Mechanisms of key gut microbial metabolites: SCFAs, BAs, and TDMs.

Gut microbial metabolites, particularly short-chain fatty acids (SCFAs), bile acid (BAs) derivatives, and tryptophan-derived metabolites (TDMs), act as signaling molecules, receptor ligands, and epigenetic modulators in regulating host metabolic and immune homeostasis. They exert local effects in the gut and reach distant tissues via hepatic, lymphatic, neural, and immune-hormonal routes, thereby regulating immunity, metabolism, and neural function. Disruptions in these pathways are associated with inflammatory diseases, neurodegeneration, and cancer. This review discusses the importance of SC

Glutamatergic Projection from the Ventrolateral Periaqueductal Gray to the Parafascicular Nucleus of the Thalamus Contributes to the Modulation of Neuropathic Pain in Mice.

Chronic pain is frequently accompanied by increased pain sensitivity and affective disturbances. The ventrolateral periaqueductal gray (vlPAG) and the parafascicular nucleus of the thalamus (PF) are key nodes in pain modulation. However, the functional role of the vlPAG-PF neural pathway remains poorly understood. In this study, we employed an integrative approach combining neuroanatomical tracing and chemogenetic techniques to investigate the role of the vlPAGvGluT2-PF neural pathway in pain modulation. Our results demonstrate that vGluT2-positive neurons in the vlPAG project to the PF. Chemo

Key circuits through which sleep affects memory and their role in Alzheimer's disease.

Sleep is a critical physiological process for maintaining the cognitive function of brain, particularly about memory consolidation. This review systematically elaborates on the multiscale regulatory mechanisms of the sleep-wake cycle, with a focus on the neural circuits involved in sleep-dependent memory consolidation, such as the hippocampus-prefrontal-thalamus network, characteristic neural oscillations, and microglia-mediated synaptic changes. Building upon these foundations, the article delves into Alzheimer's disease (AD), providing an in-depth analysis of the complex bidirectional relati

Exploring the effects of mindfulness meditation on resting-state functional connectivity: an overview.

Mindfulness meditation has received growing interest in research over the past decades. Mindfulness meditation training (MMT) can be employed as a mental practice to improve cognitive skills such as attention and emotion regulation and may promote well-being. Neuroimaging studies have emerged to understand the effects of MMT on brain functioning. However, no review exists on the effects of MMT on resting-state brain functional connectivity (rsFC) in healthy meditation-naïve adults specifically. We, therefore, aimed to provide an overview of studies that investigated the effects of MMT on rsFC

Genoarchitecture and input-output organization of the mouse basal ganglia and thalamic parafascicular nucleus.

The basal ganglia comprise interconnected subcortical nuclei essential for motor control, learning, emotion and cognition, yet how cell types relate to their circuit organization remains elusive. Here we show that spatial patterns of transcriptomic cell types in the basal ganglia and thalamic parafascicular nucleus relate to module-organized cortical inputs in the mouse. By co-registering genoarchitectural and connectivity datasets to the common coordinate framework, we delineate distinct three-dimensional subdivisions of these nuclei. Analyses of anterograde and retrograde viral tracing data

The ventral/anterior hippocampus as a central hub for stress-related dopaminergic circuit dysfunction in psychiatric disorders.

The hippocampus has long been recognized for its central role in memory and navigation. However more recent studies have shown that this structure is also involved in the pathophysiology of major psychiatric disorders. Specifically, the ventral/anterior portion of the hippocampus is a mediator of stress-induced circuit dysfunction in neuropathology. The hippocampus is comprised of a number of neurotransmissions, especially inhibitory, not unlike the neocortex, but is also vulnerable to disruption particularly in the juvenile stage. The disruption occurs via alterations in activity but also in

Resting-state motor network connectivity as a biomarker of Levodopa response in Parkinson's disease.

