Paeoniflorin alleviates urushiol-induced pruritus in mice by inhibiting Erk/CCL2 pathway.

Paeoniflorin (PAE), a bioactive monoterpene glycoside derived from the roots of Paeonia lactiflora (P. lactiflora), has been traditionally used in East Asian medicine for its anti-inflammatory and immunomodulatory properties. However, its potential therapeutic effects on plant allergen-induced dermatitis, particularly poison ivy-related allergic contact dermatitis (ACD), remain unexplored. This study aimed to investigate the immune mechanisms underlying urushiol-induced ACD, a prevalent environmental allergy caused by poison ivy, and to evaluate the efficacy of PAE in alleviating inflammatory

Heat/non-heat treatment alleviates β-conglycinin-triggered food allergy reactions by modulating the Th1/Th2 immune balance in a BALB/c mouse model.

BACKGROUND: With the increasing popularity of plant protein-based diets, soy proteins are favored as the most important source of plant protein worldwide. However, potential food allergy risks limit their use in the food industry. This work aims to reveal the mechanism of β-conglycinin-induced food allergy, and to explore the regulatory mechanism of heat treatment and high hydrostatic pressure (HHP) treatment in a BALB/c mouse model. RESULTS: Our results showed that oral administration of β-conglycinin induced severe allergic symptoms in BALB/c mice, but these symptoms were effectively allevi

Graft incompatibility between pepper and tomato elicits an immune response and triggers localized cell death.

Graft compatibility is the capacity of two plants to form cohesive vascular connections. Tomato and pepper are incompatible graft partners; however, the underlying cause of graft rejection between these two species remains unknown. We diagnosed graft incompatibility between tomato and diverse pepper varieties based on weakened biophysical stability, decreased growth, and persistent cell death using viability stains. Transcriptomic analysis of the junction was performed using RNA sequencing, and molecular signatures for incompatible graft response were characterized based on meta-transcriptomic

Melon CmRLCK VII-8 kinase genes CmRLCK27, CmRLCK30 and CmRLCK34 modulate resistance against bacterial and fungal diseases in Arabidopsis.

Receptor-like cytoplasmic kinases (RLCKs) represent a distinct class of receptor-like kinases crucial for various aspects of plant biology, including growth, development, and stress responses. This study delves into the characterization of RLCK VII-8 members within cucurbits, particularly in melon, examining both structural features and the phylogenetic relationships of these genes/proteins. The investigation extends to their potential involvement in disease resistance by employing ectopic overexpression in Arabidopsis. The promoters of CmRLCK VII-8 genes harbor multiple phytohormone- and stre

Efficient In Vitro and In Vivo Activity of Glyco-Engineered Plant-Produced Rabies Monoclonal Antibodies E559 and 62-71-3.

Rabies is a neglected zoonotic disease that has no effective treatment after onset of illness. However the disease can be prevented effectively by prompt administration of post exposure prophylaxis which includes administration of passive immunizing antibodies (Rabies Immune Globulin, RIG). Currently, human RIG suffers from many restrictions including limited availability, batch-to batch inconsistencies and potential for contamination with blood-borne pathogens. Anti-rabies monoclonal antibodies (mAbs) have been identified as a promising alternative to RIG. Here, we applied a plant-based trans

Prebiotic potential of Jerusalem artichoke (Helianthus tuberosus L.) in Wistar rats: effects of levels of supplementation on hindgut fermentation, intestinal morphology, blood metabolites and immune response.

BACKGROUND: Many studies have been conducted using purified prebiotics such as inulin or fructooligosaccharides (FOS) as nutraceuticals, but there is very little information available on the prebiotic potential of raw products rich in inulin and FOS, such as Jerusalem artichoke (JA; Helianthus tuberosus L.). The present experiment aimed to evaluate the prebiotic effects of JA tubers in rats. RESULTS: Seventy-two Wistar weanling rats divided into four groups were fed for 12 weeks on a basal diet fortified with pulverized JA tubers at 0 (control), 20, 40 and 60 g kg(-1) levels. Morinda citrifolia noni water extract enhances innate and adaptive immune responses in healthy mice, ex vivo, and in vitro.

Although Morinda citrifolia (noni) has long been used in traditional medicines for human diseases, its molecular and cellular mechanism of immunostimulatory ability to improve human health under normal healthy conditions is not fully elucidated. This study aimed to investigate the in vitro and in vivo immunostimulatory activity of M. citrifolia fruit water extract treated with enzymes (Mc-eWE). In vitro studies revealed that Mc-eWE stimulated the cells by inducing nitric oxide (NO) production and the expression of inflammatory cytokines, such as interleukin (IL)-1β, IL-6, IL-12, tumor necrosis

GRF1 fine-tunes the copper-responsive transcription module of CPK3-bHLH107 by manipulating interaction and phosphorylation.

