rare MGE-mediated fusion events, via homologous recombination or transposition, combine distinct plasmids into single multireplicon elements, expanding gene co

Cinobufagin loaded Prussian blue-like nanoparticles for chemo/gas therapy of multidrug resistant cancer.

Tumor multidrug resistance (MDR) presents a major challenge to the efficacy of cancer chemotherapy. Pyroptosis, a form of regulated cell death distinct from apoptosis and unassociated with drug resistance, may restore tumor cells’ sensitivity to therapeutic drugs. Cinobufagin (CS-1) with efficient pyroptosis inducing capability showed the potential for the treatment of MDR cancer. However, the hydrophobic nature, low bioavailability, and possible toxic side effects under high dosage pose significant limitations on its clinical application. In this research endeavor, we have engineered a unique

Leveraging synthetic genetic array screening to identify therapeutic targets and inhibitors for combatting azole resistance in Candida glabrata.

Fungal infections affect over 6.55 million individuals annually, with Candida glabrata (recently reclassified as Nakaseomyces glabratus) being the second most common cause of invasive candidiasis and candidemia, conditions that carry a high mortality rate of approximately 63.6%. C. glabrata exhibits significant genomic plasticity, enabling rapid adaptation to antifungal treatments. This ability is largely driven by gain-of-function (GOF) mutations in the CgPDR1 gene, which enhance the activity of efflux pumps and confer azole resistance. Over 80 unique CgPDR1 mutations have been identified, le

Evolutionary trajectories determine feasibility of collateral sensitivity-based antibiotic treatment strategies in critical bacterial pathogens.

The rise of antibiotic resistance among pathogenic bacteria necessitates innovative therapeutic strategies, such as collateral sensitivity where resistance to one antibiotic increases susceptibility to another. We explored the clinical relevance of collateral sensitivity through genetic engineering and experimental evolution in the six critical bacterial pathogens Escherichia coli, Salmonella Typhimurium, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Staphylococcus aureus. Our analysis of E. coli showed that clinically relevant resistance mutations did not confer

Emergence of dual drug-resistant strongylids in goats: first phenotypic and genotypic evidence from Ratchaburi Province, central Thailand.

BACKGROUND: This study provides crucial insights into the prevalence and drug resistance patterns of strongylid gastrointestinal nematodes (GINs) in goats in Thailand, highlighting resistance to albendazole and levamisole. Strongylids, particularly Haemonchus sp. and Trichostrongylus sp., pose a significant threat to goat health. With the global rise of anthelmintic resistance, the detection of multidrug resistance in Thailand's goat population is concerning, given the frequent import and export of goats. This resistance challenges effective parasite control strategies. This study aimed to ide

Proteomic Dynamics of Multidrug Resistance Mechanisms in Lucena 1 Cell Line.

The Lucena 1 cell line, derived from the human chronic myeloid leukemia cell line K562 under selective pressure of vincristine supplementation, exhibits multidrug resistance (MDR). This study aims to explore and elucidate the underlying mechanisms driving MDR in the Lucena 1 cell line. A proteomic analysis comparing K562 and Lucena 1 revealed qualitative differences, with a focus on the ATP-dependent efflux pump, Translocase ABCB1, a key contributor to drug resistance. Tubulin analysis identified two unique isoforms, Tubulin beta 8B and alpha chain-like 3, exclusive to Lucena 1, potentially in
