More than 3,100 households in 27 selected villages distributed in the main geographic regions of Guinea were surveyed for the presence of Lassa virus-specific IgG antibodies (LVA), using an enzyme-linked immunosorbent assay (ELISA) with Lassa virus nucleocapsid protein expressed in insect cells infected with a recombinant baculovirus as antigen. The highest prevalence of LVA (25-55%) was found among inhabitants of tropical secondary forest (areas near Gueckedou, Yomou, and Lola) and guinea savannah (Faranah and Kindia areas), near the southern frontiers with Sierra Leone and Liberia. This review addresses the ongoing global challenge posed by emerging and evolving viral diseases, underscoring the need for innovative vaccine development strategies. It focuses on the modern approaches to creating vaccines based on recombinant proteins produced in different expression systems, including bacteria, yeast, plants, insects, and mammals. This review analyses the advantages, limitations, and applications of these expression systems for producing vaccine antigens, as well as strategies for designing safer, more effective, and potentially 'universal' antigens. The review discusses th

A serine proteinase-like protein was isolated from the venom of Chinese red scorpion (Buthus martensii Karsch) by combination of gel filtration, ion-exchange and reveres-phase chromatography and named BMK-CBP. The apparent molecular weight of BMK-CBP was identified as 33 kDa by SDS-PAGE under non-reducing condition. The sequence of N-terminal 40 amino acids was obtained by Edman degradation. The sequence shows highest similarity to proteinase from insect source. When tested with commonly used substrates of proteinase, no significant hydrolytic activity was observed for BMK-CBP. The purified BM

The silkworm, Bombyx mori, a key economic insect, has been employed as a model organism to investigate the toxicological effects and underlying mechanisms of zinc iron oxide nanoparticle exposure on its growth and development, midgut tissue structure, and physiological functions. Exposure to zinc iron oxide inhibited larval growth and development, which was accompanied by pathological alterations in the midgut, including a reduction in columnar cells, vacuolation, chromatin condensation, microvilli shedding, and disrupted tight junctions, which directly impaired the intestinal barrier integrit

Blood-sucking insects, such as lice, act as potential vectors of vector-borne pathogens. The proposed study aimed to diagnose molecularly Bartonella sp., an arthropod-borne bacterium in Microthoracius lice collected from alpacas in Peru. Ninety-six lice of the Microthoracius genus were collected from alpaca crias from the Quenamari and Chillihua communities from Cuzco, Peru. All lice were molecularly analyzed by PCR protocols for Bartonella sp. Bartonella genus detection was performed by amplifying the 16S-23S internal transcribed spacer (ITS) region. Bartonella species were identified amplify

Members of the yellow gene family encode conserved Yellow proteins with diverse roles in insect pigmentation, cuticle formation, and reproduction, making them promising targets for RNA interference (RNAi)-mediated pest control strategies. However, little is known about their phylogeny and functions in the brown planthopper (Nilaparvata lugens), a devastating rice pest that serves as a vector for viral rice diseases. Here, we identified and characterized 11 yellow genes in N. lugens, which showed distinct structures and expression profiles. Phylogenetic analysis revealed that N. lugens has reta

The gut microbiome significantly influences insect physiology, immunity, and ecological fitness. However, mass-rearing and sterilization processes in Sterile Insect Technique (SIT) programs disrupt this natural balance. The resulting dysbiosis is a critical factor reducing the performance of sterile males. This study demonstrates that strategically administered probiotic supplementation can restore gut homeostasis and enhance the nutritional status and immunity of Ceratitis capitata sterile males. Our selected three-strain probiotic consortium L.E.K-PC (Lactococcus lactis, Klebsiella oxytoca,

Black soldier fly larvae (BSFL) have emerged as a promising and sustainable source of ingredients for animal feed due to their rapid growth and ability to efficiently bioconvert different organic waste and by-products into high-nutritional-quality insect biomass. Although considerable research has focused on the optimization of larval rearing conditions to improve the chemical composition of the insect biomass, the interplay between diet composition and immune function in BSFL remains poorly understood. Herein, we investigated how two diets differing in their protein-to-carbohydrate (P:C) rati

Zeugodacus tau Walker (Diptera: Tephritidae) is an invasive pest of global significance that inflicts substantial losses to the fruit and vegetable production sectors annually. Despite the availability of the Z. tau genome in public database, functional characterization of key genes remained limited. Vitellogenin (Vg) functions as a key precursor of yolk protein and exerts a pivotal part in the insect reproductive process. Here, we identified 3 Vg genes (Vg1, Vg2, and Vg3) based on genetic data from Z. tau. Analysis of amino acid sequences demonstrated that all ZtVgs contain conserved serine r

As human society continues to grow and evolve, so does the need for effective pest management strategies. Olfactory-mediated control methods, such as attractant and repellent compounds, are a proposed strategy for mitigating the damaging effects of some insect pests, most notably ants, that rely on olfaction for communication. To develop such compounds, it is first important to comprehensively understand the target species' olfactory transcriptome in order to guide future targeted functional characterization of relevant olfactory proteins. Here, we perform bulk RNA-seq analysis of antennae fro