BACKGROUND: Parkinson's disease (PD), the second most common neurodegenerative disorder, affects over 10 million people and leads to progressive motor and non-motor decline. Levodopa (L-DOPA) remains the primary treatment, yet long-term use causes motor fluctuations and dyskinesias, and tools to optimize therapy remain limited. OBJECTIVES: This review systematically evaluates studies assessing how L-DOPA affects functional connectivity within the motor network, to inform more precise and personalized treatment strategies. METHODS: A systematic search of PubMed and Web of Science was performe

The proto-cortical amygdala: a perspective on the functional and evolutionary relationship between the basolateral amygdala complex and neocortex in mammals.

The basolateral amygdala complex (BNC) plays an important role in learning, memory, motivation, and social behaviour, with its dysfunction implicated in a wide variety of psychiatric disorders. Despite being one of the most intensely researched areas of the brain, much is still unknown about its microanatomical structure and function, relationships and evolutionary origins. In this review, we offer a multidisciplinary approach through the lens of comparative anatomy and evolutionary developmental studies to provide a better understanding of the anatomical and functional relationship between th

A multisensory hub for resilience: LC-vLGN/IGL circuits in stress coping.

Enriched sensory experiences promote stress resilience. However, the underlying neural mechanisms remain elusive. In this issue of Neuron, Huang et al.1 identify a glutamatergic locus coeruleus-ventral lateral geniculate nucleus/intergeniculate leaflet pathway that integrates multisensory salience to promote adaptive stress coping.

Heterogeneous brain alterations in treatment-resistant depression converge on common control networks.

Treatment-resistant depression (TRD) imposes a substantial clinical and societal burden. However, it remains unclear whether neuroimaging abnormalities associated with TRD converge onto specific large-scale brain networks. We systematically reviewed neuroimaging articles comparing patients with TRD to both healthy controls (HCs) and patients with non-treatment-resistant depression (non-TRD), encompassing functional and structural imaging modalities. Using a validated coordinate network mapping (CNM) approach and a normative human brain connectome, we derived TRD-associated networks. Overlap an

Innervation Pattern of Inhibitory Projection Neurons in the Bird Sound Localization Circuit.

To navigate its environment, an animal extracts salient information from sounds using temporal and intensity cues. In birds, the nucleus laminaris (NL) detects the submillisecond differences in the arrival time of sound to the two ears, the interaural time differences (ITDs), to localize sounds. This ability is facilitated by inhibitory long-range projection neurons from the ipsilateral superior olivary nucleus (SON) that enable NL neurons to remain sensitive to ITDs across a large range of sound intensities. It is well known that the excitatory inputs to NL, from nucleus magnocellularis (NM),

Ancestral organisation and asymmetric circuit architecture of the habenula revealed in a basal ray-finned fish.

The habenula is a conserved dorsal diencephalic hub that integrates information related to motivational state, behavioural flexibility, and cognitive control. In many vertebrates, it displays pronounced left-right asymmetry, yet how structural lateralisation is reflected in circuit architecture remains poorly understood, partly because phylogenetically informative lineages remain undersampled. Here, we characterise habenular connectivity in the Senegal bichir (Polypterus senegalus), a member of an early-diverging ray-finned fish lineage with marked habenular asymmetry. Using neuronal tracing,

Post adversity changes in nigro-striatal dopamine: A mechanism for anxiety induced exacerbated innate repetitive behaviors.

Anxiety exacerbates symptoms in various psychiatric disorders. In conditions such as obsessive-compulsive disorder (OCD) or Tourette syndrome, anxiety intensifies stereotypic and repetitive behaviors. Rodent self-grooming, a structured, repetitive innate behavior, serves as an effective rodent platform for studying these behaviors in neuropsychiatric research. Anxiety is also linked to altered functioning of the dopamine (DA) system, particularly within the substantia-nigra pars compacta (SNc), the main DA source to the dorsal striatum through the nigro-striatal pathway. Striatal modulation by

BACKGROUND: Abnormalities in large-scale brain functional networks are implicated in multiple sclerosis (MS) and linked to clinical impairments, but reported findings remain heterogeneous due to methodological and sample variations. METHODS: Based on 25 resting-state functional magnetic resonance (rsfMRI) imaging studies (1,524 MS patients and 886 healthy controls (HCs)), in which the independent component analysis (ICA) method was used to evaluate the resting-state functional connectivity (rsFC) changes in default mode (DMN), sensorimotor (SMN), salience (SN), and visual (VN) networks, a met

Insight into the application of optogenetics for regulating motor-related circuits in Parkinson's disease and novel therapeutic target exploration.

