Somatic Accumulation of GluA1-AMPA Receptors Leads to Selective Cognitive Impairments in Mice.

The GluA1 subunit of the L-α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor (AMPAR) plays a crucial, but highly selective, role in cognitive function. Here we analyzed AMPAR expression, AMPAR distribution and spatial learning in mice (Gria1R/R ), expressing the "trafficking compromised" GluA1(Q600R) point mutation. Our analysis revealed somatic accumulation and reduction of GluA1(Q600R) and GluA2, but only slightly reduced CA1 synaptic localization in hippocampi of adult Gria1R/R mice. These immunohistological changes were accompanied by a strong reduction of somatic AMPAR current

Management of adipsia by a behavioural modification technique.

Adipsia combined with diabetes insipidus after hypothalamic damage may produce major difficulties in clinical management. If there is an associated memory impairment it may be impossible to teach self-regulation of fluid balance, necessitating long-term hospital supervision. The successful use of a behaviour modification technique to achieve independent drinking and allow discharge from hospital into the community is described in a patient with adipsia, diabetes insipidus and memory impairment resulting from the removal of a craniopharyngioma.

[Transient epileptic amnesia].

Transient epileptic amnesia (TEA) is a sub-type of mesial temporal lobe epilepsy, with amnesic seizures. TEA is characterized by recurrent episodes of amnesia. Diagnostic criteria are available for TEA, and these memory disturbances should not be misdiagnosed with transient global amnesia. The neuropsychological evaluation is normal, however, autobiographical memory impairment is present in 70% of the cases and accelerated long term forgetting in 44%. When a patient complains of memory disturbances, especially autobiographical memory, TEA must be considered especially if there was an amnesic e

The magnitude of the antibody and memory B cell responses during priming with a protein-polysaccharide conjugate vaccine in human infants is associated with the persistence of antibody and the intensity of booster response.

Rapid waning of anti-polysaccharide bactericidal Ab and vaccine effectiveness is observed following infant immunization with the serogroup C meningococcal (MenC) glycoconjugate vaccine. This is despite the demonstrable presence of immunological memory. Persistence of functional Ab, therefore, appears to be the key determinant of MenC conjugate vaccine effectiveness. Ab persistence is thought to depend in the short term on the survival of plasma cells generated during priming and in the longer term on the production of new Ab secreting cells from memory B cells. In this study, we found a strong

Effects of antipsychotic D2 antagonists on long-term potentiation in animals and implications for human studies.

In people with schizophrenia, cognitive abilities - including memory - are strongly associated with functional outcome. Long-term potentiation (LTP) is a form of neuroplasticity that is believed to be the physiological basis for memory. It has been postulated that antipsychotic medication can impair long-term potentiation and cognition by altering dopaminergic transmission. Thus, a systematic review was performed in order to assess the relationship between antipsychotics and D2 antagonists on long-term potentiation. The majority of studies on LTP and antipsychotics have found that acute admini

Autobiographical memory and electroconvulsive therapy: do not throw out the baby.

Retrograde amnesia for autobiographical information is the most critical adverse effect of electroconvulsive therapy (ECT). Much, if not most, modern research demonstrating long-term autobiographical amnesia after ECT has used either the Columbia University Autobiographical Memory Interview (CUAMI) or the short form of this scale (CUAMI-SF). Semkovska and McLoughlin claimed that studies using these instruments should be dismissed and the findings ignored owing to a lack of normative data, as well as concerns about the reliability and validity of these instruments. In this commentary, the devel

Immune reconstitution in children following chemotherapy for haematological malignancies: a long-term follow-up.

Modern intensive chemotherapy for childhood haematological malignancies has led to high cure rates, but has detrimental effects on the immune system. There is little knowledge concerning long-term recovery of the adaptive immune system. Here we studied the long-term reconstitution of the adaptive immune system in 31 children treated for haematological malignancies between July 2000 and October 2006. We performed detailed phenotypical and functional analyses of the various B and T cell subpopulations until 5 years after chemotherapy. We show that recovery of newly-developed transitional B cells

Long-term health outcomes of adolescent mTBI: a scoping review.

