CYP3A induction aggravates endotoxemic liver injury via reactive oxygen species in male rats.

We carried out this experiment to evaluate the relationship between isoforms of cytochrome P450 (P450) and liver injury in lipopolysaccharide (LPS)-induced endotoxemic rats. Male rats were intraperitoneally administered phenobarbital (PB), a P450 inducer, for 3 days, and 1 day later, they were intravenously given LPS. PB significantly increased P450 levels (200% of control levels) and the activities (300-400% of control) of the specific isoforms (CYP), CYP3A2 and CYP2B1, in male rats. Plasma AST and ALT increased slightly more in PB-treated rats than in PB-nontreated (control) rats with LPS tr

Effective inhibition of nitric oxide production by aminoguanidine does not reverse hypotension in endotoxaemic rats.

BACKGROUND: Excess production of nitric oxide (NO) by the inducible NO synthase (iNOS) has been implicated in the pathophysiology of septic shock. Using methaemoglobin (metHb) and the stable NO metabolite nitrate as markers of NO formation, we assessed the effect of iNOS blockade by aminoguanidine (AG) on hypotension and NO formation in endotoxaemic rats. METHODS: In 32 male Wistar rats under chloralose anaesthesia, MetHb (at 15 and 330 min, respectively) and plasma nitrate (at 330 min) were determined. Mean arterial pressure, heart rate and haematocrit were monitored. The LPS group (n=8) rec

Involvement of platelet activating factor in the endotoxin-induced effects on gastrointestinal electrical activity and some haematological parameters in the conscious miniature pig.

In conscious miniature pigs the influence of intravenous dose of lipopolysaccharide (LPS), 10 microg/kg over 10 min, with and without pretreatment with a platelet activating factor (PAF) antagonist, SAH 63-675 10 mg/kg, on gastrointestinal electrical activity, arterial pressure and clinical and haematological parameters was studied. Dose of LPS provoked mild clinical signs and hypotension, which were prevented by PAF antagonism. The LPS induced leukocytosis and increase in mature neutrophils, however, were PAF independent. Pretreatment with the PAF antagonist attenuated the LPS-provoked inhibi

Tissue-specific inflammatory responses to systemic LPS in mice with LysM-lineage Hmgb1 deletion.

The dissociation between plasma HMGB1 and genotype-associated midbrain and CLN responses indicates that total circulating HMGB1 did not fully reflect compartment-specific responses in this model, highlighting the importance of anatomical context in …

Targeting gut dysbiosis: a new frontier in MASLD management.

Globally, Metabolic dysfunction-associated steatotic liver disease (MASLD) is a prominent leading cause of CLD, which is intricately linked … red gut microbiome contributes to MASLD, by specifically emphasizing intestinal barrier disruption, endotoxemia, altered bile acid metabolism, and SCFA imbalance. These dysregulations trigger oxidative stress, leading to systemic inflammation and insulin resistance, propelling simple steatosis toward steatohepatitis and fibrosis at last. Furthermore, we also …

The Gut-Insulin Axis in Polycystic Ovary Syndrome: A Narrative Review of Gut Dysbiosis, Inflammation, and Emerging Therapeutic Strategies.

Polycystic ovary syndrome (PCOS) is a complicated, endocrine-endemic disorder with a prevalence rate of 8-13% in reproductive-age women in the world, which manifests through insulin resistance, hyperandrogenism, and reproductive dysfunction. Despite its high global prevalence, existing therapeutic interventions focus predominantly on symptom management rather than targeting the heterogeneous and multifactorial pathophysiology of the syndrome. Intestinal barrier integrity is destroyed by dysbiosis of the gut microbiota and induces endotoxemia and chronic low-grade inflammation through signallin…

Molecular Mechanisms Associated with Metabolic Dysfunction: Contributions of Nutritional Genomics.

Nutritional genomics has expanded our understanding of how dietary exposures interact with genetic and epigenetic mechanisms involved in metabolic dysfunction. In this context, metabolic dysfunction associated with excessive visceral adiposity arises from a multifaceted interaction between systemic inflammation, insulin resistance, and inter-individual biological susceptibility. Obesity, particularly when driven by diets rich in saturated fatty acids, disrupts intestinal homeostasis, thereby triggering metabolic endotoxemia and contributing to low-grade systemic inflammation and adipose tissue…

Electroacupuncture modulates gut-lung microbiota and lung EMT to attenuate airway remodeling in COPD.

The Role of Cytokines in Vascular Endothelial Glycocalyx Integrity and Impairment Following Open-Heart Surgery.

Open-heart surgery with cardiopulmonary bypass (CPB) is a high-risk procedure with significant morbidity and mortality. CPB, tissue injury, blood loss, endotoxemia and ischemia-reperfusion injury induce a pronounced systemic inflammatory response, leading to endothelial glycocalyx (EG) damage and vascular endothelial dysfunction. Consequently, immune cells, reactive oxygen species, and enzymes gain free access to vascular endothelial cells, resulting in their dysfunction and enhancing inflammation, vascular permeability, and microvascular impairment. EG degradation is most commonly assessed by…

Gastroprotective potential of canagliflozin in indomethacin-induced peptic ulcer: impact on inflammation, pyroptosis, autophagy, and gut integrity.

Additionally, it restored intestinal permeability, which, in turn, aggravated peptic ulcer via endotoxemia. Hence, Cana is a promising gastroprotective agent against peptic ulcers through multiple cascades, evidenced by network pharmacology and experimental validation.

Exercise and Intestinal Barrier Integrity: Molecular Mechanisms and Therapeutic Potential in Gut Health and Disease.

Its functional compromise-termed intestinal hyperpermeability-permits luminal antigens such as lipopolysaccharide to enter the circulation, triggering metabolic endotoxemia and low-grade systemic inflammation that underpin both intestinal and extra-intestinal diseases. This narrative review was based on a comprehensive literature search of PubMed, Web of Science, and Scopus databases up to July 2026, using keywords related to exercise, intestinal barrier, tight junction proteins, inflammation, oxidative st…
