Pseudohypoxic stabilization of HIF1α via cyclophilin D suppression promotes melanoma metastasis.

Stabilization of hypoxia-inducible factor 1 alpha (HIF1α), which plays a pivotal role in regulating cellular responses to insufficient oxygen, is implicated in cancer progression, particularly epithelial-mesenchymal transition and metastatic dissemination. Despite its crucial role in tumorigenesis, the precise mechanisms governing HIF1α stabilization under varying tumor microenvironmental conditions are not fully understood. In this study, we show that stabilization of HIF1α in metastasizing melanoma under mild hypoxia is regulated primarily by mitochondrial reactive oxygen species (ROS) rathe

Death-associated protein kinase 1 has a critical role in aberrant tau protein regulation and function.

The presence of tangles composed of phosphorylated tau is one of the neuropathological hallmarks of Alzheimer's disease (AD). Tau, a microtubule (MT)-associated protein, accumulates in AD potentially as a result of posttranslational modifications, such as hyperphosphorylation and conformational changes. However, it has not been fully understood how tau accumulation and phosphorylation are deregulated. In the present study, we identified a novel role of death-associated protein kinase 1 (DAPK1) in the regulation of the tau protein. We found that hippocampal DAPK1 expression is markedly increase

Chlamydia trachomatis-induced alterations in the host cell proteome are required for intracellular growth.

Intracellular pathogens directly alter host cells in order to replicate and survive. While infection-induced changes in host transcription can be readily assessed, posttranscriptional alterations are more difficult to catalog. We applied the global protein stability (GPS) platform, which assesses protein stability based on relative changes in an adjoining fluorescent tag, to identify changes in the host proteome following infection with the obligate intracellular bacteria Chlamydia trachomatis. Our results indicate that C. trachomatis profoundly remodels the host proteome independently of chan

TPI1 Loss Triggers a Metabolite-Driven Mitochondrial Redox Vulnerability via the SARM1-cADPR-Ca2+ Axis.

Cellular senescence is a stable cell cycle arrest program with important therapeutic implications in cancer, yet how metabolic perturbations are translated into redox-dependent senescence remains incompletely understood. Here, we identify triosephosphate isomerase 1 (TPI1) as a critical regulator of senescence in clear cell renal cell carcinoma (ccRCC) through a customized CRISPR-Cas9 metabolic screen. TPI1 depletion induces a robust senescence phenotype characterized by mitochondrial redox imbalance, DNA damage, and stable growth arrest. Mechanistically, loss of TPI1 leads to accumulation of

Mechanism of NanR transcriptional activation of sialic acid metabolism in Streptococcus pneumoniae.

In Streptococcus pneumoniae, the RpiR transcriptional regulator NanR (SpNanR) senses sialic acid in the environment and upregulates transcription of the nan and sia A operons to increase uptake and metabolism of sialic acid. The molecular basis of this activation is unknown. Here, we demonstrate that SpNanR binds N-acetylmannosamine-6-phosphate, a metabolite of sialic acid catabolism. SpNanR exists in a dimer-tetramer equilibrium, and N-acetylmannosamine-6-phosphate binding strongly stabilizes the tetramer. Crystal structures and site-specific substitutions demonstrate that N-acetylmannosamine

GPI promotes immunosuppression in anaplastic thyroid carcinoma via O-GlcNAcylated THBS1-mediated myeloid cell crosstalk.

The aggressive and immunotherapy-resistant characteristics of anaplastic thyroid carcinoma (ATC) are driven by an immunosuppressive tumor microenvironment and intercellular crosstalk; however, its regulatory mechanisms remain poorly understood. Here, we identified Glucose-6-Phosphate Isomerase (GPI) as a pivotal metabolic immune checkpoint that orchestrates myeloid cells-driven immunosuppression in ATC. We demonstrated that GPI enhances the hexosamine biosynthesis pathway to promote O-GlcNAcylation of thrombospondin-1 (THBS1) at serine-1068. This site-specific modification competes with ubiqui

Identification and functional analysis of 2,3-dihydroxybenzoic acid and catechol catabolic pathway genes in Aspergillus niger.

