The Iron Deficiency-Regulated Small Protein Effector FEP3/IRON MAN1 Modulates Interaction of BRUTUS-LIKE1 With bHLH Subgroup IVc and POPEYE Transcription Factors.

In light of climate change and human population growth one of the most challenging tasks is to generate plants that are Fe-efficient, resilient to low Fe supply and Fe-biofortified. For such endeavors, it is crucial to understand the regulation of Fe acquisition and allocation in plants. One open question is how identified Fe-regulatory proteins comprising positive and negative regulators act together to steer Fe homeostasis. bHLH transcription factors (TFs) belonging to the subgroups IVb and IVc can initiate a bHLH cascade controlling the -Fe response in roots. In Arabidopsis thaliana, the -F

Multi-omics reveals DEAD-box RNA helicase 20 as key protein enhancing wilt resistance in transgenic chickpea.

AtNPR1 expression strengthens Fusarium wilt resistance in chickpea by activating SAR. Multi-omics analyses suggest CaDEAD-box20 as a candidate gene contributing to resistance through possible interaction with AtNPR1. Traditional breeding for broad-spectrum disease resistance in crops is often slow and resource-intensive, whereas genetic engineering provides a more precise and efficient alternative. To enhance resistance in chickpea (Cicer arietinum) against Fusarium oxysporum f. sp. ciceris, the causal agent of Fusarium wilt, we introduced the Arabidopsis NPR1 (AtNPR1) gene to activate systemi

The Plant Noncanonical Antiviral Resistance Protein JAX1 Inhibits Potexviral Replication by Targeting the Viral RNA-Dependent RNA Polymerase.

Understanding the innate immune mechanisms of plants is necessary for the breeding of disease-resistant lines. Previously, we identified the antiviral resistance gene JAX1 from Arabidopsis thaliana, which inhibits infection by potexviruses. JAX1 encodes a unique jacalin-type lectin protein. In this study, we analyzed the molecular mechanisms of JAX1-mediated resistance. JAX1 restricted the multiplication of a potexviral replicon lacking movement-associated proteins, suggesting inhibition of viral replication. Therefore, we developed an in vitro potato virus X (PVX) translation/replication syst

Adaptive evolution of polyploid crops.

Crop evolution represents a fundamental biological process through which plants respond to selection in different environments. This encompasses mechanisms operating at multiple scales of biological organization, including genetic and epigenetic regulation and higher-order interactions among molecular complexes. This Review synthesizes how polyploidy shapes crop evolution by generating duplicated genes, driving genome reorganization, altering dosage relationships and promoting regulatory divergence, which together influence crop metabolism, physiology, development and environmental responses.

Respiratory burst oxidase homolog 1 plays a critical role in spermidine-induced thermotolerance by enhancing CO2 assimilation and cellular redox homeostasis in tomato.

Heat stress increasingly threatens global agriculture by disrupting photosynthesis and cellular redox homeostasis in plants. Spermidine (Spd), a stress-responsive polyamine, enhances thermotolerance by bolstering antioxidant capacity and stabilizing membranes; however, the molecular mechanisms underlying these effects remain incompletely characterized. Here, we demonstrate that Respiratory burst oxidase homolog 1 (RBOH1) is essential for Spd-induced thermotolerance in tomato (Solanum lycopersicum L.). Deficiency of RBOH1 compromised the protective effects of Spd, resulting in exacerbated membr

Exploring Aroma and Flavor Diversity in Cannabis sativa L.-A Review of Scientific Developments and Applications.

Cannabis sativa L. exhibits a complex sensory profile governed by a diverse range of volatile and non-volatile compounds. Volatile constituents-such as terpenes, aldehydes, ketones, esters, and sulfur-containing compounds-together with non-volatile taste-active molecules including flavonoids and phenolic compounds, underlie its distinctive aroma and flavor. This review examines how genetic diversity, cultivation practices, and post-harvest processing modulate the synthesis, accumulation, and chemical transformation of these metabolites in the cannabis flower. It discusses recent advancements i

Enhanced Photosynthetic Capacity, Osmotic Adjustment and Antioxidant Defenses Contribute to Improve Tolerance to Moderate Water Deficit and Recovery of Triploid Citrus Genotypes.

Currently, drought stress is a major issue for crop productivity, and future climate models predict a rise in frequency and severity of drought episodes. Polyploidy has been related to improved tolerance of plants to environmental stresses. In Citrus breeding programs, the use of triploidy is an effective way to produce seedless fruits, one of the greatest consumer expectations. The current study used physiological and biochemical parameters to assess the differential responses to moderate water deficit of 3x genotypes compared to 2x genotypes belonging to the same hybridization. Both parents,

Biological nitrogen fixation in the context of global change.

The intensive application of fertilizers during agricultural practices has led to an unprecedented perturbation of the nitrogen cycle, illustrated by the growing accumulation of nitrates in soils and waters and of nitrogen oxides in the atmosphere. Besides increasing use efficiency of current N fertilizers, priority should be given to value the process of biological nitrogen fixation (BNF) through more sustainable technologies that reduce the undesired effects of chemical N fertilization of agricultural crops. Wider legume adoption, supported by coordinated legume breeding and inoculation prog

Clones of pea aphid, Acyrthosiphon pisum (Hemiptera: aphididae) distinguished using genetic markers, differ in their damaging effect on a resistant alfalfa cultivar.