INTRODUCTION: Adolescent mild traumatic brain injury (mTBI) carries potential long-term effects. This scoping review aimed to map and explore recent literature on the long-term outcomes of mTBI in adolescents aged 13-18, defining long-term as a minimum of 12 months post-injury. METHODS: Following the JBI framework and PRISMA-ScR guidelines, we reviewed 37 English articles published in peer-reviewed academic journals between 2015 and 2024. Narrative synthesis and concept analysis were used to identify themes and patterns related to long-term outcomes. FINDINGS: The findings reveal a complex p

Preventing academic difficulties in preterm children: a randomised controlled trial of an adaptive working memory training intervention - IMPRINT study.

BACKGROUND: Very preterm children exhibit difficulties in working memory, a key cognitive ability vital to learning information and the development of academic skills. Previous research suggests that an adaptive working memory training intervention (Cogmed) may improve working memory and other cognitive and behavioural domains, although further randomised controlled trials employing long-term outcomes are needed, and with populations at risk for working memory deficits, such as children born preterm. In a cohort of extremely preterm (<28 weeks' gestation)/extremely low birthweight (<1000 g) 7-y

What a nostril knows: olfactory nerve-evoked AMPA responses increase while NMDA responses decrease at 24-h post-training for lateralized odor preference memory in neonate rat.

Increased AMPA signaling is proposed to mediate long-term memory. Rat neonates acquire odor preferences in a single olfactory bulb if one nostril is occluded at training. Memory testing here confirmed that only trained bulbs support increased odor preference at 24 h. Olfactory nerve field potentials were tested at 24 h in slices from trained and untrained bulbs. A larger AMPA component and a smaller NMDA component characterized responses in the bulb receiving odor preference training. Field potential changes were not seen in a bulbar region separate from the lateral odor-encoding area. These r

Mild cognitive impairment does entail retrograde amnesia for public events.

In this study memory for public events was evaluated in 15 amnesic mild cognitive impairment (aMCI) patients, whose clinical diagnosis was refined through a stringent selection procedure. A total of 9 patients were longitudinally reassessed over an 18-month period. About half of the participants were impaired at baseline and nearly 80% at the end of the 18-month follow-up. Moreover, retrograde memory declined significantly over time. Evidence of a pathological Ribot-type temporal gradient was found in about half of the aMCI patients. This is the first report of a remote memory deficit in aMCI.

Traumatic amnesia, repression, and hippocampus injury due to emotional stress, corticosteroids and enkephalins.

The role of stress, arousal, emotional trauma, and corticosteroid and enkephalin secretion on memory and the hippocampus, and the development of traumatic amnesia and repressed memory syndrome are detailed. Animal and human studies are reviewed. Trauma-induced memory deficits appear to be secondary to abnormal neocortical and hippocampal arousal, and corticosteroid and enkephalin secretion which can induce atrophy or seizures within the hippocampus, suppress hippocampal theta activity and long term potentiation, as well as injure hippocampal pyramidal cells. Predisposing factors include indivi

The effects of synthetic glucocorticoid treatment for inflammatory disease on brain structure, function, and dementia outcomes: A systematic review.

Many regions of the brain have a high density of glucocorticoid receptors, and the prolonged elevation of endogenous glucocorticoids may cause neurotoxicity and increase risk for cognitive decline and dementia. However, despite synthetic glucocorticoids being the first line of treatment for many inflammatory diseases, few studies have addressed whether therapeutic glucocorticoids may have similar undesirable effects on the brain. Thus, our systematic review investigated the impact of long-term glucocorticoid usage on adult brain structure, cognitive function, and dementia risk. We identified 1

Lipopolysaccharide preconditioning reduces neuroinflammation against hypoxic ischemia and provides long-term outcome of neuroprotection in neonatal rat.