Previously, the crucial role of calcium-dependent protein kinase 3 (CPK3) in Cu2+-triggered immunity (CuTI) has been established in Arabidopsis. CPK3 promotes the phosphorylation of basic helix-loop-helix 107 (bHLH107), thereby activating the expression of 1-aminocyclopropanecarboxylic acid synthesis 8 (ACS8) and downstream defense responses. However, the precise mechanisms underlying the fine-tuning of CPK3-mediated phosphorylation of bHLH107 remain unclear. To elucidate the mechanism underlying the inactivation of the CPK3-bHLH107 module, we identified a 14-3-3 protein, GRF1 (GF14χ), among c

CIP elicitors on the defense response of A. macrocephala and its related gene expression analysis.

Plant-derived elicitor is a new type of plant vaccine developed in the contemporary era, and it has safe and broad application prospects in organic agriculture. Research on defense mechanisms triggered by elicitor has become a hot topic in recent years. The Chrysanthemum indicum polysaccharide (CIP) obtained by separation and purification from Chrysanthemum indicum was used as an elicitor in this work. This elicitor has been shown to be effective in Atractylodes macrocephala Koidz (A. macrocephala) against Sclerotium rolfsii sacc (S. rolfsii) infection and soil-borne diseases. However, the mec

Abscisic acid promotes proteasome-mediated degradation of the transcription coactivator NPR1 in Arabidopsis thaliana.

Proteasome-mediated turnover of the transcription coactivator NPR1 is pivotal for efficient activation of the broad-spectrum plant immune responses known as localized acquired resistance (LAR) and systemic acquired resistance (SAR) in adjacent and systemic tissues, respectively, and requires the CUL3-based E3 ligase and its adaptor proteins, NPR3 and NPR4, which are receptors for the signaling molecule salicylic acid (SA). It has been shown that SA prevents NPR1 turnover under non-inducing and LAR/SAR-inducing conditions, but how cellular NPR1 homeostasis is maintained remains unclear. Here, w

Propolis allergy. (II). The sensitizing properties of 1,1-dimethylallyl caffeic acid ester.

As shown in the preceding paper, propolis or bee-glue is the cause of an increasing number of allergic reactions in persons using it in external preparations and cosmetics. Propolis and its main contact allergen, 1,1-dimethylallyl caffeic acid ester, designated LB-1, show strong sensitizing properties in patients as well as in guinea pig experiments. 9 patients have been patch tested with this compound, 8 of whom reacted strongly. Chemical separation of different propolis samples and poplar bud extracts reveal that LB-1 is always present. Poplar bud secretion is the bee's major source for prop

Rhizobial type III effectors as regulators of root-nodule symbiosis in legumes.

The rhizobia-legume nitrogen-fixing symbiosis begins when plants perceive rhizobial Nod factors (NF), activating the Common Symbiosis Signalling Pathway (CSSP) that drives nodule organogenesis. Rhizobia also inject Type III effectors (T3E) into host cells through a Type III Secretion System (T3SS) related to those of pathogenic bacteria. These effectors were long viewed only as suppressors of plant immunity and determinants of host compatibility, but this picture has changed. Recent studies revealed that rhizobial effectors (known as Nodulation Outer Proteins, Nops) such as NopT, NopM, NopL, N…

A glycine rich protein (NbRBGA) confers broad-spectrum tolerance to begomovirus-betasatellite complex through activation of salicylic acid signalling pathway.

Begomoviruses, the largest genus of the family Geminiviridiae, are ssDNA viruses transmitted by whiteflies. Betasatellite molecules are occasionally found to be associated with begomoviruses and greatly influence viral pathogenesis. During viral invasion, plants activate a well-coordinated antiviral immune response to inhibit the propagation of the pathogen. In this study, we have reported an RNA-binding glycine-rich protein (RBGA) getting differentially expressed during infection due to begomovirus-betasatellite complex in Nicotiana benthamiana. Over-expression of the protein successfully res

Immunomodulatory Effects of Medicinal Plants used for Vitiligo in Traditional Persian Medicine.

BACKGROUND: Vitiligo is a hypopigmentation disorder that affects 1% of the world's population. Vitiligo causes white spots on the skin, mucous membranes, or white hair by destroying skin melanocytes. The pathogenesis of vitiligo is unknown but autoimmune, autocytotoxic, and neural mechanisms are suggested. According to the autoimmune theory, in people with vitiligo, immune cells invade and damage melanocytes. T cells are more commonly present in vitiligo patients' skin and remain in the lesion site, which is composed of CD8 and CD4 T cells. Many studies have been conducted on the presence and
