ATH434 Rescues Pre-motor Hyposmia in a Mouse Model of Parkinsonism.

Hyposmia is a prevalent prodromal feature of Parkinson's disease (PD), though the neuropathology that underlies this symptom is poorly understood. Unlike the substantia nigra, the status of metal homeostasis in the olfactory bulbs has not been characterized in PD. Given the increasing interest in metal modulation as a therapeutic avenue in PD, we sought to investigate bulbar metals and the effect of AT434 (formerly PBT434) an orally bioavailable, small molecule modulator of metal homeostasis on hyposmia in a mouse model of parkinsonism (the tau knockout (tau-/-) mouse). 5.5 (pre-hyposmia) and …

A subset of peripherin positive olfactory axons delineates the luteinizing hormone releasing hormone neuronal migratory pathway in developing mouse.

Luteinizing hormone releasing hormone (LHRH) neurons in the CNS are derived from cells of the olfactory placode and thereafter migrate from … herin and N-CAM immunoreactive. Although the majority of these axonal tracks entered the developing olfactory bulbs, a small population of peripherin positive but N-CAM negative axons turned caudally into the developing forebrain. LHRH cells were consistently juxtaposed to these axons. We propose that this peripherin positive/N-CAM negative fiber track is the an …

Substrate-bound carbohydrates stimulate signal transduction and neurite outgrowth in an olfactory neuron cell line.

Subpopulations of olfactory receptor neurons, which are dispersed throughout the olfactory neuroepithelium, express specific cell surface carbohydrates and project to discrete regions of the olfactory bulb. Cell surface carbohydrates such as N-acetyl-lactosamine have been postulated to mediate sorting and selective fasciculation of discrete axon subpopulations during development of the olfactory pathway. Substrate-bound N-acetyl-lactosamine promotes neurite outgrowth by both clonal olfactory receptor neuron cell lines and olfactory receptor neurons in vitro, indicating that cell surface carboh…

Olfaction: New Understandings, Diagnostic Applications.

Estimates indicate that 14 million Americans have olfactory dysfunction. As with other senses, such as sight and hearing, olfaction frequently declines with age. Impaired olfaction can be a warning sign of Parkinson's disease, sometimes occurring before motor symptoms develop. It's also an initial symptom of Alzheimer's dementia (AD); the amyloid plaques and tangles characterizing AD invade the olfactory bulb and hippocampus early in its course, hampering odor identification. Olfactory dysfunction is associated with some serious problems, including inability to smell warning odors (fire, gas) …

Primary olfactory terminations in the forebrain of amphibia: a comparative study with soybean agglutinin.

Ambystoma and Xenopus were used to investigate olfactory terminations in the forebrain by exploiting the selective binding of soybean agglut … irst time, which terminate in a discrete bilateral region of the forebrain localized below the main olfactory bulb. The presence of three different sets of primary olfactory neurons, the experimental potentials and the possible functional significance of SBA binding sites are discussed.

In the central olfactory pathway, a decrease in the turnover of interneurons in the olfactory bulb (OB) and reduced activity in the olfactory cortex under olfactory stimulation is observed. Recently, the association between olfactory impairment and neurodegenerative diseases, such as Alzheimer's disease (AD) and Parkinson's disease (PD), has gain …

Expression of non-N-methyl-D-aspartate glutamate receptor subunits in the olfactory epithelium.

The channel properties of the multimeric ionotropic glutamate receptors can be regulated by their subunit composition. The relationship betw … he axons of these neurons, however, terminate and release glutamate in the glomerular region of the olfactory bulb, and may contain presynaptic glutamate receptors. By reverse transcriptase-polymerase chain reaction amplification and RNase protection assays, we showed that a subset of non-N-methyl-D-aspartate receptor subunits is expressed in the olfactory epith …

Wide-ranging migration and destination of early olfactory placode-derived neurons in chick embryos.