CUF 101, a resistant cultivar of alfalfa, was exposed to 15 clones of Acyrthosiphon pisum Harris collected from alfalfa fields in three regions of France (east, south, central west) to determine whether the level of resistance varied across the different clones. The survival of alfalfa seedlings infested at the cotyledon stage was assessed using a standardized method. Although no difference in seedling mortality was detected between clones grouped by region, there was a significant variation among the 15 pea aphid clones. In particular, two clones of southern origin were more aggressive. In ad

UAV-based phenotyping outperforms visual canopy wilting for evaluating soybean drought tolerance and yield retention under rainfed conditions.

Drought is the major abiotic stress limiting soybean growth and yield, yet accurately identifying genotypes that sustain yield under rainfed conditions remains a major bottleneck in soybean breeding. Canopy wilting scores are widely used as a proxy for evaluating plant responses to drought stress. However, most assessments rely on leaf-level visual observations that are inherently subjective and typically based on single time-point scores, providing only a snapshot of stress expression and failing to capture their relationship with yield retention under rainfed conditions. To address these lim

Assessment of cold tolerance in early field emergence in sweet, fiber, and grain sorghums [Sorghum bicolor (L.) Moench].

INTRODUCTION: Constraints to the adoption of early sowings in biomass sorghum were found in its sensitivity to cold stress during germination. METHODS: A preliminary test was conducted in the laboratory on seven sorghums among sweet, fiber, and grain types, to assess their cold tolerance during germination. Sorghums were germinated at 8 °C and 25 °C (control). At the end of the test, final germination percentage (FG), mean germination time (MGT), and germination index (GI) were calculated. Thereafter, a first-year (2009) field study was conducted under the semi-arid climate of Southern Italy,

Molecular mechanisms and regulatory networks of salt stress tolerance in plants.

Soil salinity is a major threat to crop productivity, sustainable agriculture, and global food security, with more than 833 million hectares of land affected worldwide. Salt stress restricts plant growth through osmotic stress, ion toxicity, oxidative damage, membrane disruption, reduced photosynthesis, and yield loss. Plants respond through coordinated regulatory networks that connect early stress perception with ion balance, osmotic adjustment, hormone signaling, transcript regulation, and protein modification. Recent advances have identified several sensory and signaling modules involved in

Pathway Crosstalk and Metabolic Relay in the Phenylpropanoid-Carotenoid Axis Orchestrate Petal Coloration in Kalanchoe blossfeldiana.

Background: Flower color is a primary determinant of ornamental value in flowering plants. Although the biosynthetic pathways of major petal pigments have been extensively documented, the systems-level mechanisms coordinating color diversification across multiple cultivars of Kalanchoe blossfeldiana remain less understood. This study aimed to establish a comparative metabolomic and transcriptomic framework to elucidate these mechanisms. Methods: Petal tissues were collected from five cultivars with white, yellow, orange, red, and purple-red flowers at the full-bloom stage. Metabolomics was per

CtChi19: a valuable gene for improving resistance to Botrytis cinerea and Alternaria alternata in Carthamus tinctorius.

Safflower (Carthamus tinctorius L.) is an economically important crop with both medicinal and oil-producing values, However, its production is severely threatened by fungal diseases, including those caused by Alternaria alternata and other pathogenic fungi. To date, molecular investigations into the disease resistance mechanisms of safflower remain largely limited. In this study, proteomic analysis was performed on safflower following infection by A. alternata, identifying 490 and 921 differentially expressed proteins (DEPs) at 48 hours post-infection (hpi) and 72 hpi, respectively, Among thes

Focus on DnaJ protein research from Arabidopsis to crop improvement.

DnaJ proteins (Hsp40s) are essential components of the cellular proteostasis network, functioning as molecular co-chaperones that regulate protein folding, stability, and stress-responsive homeostasis in plants. Beyond their classical role as Hsp70 partners, accumulating evidence demonstrates that DnaJ proteins participate in diverse biological processes by integrating proteostasis with hormonal, developmental, and defense signaling pathways. This review synthesizes recent advances in the structural diversity, evolutionary distribution, and functional specialization of DnaJ proteins across mod

Species-specific functional divergence of MAH1-L genes enhances wax biosynthesis and drought tolerance in cotton (Gossypium hirsutum L.).

Drought remains a major limitation in Cotton (Gossypium spp.) productivity, with high-yielding Gossypium hirsutum being stress-sensitive, whereas Gossypium arboreum exhibits inherent drought resilience. The superior drought tolerance of G. arboreum is closely associated with its higher epicuticular wax content, yet the mechanistic basis underlying this trait is poorly understood. Here, we revealed species-specific divergence of Midchain Alkane Hydroxylase-Like (MAH1-L) genes between the two species and evaluated their potential to enhance drought tolerance in G. hirsutum. Introduction of GaMAH

Unravelling Mechanisms of Acquired Drought Tolerance by Priming in Tomato Using Integrative Multi-Omics Phenotypic Analysis.

The escalating frequency and intensity of recent droughts threaten global crop production and food security. Although drought priming is crucial for plant drought resilience, the mechanisms imparting drought stress memory remain poorly understood, especially in crops such as tomato. This study elucidates the mechanisms of drought stress memory in tomato by integrating phenotypic screening with transcriptomic and proteomic analyses. Initial screening of 30 genotypes identified varieties with contrasting basal drought tolerance as indicated by gas exchange and drought damage index. Subsequent pr
