Cyclic AMP compartmentalization drives signal specificity to control vector colonization and mammalian host infection by American trypanosomes.

Cyclic AMP (cAMP) signaling is crucial for environmental sensing and response to stress conditions in trypanosomatids. However, the mechanisms driving the specificity of cAMP signals remain poorly understood in these protozoan parasites. We recently identified two putative cAMP microdomains in Trypanosoma cruzi, the causative agent of Chagas disease. Here, considering the localization of three phosphodiesterases, PDEC at the contractile vacuole complex (CVC), and PDEB1 and PDEB2 along the flagellum, we modulated their expression to functionally characterize the flagellar tip (FT) and the CVC a

Microfluidics-based engineered silver nanoparticles to control growth and biofilm formation in bacterial pathogens causing dental infection.

Dental infections caused by Streptococcus mutans and Enterococcus faecalis are major contributors to dental caries and root canal infections due to their strong biofilm forming ability across all age groups. This present study aimed to develop a microfluidics-assisted green synthesis approach for the fabrication of Clitoria ternatea-derived silver nanoparticles (CtAgNPs) and to evaluate their antibacterial and antibiofilm potential in dental pathogens. The bioactive compounds present in Clitoria ternatea floral extract act as a natural reducing and stabilizing agent within a microfluidics syst

Biological control evaluation of bacteriophage freeze-dried powder against Aeromonas hydrophila infection, based on whey protein/trehalose.

Aeromonas hydrophila, as an important zoonotic conditional pathogen, can cause various diseases such as bacterial septicemia in fish, as well as acute gastroenteritis in humans. Bacteriophages, due to their significant advantage in combating antibiotic-resistant bacteria, have become a research hotspot for novel biological control tools. This study focused on A. hydrophila phage 003s1, which maintained high activity under a wide range of conditions from pH 3 to 11 and at temperatures from 30 to 60°C. After 60 min of ultraviolet irradiation, 63.97% of the original activity remained. To develop

Loss of 7-Dehydrocholesterol Reductase mediated cholesterol biosynthesis activates IRF3 and inhibits control of Mycobacterium marinum infection.

Cholesterol immunometabolism is a critical controller of immunopathology in respiratory infections such as tuberculosis. Smith-Lemli-Opitz syndrome (SLOS) patients are affected by a loss of 7-dehydrocholesterol reductase (DHCR7) function and have elevated 7-dehydrocholesterol (7DHC) and reduced cholesterol. Increased 7DHC has been found to be protective against viral infections in a range of infection models however SLOS patients have a higher susceptibility to respiratory infection. Here we use the zebrafish-Mycobacterium marinum infection model to demonstrate a compromised innate immune resp

Alcohol use disorder and risk of incident COVID-19 infection: A nested case-control study in Taiwan.

BACKGROUND AND AIMS: Alcohol use disorder (AUD) may induce angiotensin-converting enzyme 2 (ACE2) overexpression, potentially increasing vulnerability to severe acute respiratory syndrome coronavirus-2 infection. However, the relationship between AUD and the risk of coronavirus disease 2019 (COVID-19) infection remains unclear. This study aimed to measure the association between AUD and the incidence of COVID-19 infection. DESIGN: We identified 247 000 individuals with AUD between 2001 and 2019 from the Taiwan National Health Insurance Research Database. Another 2 470 000 age- and sex-matched

Burnout in Infection Control Practitioners During Extended Public Health Crisis Events: A Mixed Methods Systematic Review Protocol.

BACKGROUND: Infection prevention and control work environments are complex in nature and have long been associated with extended public health crisis events; major global outbreaks, including epidemics, and pandemics. The role of infection control practitioners shifted exponentially during the COVID-19 pandemic and with this rapid change came anxiety, emotional exhaustion and burnout. Burnout at work generally occurs in response to chronic and prolonged exposure to emotionally challenging events, causing emotional exhaustion, feelings of cynicism, and lack of accomplishment at work. This syste…

CpG oligodeoxynucleotides and pan-serotype inhibitors control neurotropic dengue infection in novel immune competent neonatal mouse model.

Dengue virus (DENV) is a growing global public health threat. The lack of symptomatic immune-competent animal models for dengue has hindered the screening and development of effective therapeutics that can be used to control dengue virus replication and thereby control the progression to severe dengue disease. To address this, we established an infection model in neonatal C57BL/6 mice and showed that a systemic Dengue challenge leads to ataxia, seizures, paralysis, and death within 15 days. The virus was found predominantly in the eye and brain where DENV infects neurons but not astrocytes and

Investigating the association between yellow fever vaccination and symptomatic acute Zika virus infection: a case-control study.

BACKGROUND: During the Zika virus (ZIKV) epidemic in Latin America (2015-2016), a high incidence of microcephaly was observed in places with low yellow fever (YF) vaccination coverage. Cross-reactivity between ZIKV and YF virus was hypothesized as a possible explanation. METHODS: We performed a case-control study to estimate the odds of ZIKV infection according to patients' YF vaccination status. The study considered ZIKV fever as the outcome, and previous vaccination against YF as exposure. We included all confirmed cases of acute ZIKV fever in Campo Grande, Mato Grosso do Sul, from January

Specific CD4+ T cell phenotypes associate with bacterial control in people who 'resist' infection with Mycobacterium tuberculosis.

A subset of individuals exposed to Mycobacterium tuberculosis (Mtb) that we refer to as 'resisters' (RSTR) show evidence of IFN-γ- T cell responses to Mtb-specific antigens despite serially negative results on clinical testing. Here we found that Mtb-specific T cells in RSTR were clonally expanded, confirming the priming of adaptive immune responses following Mtb exposure. RSTR CD4+ T cells showed enrichment of TH17 and regulatory T cell-like functional programs compared to Mtb-specific T cells from individuals with latent Mtb infection. Using public datasets, we showed that these TH17 cell-li

Fetal MAVS and type I IFN signaling pathways control ZIKV infection in the placenta and maternal decidua.

The contribution of placental immune responses to congenital Zika virus (ZIKV) syndrome remains poorly understood. Here, we leveraged a mouse model of ZIKV infection to identify mechanisms of innate immune restriction exclusively in the fetal compartment of the placenta. ZIKV principally infected mononuclear trophoblasts in the junctional zone, which was limited by mitochondrial antiviral-signaling protein (MAVS) and type I interferon (IFN) signaling mechanisms. Single nuclear RNA sequencing revealed MAVS-dependent expression of IFN-stimulated genes (ISGs) in spongiotrophoblasts but not in oth