In eukaryotes, a lot of proteins are transported across the endoplasmic reticulum by the heterotrimeric Sec61 channel. And post-translational transport needs another Sec62/Sec63 complex. However, functions of these genes are poorly explored in insects. In this study, we first identified five Sec genes, named Sec61α, Sec61β, Sec61γ, Sec62 and Sec63, in Locusta migratoria. Quantitative reverse-transcription polymerase chain reaction (RT-qPCR) analysis showed that these five genes were expressed in muti-tissues, including wing pad, leg, foregut, midgut, gastric cecum, hindgut, and highly expresse

BACKGROUND: Liriomyza trifolii is an economically significant, invasive pest of horticultural and vegetable crops. The larvae form tunnels in foliage and hasten senescence and death. Outbreaks of L. trifolii often erupt in hot weather and are driven by thermotolerance; furthermore, the poor effectiveness of pesticides has made outbreaks more severe. But it is still unclear whether the development of insecticide tolerance will contribute to thermotolerance in L. trifolii.
RESULTS: To explore potential synergistic relationships between insecticide exposure and thermotolerance in L. trifolii, we

Haemophilus ducreyi was historically known as the causative agent of chancroid, a sexually-transmitted disease causing painful genital ulcers endemic in many low/middle-income nations. In recent years the species has been implicated as the causative agent of nongenital cutaneous ulcers affecting children of the South Pacific Islands and West African countries. Much is still unknown about the mechanism of H. ducreyi transmission in these areas, and recent studies have identified local insect species, namely flies, as potential transmission vectors. H. ducreyi DNA has been detected on the surfac

The genus Flavivirus is a group of arthropod-borne single-stranded RNA viruses, which includes important human and animal pathogens such as Japanese encephalitis virus (JEV), Zika virus (ZIKV), Dengue virus (DENV), yellow fever virus (YFV), West Nile virus (WNV), and Tick-borne encephalitis virus (TBEV). Reverse genetics has been a useful tool for understanding biological properties and the pathogenesis of flaviviruses. However, the conventional construction of full-length infectious clones for flavivirus is time-consuming and difficult due to the toxicity of the flavivirus genome to E. coli.

Phlebotomus argentipes is an established vector for Visceral leishmaniasis prevalent in the Indian subcontinent. Insect Glutathione S-transferases (GST) enzyme plays a pivotal role in the metabolism of xenobiotics and chemical insecticides. We report herein the identification and characterization of a delta class GST from the sandfly, P. argentipes. The resulting clone (rParg-GSTδ) is successfully sequenced, which revealed 76.43% and 66.32% gene identity with GST from Phlebotomus papatasi (Scopoli; Diptera: Psychodidae) and Lutzomiya longipalpis (Lutz and Neiva; Diptera: Psychodidae), respecti

The black fig fly (Silba adipata) is one of the major pests of figs worldwide. This study investigated the effect of pollination on black fig fly infestation and volatile emission during fruit development of facultative parthenocarpic Ficus carica. The results from in-field oviposition preference of black fig fly, olfactory analysis, and fruit volatile profiles indicate that the black fig fly gave a strong preference to unpollinated figs that showed higher emissions of volatile organic compounds. Terpenes are known to be important compounds determining many insect-plant interactions, so we rep

The fall armyworm (Spodoptera frugiperda) is a lepidopteran insect pest that causes huge economic losses. This notorious insect pest has rapidly spread over the world in the past few years. However, the mechanisms of rapid dispersal are not well understood. Here, we report a chromosome-level assembled genome of the fall armyworm, named the ZJ-version, using PacBio and Hi-C technology. The sequenced individual was a female collected from the Zhejiang province of China and had high heterozygosity. The assembled genome size of ZJ-version was 486 Mb, containing 361 contigs with an N50 of 1.13 Mb.

The yellow fever mosquito, Aedes aegypti, vectors human pathogens. Juvenile hormones (JH) control almost every aspect of an insect's life, and JH analogs are currently used to control mosquito larvae. Since RNA interference does not work efficiently during the larval stages of this insect, JH regulation of larval development and mode of action of JH analogs are not well studied. To overcome this limitation, we used a multiple single guide RNA-based CRISPR/Cas9 genome-editing method to knockout the methoprene-tolerant (Met) gene coding for a JH receptor. The Met knockout larvae exhibited a blac

The circadian clock of social insects has become a focal point of interest for research, as social insects show complex forms of timed behavior and organization within their colonies. These behaviors include brood care, nest maintenance, foraging, swarming, defense, and many other tasks, of which several require social synchronization and accurate timing. Ants of the genus Camponotus have been shown to display a variety of daily timed behaviors such as the emergence of males from the nest, foraging, and relocation of brood. Nevertheless, circadian rhythms of isolated individuals have been stud

Blood-feeding arthropods are vectors of infectious diseases such as dengue, Zika, Chagas disease, and malaria [1], and vector control is essential to limiting disease spread. Because these arthropods ingest very large amounts of blood, a protein-rich meal, huge amounts of amino acids are produced during digestion. Previous work on Rhodnius prolixus, a vector of Chagas disease, showed that, among all amino acids, only tyrosine degradation enzymes were overexpressed in the midgut compared to other tissues [2]. Here we demonstrate that tyrosine detoxification is an essential trait in the life his