Hypoxic ischemia (HI) in newborns causes long-term neurologic abnormalities. Systemic lipopolysaccharide (LPS) is neuroprotective in neonatal rats when injected 24 h before HI. However, the effect on HI-induced neuroinflammation and the long-term outcome of LPS preconditioning in neonatal rats have not been examined. In a rat-pup HI model, compared with normal saline (NS), 0.3 mg/kg of LPS injected 24 h before HI greatly increased microglial cell and macrophage activation and up-regulated TNF-alpha and inducible NOS expression 12-h postinjection and resulted in high mortality during HI. 'Paradoxical' alpha synchronization in a memory task.

The results of a specially designed memory search paradigm which maximizes episodic short-term memory (STM) and minimizes semantic long-term memory (LTM) demands show that the upper alpha band synchronizes selectively in those conditions and time intervals where episodic STM demands are maximal. This finding of a selective alpha synchronization occurring only in the upper alpha band and during highest task demands is surprising because it is well known that usually alpha desynchronizes during mental activity. Because experiments from our laboratory indicate that desynchronization in the upper

Chronic hypertension and social isolation disrupt the blood-brain barrier and memory in a sex-dependent manner.

Chronic hypertension in mid-life is a leading cause of vascular cognitive impairment (VCI), the second leading cause of age-related cognitive decline. Further, social isolation is another risk factor for dementia, and disproportionately impacts middle-aged adults. However, mechanisms by which social isolation may compound chronic hypertension to damage the cerebrovasculature and contribute to VCI remain unclear. The current study used male and female adult Wistar rats and spontaneously hypertensive rats (SHR) to investigate how social isolation during chronic hypertension disrupted blood-brain

Origin and differentiation of human memory CD8 T cells after vaccination.

The differentiation of human memory CD8 T cells is not well understood. Here we address this issue using the live yellow fever virus (YFV) vaccine, which induces long-term immunity in humans. We used in vivo deuterium labelling to mark CD8 T cells that proliferated in response to the virus and then assessed cellular turnover and longevity by quantifying deuterium dilution kinetics in YFV-specific CD8 T cells using mass spectrometry. This longitudinal analysis showed that the memory pool originates from CD8 T cells that divided extensively during the first two weeks after infection and is maint

Accelerated long-term forgetting in patients with acquired brain injury.

OBJECTIVE: Recent research suggests that patients with neurological disorders without overt seizures may also experience accelerated long-term forgetting (ALF). This term describes unimpaired learning and memory performance after standard retention intervals, but an excessive rate of forgetting over delays of days or weeks. The objective of this retrospective study was to investigate ALF in patients with an acquired brain injury (ABI) and to associate memory performance with executive functions. METHODS: Verbal memory performance (short-term recall, 30-min recall, 1-week recall) was assessed

Non-invasive transcutaneous vagus nerve stimulation during memory retrieval enhances recollection of emotionally salient memories.

Remembering emotionally significant events is critical for the organisms' behavior and survival. While the mechanisms underlying the formation of such memories are well understood and closely linked to the vagus nerve and the brain's arousal system, less is known about its contribution to memory retrieval. The current study tested whether non-invasive transcutaneous auricular vagus nerve stimulation (taVNS) applied during a long-term recognition memory task influenced retrieval of unpleasant and neutral scenes that had been incidentally encoded one week earlier. Results showed that taVNS, comp

Valproate improves memory deficits in an Alzheimer's disease mouse model: investigation of possible mechanisms of action.

Alzheimer's disease (AD) is a very common progressive neurodegenerative disorder affecting the learning and memory abilities in the brain. Key findings from recent studies of epigenetic mechanisms of memory suggest chromatin remodeling disorders via histone hypoacetylation of the lysine residue contribute to the cognitive impairment in AD. Therefore, the deinhibition of histone acetylation induced by histone deacetylases (HDACs) inhibitors contributes to recovery of learning and memory. We show here that the antiepileptic drug sodium valproate (VPA) potently enhanced long-term recognition memo