Parkinson's disease (PD) is one of the most pervasive neurodegenerative diseases. However, the etiology and pathogenesis of PD have not been fully elucidated, the underlying neural circuits or neuron types have not been fully identified, and the roles that different neuronal subtypes in distinct deep brain nuclei play in the pathophysiological process of PD need to be explored. Optogenetics is a technique that can realize the exact temporal and spatial specific control of a defined neuron subpopulation, which may contribute to the comprehension of pathophysiological mechanism of PD. For this s

Amygdala-Dependent Signaling Pathways and Regulating Circuits in Fear Memory.

Fear memory, a profound emotional memory formed in response to threatening events, is vital to the survival and adaptation of individuals. The amygdala, a central hub in the formation, storage, and expression of fear memory, extensively connects with other brain regions, particularly the hippocampus and hypothalamus, allowing it to rapidly integrate sensory information and generate emotional responses. Past research on the regulatory circuits of fear memory has focused on the amygdala, prefrontal cortex, and hippocampus, emphasizing their interactive roles in the formation, consolidation, and

Gateway to the study of the amygdala and emotion.

The study of the amygdala and its role in the processing of emotions has become a common focus in neuroscience. The modern expansion of research in this area is partly due to the discovery of a subcortical pathway for the transmission of emotional information and the experimental paradigm that was developed to study it. Groundbreaking experiments during the 90s utilized anatomical, neurophysiological, and behavioral lesion studies in a rodent animal model to uncover the neural circuitry of a simple emotional memory. These studies demonstrated the essential role of a specific monosynaptic pathw

Projections from the supragenual nucleus to the lateral mammillary and dorsal tegmental nuclei.

An organism's sense of direction depends on vestibular input to thalamic and forebrain structures. The supragenual nucleus (SGN) sits at a prime location in the network to convey vestibular signals forward to update the representation of head direction (HD) with ongoing head movement. The SGN receives anatomical projections from the medial vestibular nuclei and nucleus prepositus hypoglossi and prior reports have suggested that the SGN sends projections to both the dorsal tegmental nucleus (DTN) and lateral mammillary nucleus (LMN), two midbrain nuclei crucial in generating a representation of

Aberrant dynamic functional connectivity of amygdala subregions in adolescents with depression.

OBJECTIVE: Research has indicated that aberrant amygdala connectivity is a crucial neurobiological underpinning of juvenile depression. This study aimed to further identify resting-state dynamic functional connectivity (dFC) changes between the lateral amygdala (LA), medial amygdala (MeA), and whole-brain voxels, as well as their correlations with clinical symptoms in adolescents with depression. MATERIALS AND METHODS: The bilateral LA and MeA were selected as regions of interest, and the sliding window method was applied to investigate dFC changes in 47 adolescents with depression, compared

Wnt signaling pathway and retinoic acid signaling pathway involved in delamination and migration of chicken trunk NCCs and contributing to HVP phenotype.

Hyperpigmentation of the visceral peritoneum (HVP) is a hereditary trait that significantly affects the carcass quality in bearded chickens, yet its molecular mechanisms remain unclear. This study utilized data-independent acquisition proteomics to analyze the protein expression profiles of black peritoneum (B), faded peritoneum (F), and normal peritoneum (N) in bearded chickens at 40 and 120 d of age. Combined with histopathological and functional enrichment analyses, we revealed the regulatory network underlying HVP formation. Results indicated that the melanin content was significantly elev
