Inhibition of fucosylation of cell wall components by 2-fluoro 2-deoxy-L-fucose induces defects in root cell elongation.

Screening of commercially available fluoro monosaccharides as putative growth inhibitors in Arabidopsis thaliana revealed that 2-fluoro 2-l-fucose (2F-Fuc) reduces root growth at micromolar concentrations. The inability of 2F-Fuc to affect an Atfkgp mutant that is defective in the fucose salvage pathway indicates that 2F-Fuc must be converted to its cognate GDP nucleotide sugar in order to inhibit root growth. Chemical analysis of cell wall polysaccharides and glycoproteins demonstrated that fucosylation of xyloglucans and of N-linked glycans is fully inhibited by 10 μm 2F-Fuc in Arabidopsis s…

Glycosylated antitumor ether lipids: activity and mechanism of action.

Glycosylated antitumor ether lipids (GAELs) are distinguished from the alkyllysophospholipids or alkylphosphocholines classes of antitumor ether lipids (AEL) by the presence of a sugar moiety. Non-phosphorus GAELs, the subject of this review, have a sugar moiety in place of the phosphobase found in alkyllysophospholipids. Analogues of non-phosphorus GAELs with glucose, maltose, arabinose, or disaccharide moieties have been synthesized. Non-phosphorus GAELs with monosaccharides have cytotoxic and antiproliferative effects against cancer cells derived from a wide range of tissues, including drug…

Flower opening and closure: a review.

Flower opening and closure are traits of a reproductive syndrome, as it allows pollen removal and/or pollination. Various types of opening can be distinguished such as nocturnal and diurnal and single or repetitive. Opening is generally due to cell expansion. Osmotic solute levels increase by the conversion of polysaccharides (starch or fructan) to monosaccharides, and/or the uptake of sugars from the apoplast. Repeated opening and closure movements are often brought about by differential elongation. In tulip petals, for example, the upper and lower sides of the mesophyll exhibit a 10 degrees …

Ultrasound-assisted deep eutectic solvent extraction of Moringa oleifera leaf polysaccharides: process optimization, purification, physicochemical characterization, and bioactivities.

Ultrasound-assisted deep eutectic solvent (DES) extraction was developed for the efficient recovery of polysaccharides from Moringa oleifera leaves. Among seven DES systems, choline chloride-urea exhibited the highest extraction efficiency. Response surface methodology further optimized the extraction conditions, achieving a maximum polysaccharide yield of 17.1 %. The crude polysaccharide fraction (MOLP) was subsequently purified by DEAE-52 ion-exchange chromatography and Sephadex G-100 gel filtration, yielding two major purified fractions, MOLPs0 and MOLPs0.5. Structural characterization show

Isolated lignin loading elicits hydrolysis-centered cross-scale responses during anaerobic cellulose digestion with a mesophilic inoculum at two temperatures.

Lignin constrains cellulose hydrolysis during anaerobic digestion of lignocellulosic biomass, but its effects on downstream conversion kinetics and microbial community states remain unclear. Here, microcrystalline cellulose was co-digested with isolated lignin using a mesophilic inoculum at 35 and 55℃ conditions to characterize loading-dependent changes in methane conversion, kinetic parameters derived from Anaerobic Digestion Model No. 1 (ADM1), and microbial community states. Increasing lignin loading progressively delayed methane conversion, prolonged the times required to reach 50% and 90%

Structure-activity relationship of uronic acid oligosaccharides as potential anti-Alzheimer's disease agents.

Alzheimer's disease (AD) is the most common neurodegenerative disorder and the primary cause of dementia. Although natural polysaccharides exhibit significant therapeutic potential against AD due to their multi-target activities, the structure-activity relationship (SAR) of uronic acid oligosaccharides-particularly the influence of specific glycosidic linkages and monosaccharide compositions-remains insufficiently characterized. To address this, we utilized a highly selective TEMPO oxidation system to prepare four structurally distinct uronic acid oligosaccharides (α1,4-G, β1,4-G, β1,3-G, and

Structure elucidation of duckweed polysaccharide from Spirodela oligorrhiza and its anti-inflammatory activity.

Duckweed polysaccharides constitute an underexplored yet functionally important class of biomolecules within these rapidly proliferating aquatic plants. Although abundant in cell walls and implicated in diverse bioactivities, their structural elucidation beyond pectic fractions has remained scarce. In this study, the first comprehensive structural analysis of a purified polysaccharide (WHL-N1) derived from Spirodela oligorrhiza duckweed is reported. Monosaccharide composition analysis demonstrated galactose (46.15 mol%) and arabinose (39.44 mol%) as the predominant constituents. Structural inv

Glucose and fructose differently mediate alcohol cocktail drinking in female and male rats: interaction of glucose and alcohol on post-ingestive behavior.

Sweetened alcoholic beverages are thought to contribute to developing Alcohol Use Disorder by increasing palatability. One monosaccharide, glucose, readily enters the brain more than fructose and directly impacts the activity of central neurons. The objective of this study is to determine the impact of glucose versus fructose on alcohol drinking patterns in female and male rats. Rats drank alcohol cocktails (1.25%-10%) containing either glucose or fructose (10%) in 4-hour sessions. We sought to parse orosensory effects from post-ingestive central effects by analyzing drinking microstructure. W

Genomic and functional characterization of sugar transporters reveals potential roles in sugar accumulation in a modern sugarcane cultivar.

Sugarcane (Saccharum spp.) is a globally important sugar crop whose productivity depends on efficient sugar transport from source to sink organs. However, systematic identification and functional characterization of sugar transporters (STs) in sugarcane cultivars remain limited. Here, we identified 190 non-redundant ST genes in sugarcane cultivar Guitang 42 (GT42) and phylogenetically classified them into nine groups within the Monosaccharide Transporter (MST), Sucrose Transporter (SUT), and Sugars Will Eventually be Exported Transporters (SWEET) families. Fabrication and application of a Dendrobium officinale polysaccharide-based active film incorporated with carbon dots and anthocyanins for shrimp preservation.

This study developed an intelligent active packaging film based on Dendrobium officinale polysaccharides (DOP), incorporating copper-doped carbon dots (Cu-CD) derived from the residue of DOP extraction and anthocyanins (AMA) extracted from Aronia melanocarpa. To clarify the fundamental physicochemical properties of DOP and AMA, both were systematically characterized. For DOP, the molecular weight, monosaccharide composition, total sugar content, and in vitro antioxidant activity were determined. For AMA, the total anthocyanin content and antioxidant activity were measured. On this basis, the e

Structure characterization and multi-target inhibitory activities of fucosylated chondroitin sulfate from sea cucumber Thelenota anax.

A fucosylated chondroitin sulfate (FCS) was extracted from the body wall of Thelenota anax (FCSTx). Physicochemical characterization confirmed its typical FCS structural features, and monosaccharide composition analysis revealed the presence of Gal. Combined NMR spectroscopic analysis of native FCSTx and fifteen oligosaccharides prepared by partial acid hydrolysis and β-elimination depolymerization comprehensively established that FCSTx possesses a chondroitin sulfate E backbone with branches composed of 60% Fuc2S4S, 8% Fuc4S, and 32% D-Gal4S(6S)-α1,2-L-Fuc3S, all attached at the O-3 of GlcA.
