Interaction of GABAA and GABAB antagonists after status epilepticus in immature rats.

Among neurotransmitter systems affected by status epilepticus (SE) in adult rats are both GABAergic systems. To analyze possible changes of GABAA and GABAB systems in developing rats lithium-pilocarpine SE was induced at postnatal day 12 (P12). Seizures were elicited by a GABAA antagonist pentylenetetrazol (PTZ) 3, 6, 9, and 13 days after SE (i.e., in P15, P18, P21, and P25 rats), and their possible potentiation by a GABAB receptor antagonist CGP46381 was studied. Pilocarpine was replaced by saline in control animals (lithium-paraldehyde [LiPAR]). Pentylenetetrazol in a dose of 50 mg/kg s.c. e

Identification of a putative methyltransferase gene of Babesia bigemina as a novel molecular biomarker uniquely expressed in parasite tick stages.

BACKGROUND: Bovine babesiosis is caused by apicomplexan pathogens of the genus Babesia such as B. bigemina and B. bovis. These tick-borne pathogens have a complex life-cycle involving asexual multiplication in vertebrate hosts and sexual reproduction in invertebrate vectors. In the tick midgut, extracellular Babesia parasites transform into gametes that fuse to form zygotes. Understanding the mechanisms that underlie formation of extracellular Babesia tick stages is an important step towards developing control strategies for preventing tick infection and subsequent parasite transmission. Molecular tools that block maturation of the nuclear lamin A and decelerate cancer cell migration.

Lamin A contributes to the structure of nuclei in all mammalian cells and plays an important role in cell division and migration. Mature lamin A is derived from a farnesylated precursor protein, known as prelamin A, which undergoes post-translational cleavage catalyzed by the zinc metalloprotease STE24 (ZPMSTE24). Accumulation of farnesylated prelamin A in the nuclear envelope compromises cell division, impairs mitosis and induces an increased expression of inflammatory gene products. ZMPSTE24 has been proposed as a potential therapeutic target in oncology. A library of peptidomimetic compound

Differential alkylation-based redox proteomics--Lessons learnt.

Cysteine is one of the most reactive amino acids. This is due to the electronegativity of sulphur atom in the side chain of thiolate group. It results in cysteine being present in several distinct redox forms inside the cell. Amongst these, reversible oxidations, S-nitrosylation and S-sulfenylation are crucial mediators of intracellular redox signalling, with known associations to health and disease. Study of their functionalities has intensified thanks to the development of various analytical strategies, with particular contribution from differential alkylation-based proteomics methods. Prese

Microbial metabolism of reduced phosphorus compounds.

The field of bacterial phosphorus (P) metabolism has undergone a significant transformation in the past decade owing to the elucidation of widespread and diverse pathways for the metabolism of reduced P compounds. The characterization of these pathways dramatically changes the current and narrow view of P metabolism and our understanding of the forms in which P is produced and available in the environment. In this review, recent investigations into the biochemical pathways and molecular genetics of reduced P metabolism in bacteria are discussed. Particular attention is paid to recently elucida

Engineering InAs(x)P(1-x)/InP/ZnSe III-V alloyed core/shell quantum dots for the near-infrared.

Quantum dots with a core/shell/shell structure consisting of an alloyed core of InAs(x)P(1-x), an intermediate shell of InP, and an outer shell of ZnSe were developed. The InAs(x)P(1-x) alloyed core has a graded internal composition with increasing arsenic content from the center to the edge of the dots. This compositional gradient results from two apparent effects: (1) the faster reaction kinetics of the phosphorus precursor compared to the arsenic precursor, and (2) a post-growth arsenic-phosphorus exchange reaction that increases the arsenic content. The cores have a zinc blend structure fo

Epidemiology of hydrogen phosphide exposures in humans reported to the poison center in Mainz, Germany, 1983-2003.

BACKGROUND: Poisonings with rodenticides containing hydrogen phosphide-releasing compounds may lead to deleterious organ dysfunction and death. Since data of hydrogen phosphide poisonings is limited to case reports/series, this study was intended to elucidate hydrogen phosphide poisonings based on a 20-year data collection. METHODS: Explorative data analysis of the Poison Center Mainz database looking for route of exposure, symptoms, and severity using the Poisoning Severity Score. RESULTS: From 1983-2003, 188 hydrogen phosphide poisonings were reported. Sixty-five percent of these were unin

Medicinal chemistry and molecular pharmacology of GABA(C) receptors.

GABA(C) receptors belong to the nicotinicoid superfamily of ionotropic receptors that include nicotinic acetylcholine receptors, bicuculline-sensitive GABA(A) receptors, strychnine-sensitive glycine receptors and 5HT3 serotonin receptors. The GABA(C) receptor concept arose from medicinal chemical studies of a conformationally restricted analog of GABA. Receptors matching the predicted properties of GABA(C) receptors were cloned from the retina. Post cloning studies revealed the unique physiology and pharmacology of these relatively simple homomeric receptors. Three subtypes of GABA(C) receptor

Mutagenicity testing of a rodenticide, zinctox (zinc phosphide) in a mouse in vivo system.

