Novel blood biomarkers are crucial for HPV-related cancers to overcome the limitations of imaging and biopsies. This review evaluates the diagnostic performance of circulating cell-free DNA (cfDNA) fragmentomic patterns, distinguishing host-genome integrity from specific viral signatures. Following PRISMA guidelines, we searched PubMed, EMBASE, and Web of Science up to February 2026. Methodological quality was assessed via QUADAS-2, and diagnostic performance was synthesized using a random-effects model. From 489 records, six studies (215 patients, 209 controls) met inclusion criteria. Lacking

Adaptive platform trials can be more efficient than classic trials for developing new treatments. Moving from culture-based to simpler- or faster-to-measure biomarkers as efficacy surrogates may enhance this advantage. We performed a systematic review of treatment efficacy biomarkers in adults with tuberculosis. Platform trials can span different development phases. We grouped biomarkers as: α, bacterial load estimates used in phase 2a trials; β, early and end-of treatment end points, phase 2b-c trials; γ, posttreatment or trial-level estimates, phase 2c-3 trials. We considered as analysis uni

Malaria is a life-threatening and devastating parasitic disease. Our previous work showed that parasite development requires the import of exogenous transfer RNAs (tRNAs), which represents a novel and unique form of host-pathogen interaction, as well as a potentially druggable target. This import is mediated by tRip (tRNA import protein), a membrane protein located on the parasite surface. tRip displays an extracellular domain homologous to the well-characterized OB-fold tRNA-binding domain, a structural motif known to indiscriminately interact with tRNAs. We used MIST (Microarray Identificati

Several pathogenic viruses such as hepatitis B and human immunodeficiency viruses may integrate into the host genome. These virus/host integrations are detectable using paired-end next generation sequencing. However, the low number of expected true virus integrations may be difficult to distinguish from the noise of many false positive candidates. Here, we propose a novel filtering approach that increases specificity without compromising sensitivity for virus/host chimera detection. Our detection pipeline termed Vy-PER (Virus integration detection bY Paired End Reads) outperforms existing simi

Understanding how environmental parameters interact to govern species distributions is a shared goal of ecology and biogeography. Biotic and abiotic conditions can change distributions by affecting the nature of interspecific interactions. Although documented in free-living systems, this context dependency has been neglected in parasite interactions. We investigated the influence of condition-specific competition on the specificity of two species of feather lice (Phthiraptera: Ischnocera) that share a host, the mourning dove (Zenaida macroura). We show that relative humidity restricts the rang

Current treatments for autoimmune disease are hampered by the non-specificity of immunomodulatory interventions, having to accept broad suppression of immunoresponsiveness with potentially serious side effects, such as infection or malignancy. The development of antigen-specific approaches, downregulating pathogenic immune responses while maintaining protective immunity, would be a major step forward. One possible approach involves the targeting of physiological regulatory mechanisms, such as inhibitory CD8 T cells that are now recognized to fine-tune many aspects of immune responses. CD8 T su

The topics in this review are the usefulness of immunodeficient and inbred mice for studies of developmental biology, drug efficacy and host specificity in cysticercosis and echinococcosis. In non-obese diabetic severe combined immunodeficiency (NOD/Shi-scid) mice of both sexes, in vitro hatched oncospheres of all three human taeniid species (Taenia solium, Taenia saginata and Taenia asiatica) developed into cysticerci comparable to or bigger than those developed in their known intermediate host animals, whereas only females were susceptible to these infections in other scid mice of BALB/c, C5…

A 58-year-old woman presented with hemolysis and thrombocytopenia 2 weeks after receiving a kidney graft. Hemolytic uremic syndrome was initially suspected, because in addition to hematological changes the graft function was missing. Unexpectedly, the results of the direct antiglobulin test became positive (4+), which is not normally observed in the hemolytic uremic syndrome. Differentiation of the eluted antibodies revealed anti-rhesus D specificity, which had to be interpreted either as an autoantibody of patient's origin or, hypothetically, as a "graft versus host" antibody of donor origin.

Salmonellae are gastrointestinal pathogens of man and animals. However, strains that are host-specific avian pathogens are often avirulent in mammals, and those which are nonspecific are commensal in poultry. The objective of this study was to determine whether host specificity was exhibited by bacterial abilities to invade epithelial cells or resist leukocyte killing. In this study, leukocytes isolated from humans and chickens were used to kill Salmonella in vitro. Both Salmonella pullorum, an avian-specific serotype, and Salmonella typhimurium, a broad-host-range serotype, were sensitive to …

Parasitic infection often results in alterations to the host's phenotype, and may modify selection pressures for host populations. Elucidating the mechanisms underlying these changes is essential to understand the evolution of host-parasite interactions. A variety of mechanisms may result in changes in the host's behavioural phenotype, ranging from simple by-products of infection to chemicals directly released by the parasite to alter behaviour. Another possibility may involve parasites freely moving to certain sites within tissues, at specific times of the day to induce behavioural changes in