Microorganisms catabolize many plant-derived aromatic compounds through the β-ketoadipate pathway. This pathway has two branches, beginning with protocatechuic acid and catechol, which converge on β-ketoadipate before further catalysis to produce tricarboxylic acid cycle intermediates. The protocatechuic acid branch of the pathway has been characterized in fungi. Here, we characterized the genes of the catechol branch of the β-ketoadipate pathway in the filamentous fungus Aspergillus niger. We predicted the genes involved using comparative transcriptomics and homology to characterized genes. We

Cadmium-induced DAG-PKC/IP3-Ca2+ double signaling pathway and cell injury in gill of freshwater crabs Sinopotamon henanense.

Cadmium (Cd) is an extremely toxic heavy metal and widespread in the environment, which has been demonstrated closely associated with numerous organs damage and even death. To evaluate the signaling pathway and the toxicity induced by Cd2+ in aquatic organisms, freshwater crabs Sinopotamon henanense were exposed to Cd2+ (0, 14.5, 29 and 58 mg/L) for 4 d. The results showed that Cd2+ induced the increase of diacylglycerol (DAG) and inositol-1,4,5-triphosphate (IP3), the rapid change of Ca2+ concentration in the cytoplasm and activation of protein kinase C (PKC) in the gill of crab. Also, Cd2+ r

Cobalt complexes in biology and medicine: enzymatic functions, pharmacological applications, and health challenges.

Cobalt is an essential trace element in biochemistry that plays a crucial role in the structure and function of several important biomolecules. In this review, vitamin B12 is discussed as one of the best-known examples in this area. Various forms of this vitamin, including methylcobalamin and adenosylcobalamin, play a crucial role in metabolic reactions in mammals and prokaryotes. It also discusses cobalt-containing enzymes that are essential for various biological processes. These enzymes are B12-dependent enzymes, which are well studied, and cobalt-containing enzymes, which are less well kno

The R2R3-MYB transcription factor ApMYB9 links UV-B signaling to andrographolide biosynthesis by activating ApIPPI in Andrographis paniculata.

Andrographis paniculata, a medicinal species within the Acanthaceae family, is widely recognized as a "traditional Chinese herbal antibiotic," with andrographolides (ADs) serving as its primary bioactive components. Nevertheless, the molecular regulatory mechanisms governing the biosynthesis of ADs remain poorly understood. In this study, Ultraviolet-B (UV-B) radiation significantly induced the expression of ApIPPI, a gene encoding isopentenyl pyrophosphate isomerase, which is correlated with the accumulation of ADs. Sequence analysis revealed that the ApIPPI promoter contains a variety of MYB

Drynaria roosii-derived exosome-like nanovesicles promote alveolar socket healing via activation of ITPR3-mediated calcium flux.

Alveolar socket healing post-extraction is accompanied by alveolar bone loss, which is negative to subsequent prosthetic rehabilitation. Plant-derived exosome-like nanovesicles are extracellular vehicles with great potential for disease treatment and are advantageous for drug delivery and absorption. Herein, we report the mineralization-promoting effects of Drynaria roosii-derived exosome-like nanovesicles (DRDENs) on osteoblasts and in a mouse alveolar socket healing model. DRDENs were isolated and characterized. Treatment with DRDENs increased the formation of calcium nodules and the express

PDIA6-SCD1 Axis Rewires Lipid Metabolism to Drive Gastric Cancer Progression.

Gastric cancer (GC) remains an aggressive malignancy with limited effective therapeutic options. Integrated single-cell and bulk transcriptomic analyses identify protein disulfide isomerase A6 (PDIA6) as a tumor epithelial-enriched gene associated with advanced tumor‑ node‑metastasis (TNM) stage and unfavorable survival. Multi-omics profiling reveals that PDIA6 drives lipid metabolic reprogramming by sustaining the monounsaturated fatty acid (MUFA)-enriched neutral lipid pools required for lipid droplet homeostasis and redox balance. Mechanistically, PDIA6 directly associates with stearoyl-CoA
