[ARDS and leukotoxin].

It is well known that pulmonary influx of neutrophils is involved in lung injury in patients with adult respiratory distress syndrome (ARDS). Neutrophils are major contributors to the self-defence mechanism, however, adverse effects of neutrophils have also been recognized. Recently, we found that a highly toxic substance, 9, 10-epoxy-12-octadecenoate (leukotoxin) is biosynthesized by human neutrophils. This study was designed to investigate whether or not leukotoxin participates in lung injury in ARDS and coagulation abnormality which is often associated with ARDS. Intravenous injection of le

Effect of High Temperature and Drought on the Fatty Acid Composition of Oil of Different Linseed Varieties and Its Association with Gene Expression.

Flax is an important food and industrial crop. Linolenic acid (LIN) content of linseed oil differs greatly among varieties, ranging from 2% to 70%. In addition to genetic factors, environmental conditions can also significantly impact the fatty acid composition of linseed oil. Our study aimed to compare the effects of different temperature and watering conditions on the fatty acid composition of oil from nine flax varieties with diverse LIN content induced by genotype. Elevated temperature with reduced watering resulted in increased oleic acid (OLE) levels in all varieties. Decreases in linole

Modulation of cellular phospholipid fatty acids and leukotriene B4 synthesis in the human intestinal cell (CaCo-2).

The ability of a human colonocyte epithelial cell line (CaCo-2) to synthesise leukotriene B4 was examined. In addition, the effects of stimulation with calcium ionophore, inhibition by a drug which specifically prevents activation of 5-lipoxygenase, and modification of the fatty acid composition of membrane phospholipids on leukotriene B4 synthesis were assessed. Incubation with calcium ionophore (A23187) resulted in a dose and time dependent increase in leukotriene B4 synthesis. After cell phospholipids had been enriched with oleic acid, linoleic acid, and arachidonic acid, leukotriene B4 syn

Antioxidant and protective effects of extra virgin olive oil incorporated with diallyl sulfide against CCl4-induced acute liver injury in mice.

The present study delineates the effects of incorporation of 1% diallyl sulfide (DAS) into extra virgin olive oil (EVOO) on the physico-chemical characteristics, in vitro antioxidant, and in vivo hepatoprotective properties in CCl4-induced acute liver injury in mice. Results showed that the DAS-rich EVOO exhibited good oxidative stability over one-month storage and preserved its original quality-related parameters including major components (oleic acid, linoleic acid, and palmitic acid), and minor components (tocopherols, chlorophylls and carotenoids, tyrosol, hydroxytyrosol, elenolic acid, ol

Changes in Essential Fatty Acids and Ileal Genes Associated with Metabolizing Enzymes and Fatty Acid Transporters in Rodent Models of Cystic Fibrosis.

Cystic fibrosis (CF), the result of mutations in the CF transmembrane conductance regulator (CFTR), causes essential fatty acid deficiency. The aim of this study was to characterize fatty acid handling in two rodent models of CF; one strain which harbors the loss of phenylalanine at position 508 (Phe508del) in CFTR and the other lacks functional CFTR (510X). Fatty acid concentrations were determined using gas chromatography in serum from Phe508del and 510X rats. The relative expression of genes responsible for fatty acid transport and metabolism were quantified using real-time PCR. Ileal tissu

Transdermal insulin delivery with microwave and fatty acids as permeation enhancers.

Inhaled/oral insulin have been investigated as an alternative to injectable insulin, but are met with unsatisfactory outcomes. Transdermal administration bears several advantages unmet by inhalation/oral delivery, but macromolecular drugs permeation is poor. This study explored microwave to elicit transdermal insulin permeation, and compared against conventional permeation enhancers (fatty acids) in vitro/in vivo. The transdermal insulin permeation was promoted by microwave (2450 MHz/1 mW) > oleic acid (monounsaturated) ~ linoleic acid (double unsaturated bonds). The linolenic acid (triple uns

Free radical generation by early glycation products: a mechanism for accelerated atherogenesis in diabetes.