Cell migration from the olfactory placode (OP) is a well-known phenomenon wherein various cell types, such as gonadotropin-releasing hormone … EL at HH23 and HH27. At HH36, labeled cells were observed in the rostral-lateral TEL, including the olfactory bulb (OB) region. GFP-labeled and calretinin-positive neurons were detected in the OB, suggesting that early OP-derived neurons enter the forebrain and function as interneurons in the OB.

Recurrent circuitry is required to stabilize piriform cortex odor representations across brain states.

Pattern completion, or the ability to retrieve stable neural activity patterns from noisy or partial cues, is a fundamental feature of memory. Theoretical studies indicate that recurrently connected auto-associative or discrete attractor networks can perform this process. Although pattern completion and attractor dynamics have been observed in various recurrent neural circuits, the role recurrent circuitry plays in implementing these processes remains unclear. In recordings from head-fixed mice, we found that odor responses in olfactory bulb degrade under ketamine/xylazine anesthesia while res…

The terminal nerve and its relation with extrabulbar "olfactory" projections: lessons from lampreys and lungfishes.

The definition of the terminal nerve has led to considerable confusion and controversy. This review analyzes the current state of knowledge as well as diverging opinions about the existence, components, and definition of terminal nerves or their components, with emphasis on lampreys and lungfishes. I will argue that the historical terminology regarding this cranial nerve embraces a definition of a terminal nerve that is compatible with its existence in all vertebrate species. This review further summarizes classical and more recent anatomical, developmental, neurochemical, and molecular eviden

The role of olfaction in oil preference in the chicken.

The role of olfaction on the preference of diets containing 20% medium-chain (MCT) or long-chain triacylglycerol (LCT) was investigated in the chicken. Olfactory bulbectomized, sham-operated (Sham) or intact (Intact) birds were offered a choice between LCT or MCT diet and food intake was measured over a short time period. Intact and Sham groups showed a significant preference for LCT over MCT diet, but olfactory-bulbectomized chickens lost the preference for LCT over MCT. The bilateral cutting of the olfactory nerves confirmed the results taken in olfactory bulbectomy. It is concluded that olf…

Early Odorant Exposure Increases the Number of Mitral and Tufted Cells Associated with a Single Glomerulus.

UNLABELLED: The highly specific organization of the olfactory bulb (OB) is well known, but the impact of early odorant experience on its circuit structure is unclear. Olfactory sensory neurons (OSNs) project axons from the olfactory epithelium to the OB, where they form spherical neuropil structures called glomeruli. These glomeruli and the postsynaptic targets of OSNs, including mitral and tufted cells (M/TCs) and juxtaglomerular cells, form glomerular modules, which represent the basic odor-coding units of the OB. Here, we labeled M/TCs within a single glomerular module of the mouse OB and s…

Origin and endpoint of the olfactory nerve fibers: as described by Santiago Ramón y Cajal.

In the late Nineteenth Century, Santiago Ramón y Cajal was able to reproduce an exceptional illustration of the Olfactory Nerve pathway and … al and central nervous system and was able to assert that these fibers were not continuous from the olfactory bulb to the bipolar cells that relinquish into the olfactory epithelium, but that the olfactory system was made up of various cell types each having distinct morphologies and functions. This may very well be the first definitive description of the olfact …

Role of neuropeptide Y in the regulation of gonadotropin releasing hormone system in the forebrain of Clarias batrachus (Linn.): immunocytochemistry and high performance liquid chromatography-electrospray ionization-mass spectrometric analysis.

Although the importance of neuropeptide Y (NPY) in the regulation of gonadotropin releasing hormone (GnRH) and reproduction has been highlig … on, a dramatic increase was observed in the GnRH immunoreactivity in the ORNs, in the fibers of the olfactory bulb (163%) and medial olfactory tract (351%). High performance liquid chromatography-electrospray ionization-mass spectrometric analysis confirmed the immunocytochemical data. Significant rise in the salmon GnRH (sGnRH)-like peptide content was observed …