The genotoxic potential of zinctox (zinc phosphide) was evaluated in mice in vivo employing different bioassays: bone marrow chromosome aberration, micronucleus and sperm shape abnormality. A significant increase of chromosome aberrations was induced by three doses. Interestingly, the highest percentage was induced by the middle dose. In a route-responsive study the highest percentage of chromosome aberration was induced by the oral route. A dose-dependent increase in the incidence of micronuclei was noticed in the bone marrow cells. In the sperm shape abnormality test a high percentage of abn

A unified platform for the rapid assembly of glutarimides for Cereblon E3 ligase modulatory drugs.

Glutarimide-containing Cereblon (CRBN) ligands are critical motifs for PROTACs, molecular glue degraders and next-generation Cereblon E3 ligase modulatory drugs (CELMoDs), which represent promising therapeutic modalities in targeted protein degradation. However, the multistep synthetic routes required to access glutarimide scaffolds continue to present formidable challenges for medicinal chemists, limiting rapid structure-activity relationship (SAR) exploration and late-stage diversification. To streamline access to these privileged motifs, modular and efficient methodologies are still highly

Lanthanide Complexes Supported by Tripodal Chalcogenophosphinic Amide Ligands.

While transition metal complexes containing tripodal phosphinic amide ligands such as [N{CH2CH2NP(O)Ph2}3]3- ([poat]3-) are well documented, analogous complexes with lanthanide elements have not been explored. Herein, we report on the synthesis, structure, and reactivity of lanthanide complexes supported by the tripodal chalcogenophosphinic amide ligands [N{CH2CH2NP(E)R2}3]3- (R = Ph, E = O; R = iPr, E = S or Se). Treatment of LaCl3 and CeBr3 in tetrahydrofuran with K3poat afforded dinuclear [M2(poat)2] that reacted with trimethylphosphine oxide to yield the adducts [M2(OPMe3)2(poat)2] (M = La

Profiling Selectivity for the Shigella Virulence Factor OspF.

Phosphothreonine lyases are a small family of bacterial virulence factors that are secreted into host cells during infection by some Gram-negative pathogens. Phosphothreonine lyases catalyze an irreversible phosphate β-elimination that converts phosphothreonine (pThr) to dehydrobutyrine (Dhb) on host proteins. Here, we explore the substrate profile for OspF, a phosphothreonine lyase secreted during Shigella flexneri infection, which has been previously reported to modify mitogen-activated protein kinases (MAPKs) to silence the host immune response. In this work, we use a combination of in vitr

Cu-Catalyzed Regioselective and Enantioselective Allylation of Ketones with Allenylsilanes: Mechanistic Insights and Selectivity Origins via Density Functional Theory Analysis.

Comprehensive density functional theory investigations were conducted to elucidate the mechanism and ligand-regulated regio- and enantioselectivity in the Cu-catalyzed asymmetric allylation of ketones with allenylsilanes. Reaction pathways involving two distinct ligands were explored, specifically Josiphos-type bidentate phosphine ligand L1 (reaction 1) and P-stereogenic ligand BenzP* L2 (reaction 2). For both catalytic systems, the catalytic cycle comprises catalyst activation, C-C bond formation accompanied by [Cu] migration, turnover-limiting σ-bond metathesis, and desilication. The 1,3-cop

Cell Surface Thiol Engineering Mechanoregulates Myogenic Differentiation via the FAK-PI3K-AKT Axis.

Skeletal muscle regeneration is essential in conditions like muscle injury and muscular dystrophy, primarily relying on the activation and differentiation of myogenic progenitors. This study explores the role of redox-modulated surface thiol modification in myogenesis, using tris(2-carboxyethyl)phosphine hydrochloride (TCEP) to reduce disulfide bonds and generate free thiol groups on the cell surface. The findings show that TCEP treatment significantly increases cell surface thiols, activating the focal adhesion kinase (FAK)-phosphoinositide 3-kinase (PI3K)-protein kinase B (AKT) signaling pat

Catalytic Conversion of Carbon Dioxide to Long Chain Esters via Formate Hydroesterification.

CO2-based hydroesterification is an attractive route to produce value added ester compounds, which could replace widely used CO-based hydroesterification processes if sufficient catalytic technologies are developed. One path to CO2-based hydroesterification could proceed through an organoformate intermediate, which is then used in olefin hydroesterification to generate the desirable esters. This route opens the possibility of creating a net CO2-based hydroesterification process using tandem catalytic systems for CO2 hydrogenation to organoformate paired with formate-olefin hydroesterification.