Non-enzymatic glycation of reactive amino groups in model proteins increased the rate of free radical production at physiologic pH by nearly fifty-fold over non-glycated protein. Superoxide generation was confirmed by electron paramagnetic resonance measurements with the spin-trap phenyl-t-butyl-nitrone. Both Schiff base and Amadori glycation products were found to generate free radicals in a ratio of 1:1.5. Free radicals generated by glycated protein increased peroxidation of membranes of linoleic/arachidonic acid vesicles nearly 2-fold over control, suggesting that the increased glycation of

Changes of Erythrocyte Fatty Acids after Supplementation with Highly Concentrated Docosahexaenoic Acid (DHA) in Pediatric Cystic Fibrosis: A Randomized Double-Blind Controlled Trial.

We characterized the fatty acid profiles in the erythrocyte membrane of pediatric patients with cystic fibrosis (CF) receiving highly concentrated docosahexaenoic acid (DHA) supplementation (Tridocosahexanoin-AOX® 70%) at 50 mg/kg/day (n = 11) or matching placebo (n = 11) for 12 months. The mean age was 11.7 years. The DHA group showed a statistically significant improvement in n-3 polyunsaturated fatty acids (PUFAs), which was observed as early as 6 months and further increased at 12 months. Among the n-3 PUFAs, there was a significant increase in DHA and eicosapentaenoic acid (EPA). Addition

Mechanisms underlying the response of different resistant quinoa cultivars to Spodoptera exigua feeding.

BACKGROUND: Spodoptera exigua is the main pest that harms quinoa. Its larvae feed on leaves, flowers, and grains, seriously affecting the quality and yield of quinoa. However, faced with the feeding of S. exigua, some quinoa cultivars have the characteristics of avoiding or reducing damage and having compensatory ability after being harmed. This study aims to dissect the defensive responses of quinoa cultivars with differing resistance levels to feeding by S. exigua. METHODS: We compared defensive metabolites and transcriptomes among high-resistant (HR), medium-resistant (MR) and highly-susce

13/16-Specific Allene Oxide Synthase BnAOS (CYP74A) of Rapeseed (Brassica napus L.): Identification and Biochemical Properties.

Lipoxygenase cascade is the source of key plant bioregulators, oxylipins, which are oxidized derivatives of polyunsaturated fatty acids that play a crucial role in growth and development, as well as stress response. The key enzymes of the lipoxygenase cascade are allene oxide synthases (AOS) and hydroperoxide lyases, which are present in all known angiosperms. Although these enzymes play a central role in defining the pattern of the final metabolites of the cascade, significant gaps still exist in understanding their catalytic properties and positional specificity. This paper describes the all

Integrative analysis of placental metabolic reprogramming and microbiome alterations in gestational diabetes mellitus (GDM).

Gestational diabetes mellitus (GDM) significantly impacts placental metabolism and microbial composition, leading to transformations at the maternal–fetal interface. In response to hyperglycemia, the placenta undergoes extensive metabolic reprogramming, including disturbances in lipid metabolism, reduced efficiency in fatty acid oxidation, altered amino acid transport, and adaptations in glycolysis. These metabolic changes are associated with inflammatory cytokine activity, mitochondrial stress, and endoplasmic reticulum dysfunction, collectively driving placental lipotoxicity and disruptions

Barn swallows and purple martins convert precursors to long-chain polyunsaturated fatty acids: implications for conservation of riparian- vs inland-nesting habitats.

For aerial insectivorous birds, whose populations have declined significantly in North America, long-chain polyunsaturated fatty acids (LC-PUFA) that are more abundant in aquatic-emergent insects than terrestrial insects, are important for the development, somatic growth, and health of young birds. Some bird species, however, can convert shorter chain PUFA that occur abundantly in terrestrial insects into LC-PUFA. Our study aimed to evaluate the ability of two aerial insectivore species to synthesize their own LC-PUFA. We hypothesized that terrestrially associated aerial insectivores rely on h