The ability of bacterial pathogens to colonize specific host niches is largely dependent on acquisition of essential metabolites and co-factors for growth and sensing and adapting in response to specific environmental cues. Nutrient availability in host environments is strongly influenced by host physiology and immunity, diet, and competition with other members of the host microbiota. Rapid adaptation to environmental cues and nutrient availability is a hallmark of bacterial fitness and virulence. This adaptability requires complex regulatory networks that tightly link sensing of nutrient avai

Host-parasite interactions occur at a number of stages during the process of infection with Cryptosporidium. Until recently, very little was known about the molecular basis of these interactions or of specific parasite and host molecules involved in them. Within the past decade significant advances have been made in our understanding of Cryptosporidium host-parasite interactions and in identifying molecules involved in them. However, with most interactions the story is far from complete and a number of gaps remain to be filled. This chapter reviews the existing knowledge of the molecular basis

African clawed frogs (Xenopus species) are distributed across sub-Saharan Africa, live in water, and are hosts to diverse parasites whose distributions and host-specificities are incompletely characterized. To better understand this host/parasite biodiversity, we used morphology and Sanger sequencing to characterize Xenopus species and their helminth parasites in several ecological zones of Nigeria. Five Xenopus species were identified in Nigeria (X. fraseri, X. fischbergi, X. poweri, X. tropicalis, and X. calcaratus), and one - Xenopus fraseri - was found to have a wide ecological tolerance i

Fibrinogen is an essential part of the blood coagulation cascade and a major component of the extracellular matrix in mammals. The interface between fibrinogen and bacterial pathogens is an important determinant of the outcome of infection. Here, we demonstrate that a canine host-restricted skin pathogen, Staphylococcus pseudintermedius, produces a cell wall-associated protein (SpsL) that has evolved the capacity for high strength binding to canine fibrinogen, with reduced binding to fibrinogen of other mammalian species including humans. Binding occurs via the surface-expressed N2N3 subdomain

Donor lymphocyte infusions (DLI) can induce durable remissions in multiple myeloma (MM) patients, but this occurs rather infrequently. As the graft-versus-tumor (GvT) effect of DLI depends on the presence of host-dendritic cells (DCs), we tested in a phase I/II trial whether the efficacy of DLI could be improved by simultaneous vaccination with host-DCs. We also analyzed the possibility of further improving the GvT effect by loading the DCs with peptides of mismatched hematopoietic cell-specific minor histocompatibility antigens (mHags). Fifteen MM patients not responding to a first DLI were i

Seasonal infection dynamics characterise many host-parasite interactions, as abiotic conditions drive fundamental biological processes in both hosts and parasites. The extent to which seasonal cycles in parasite abundance are universal, and whether the same underlying mechanisms are responsible for seasonality across different host-parasite associations, have received little attention to date. Here, global patterns of seasonal infection dynamics are tested using a meta-analysis of 693 seasonal contrasts in the local abundance of metazoan parasites in aquatic vertebrates, representing 343 paras

Despite the high disease burden of dengue virus, there is no approved antiviral treatment or broadly applicable vaccine to treat or prevent dengue virus infection. In the last decade, many antiviral compounds have been identified but only few have been further evaluated in pre-clinical or clinical trials. This review will give an overview of the direct-acting and host-directed antivirals identified to date. Furthermore, important parameters for further development that is, drug properties including efficacy, specificity and stability, pre-clinical animal testing, and combinational drug therapy

The brown ear tick Rhipicephalus appendiculatus is the vector for East Coast fever, a disease that seriously limits livestock production in East Africa. The sporozoites of the infectious agent Theileria parva develop in the tick salivary gland. This paper describes the organization of the type III acinus of the gland and establishes unambiguous ultrastructural criteria for identification of the three secretory cell types: the d-cell, e-cell and f-cell. These observations are basic to exploration of possible cell-type specificity of the invading theileria and other aspects of host-parasite rela

Nine Lichtenstein's hartebeest ( Alcelaphus lichtensteini ) were sampled for nasal bots ( Oestrinae ) in a woodland area of central western Zambia. Larvae of the genera Gedoelstia , Oestrus and Kirkioestrus were found in mixed infestations in the nasal cavities and paranasal sinuses. A maximum count of 252 larvae was recorded from one head but no obvious pathogenicity was detectable in this or any other hartebeest. The significance of these findings is discussed in relation to the host specificity of the nasal bots and to gedoelstial myiasis of domestic livestock in the area where they interac…

We studied the specificity of the individual developmental stages of Fasciola hepatica for evoking immune response of the host to reinfection with this parasite, whereby the primary infection was eliminated by a dose of 150 mg/kg diamphenethide administered in various intervals. In rats we observed a state of hypersensitivity demonstrated by retarded migration and growth of the flukes and the reduction in the number of sexually developed parasites. The changes were most marked, if the elimination of the immunizing infection followed 8-10 weeks after primary infection.
