Phylogeny of trypanosomes as inferred from the small and large subunit rRNAs: implications for the evolution of parasitism in the trypanosomatid protozoa.

Sequences of the small rRNA genes and partial sequences of the large rRNA genes were obtained by PCR amplification from a variety of vertebrate trypanosomes. The trypanosome species and hosts included Trypanosoma avium from a bird, T. rotatorium from an amphibian, T. boissoni from an elasmobranch, T. triglae from a marine teleost and T. carassii from a freshwater teleost. Phylogenetic relationships among these species and other representatives of the family Trypanosomatidae were inferred using maximum likelihood, maximum parsimony and evolutionary parsimony. The trypanosomatid tree was rooted

Mitochondrial Genome Characteristics of Rhodobryum laxelimbatum and Comparative Analysis of RNA Editing and Codon Usage Bias With the Chloroplast Genome.

Rhodobryum laxelimbatum is a traditional medicinal plant belonging to the genus Rhodobryum. To date, studies on this genus have focused primarily on its geographical distribution, chemical constituents, and biological activities, whereas genomic data within the genus are limited. The mitochondrial genome of R. laxelimbatum was sequenced, assembled, and annotated. Genomic structural features and simple sequence repeats (SSRs) were comprehensively characterized. Comparative analyses of RNA editing patterns and codon usage bias were performed using the mitochondrial and chloroplast genomes of R.

tRF-Ser-GCT-108 regulates thermal recovery in HUVECs by targeting MAPK1.

BACKGROUND: Transfer RNA-derived fragments (tRFs) are emerging as critical post-transcriptional regulators in stress responses and tissue repair; meanwhile, endothelial cells orchestrate burn-wound vascular recovery by sprouting new vessels, restoring perfusion, and guiding epithelial regeneration. However, the role of tRFs in thermal injury-induced endothelial cell dysfunction remains unexplored. Here, we identify a novel stress-induced tRF, tRF-Ser-GCT-108, as a key suppressor of endothelial cell recovery following thermal damage. METHODS: Small-RNA sequencing was performed on paired normal

Viral hepatitis and the cascade of care among people living with HIV in the Asia-Pacific.

BACKGROUND: Although the prevalence and mortality of hepatitis is high in the Asia-Pacific region, few studies are available on the diagnosis, treatment, and cure rates for viral hepatitis among people living with HIV in this area. This study aims to report the cascade of care (CoC) for hepatitis B (HBV) and C (HCV) among people living with HIV receiving combined antiretroviral therapy (ART). METHODS: Patients enrolled in the TREAT Asia HIV Observational Database Low Intensity Transfer (TAHOD-LITE) cohort, on ART, and with follow-up data from 2010 to 2019 were included. Patients were determin

West Nile Virus Activity in a Winter Roost of American Crows (Corvus brachyrhynchos): Is Bird-To-Bird Transmission Important in Persistence and Amplification?

Since its emergence in North America, West Nile virus (WNV) has had a large impact on equines, humans, and wild bird communities, yet gaps remain in our understanding of how the virus persists at temperate latitudes when winter temperatures preclude virus replication and host-seeking activity by mosquito vectors. Bird-to-bird transmission at large communal American Crow roosts could provide one mechanism for WNV persistence. Herein, we describe seasonal patterns of crow and Culex mosquito abundance, WNV infection rates, and the prevalence of WNV-positive fecal samples at a winter crow roost to

IGF-1 Gene Transfer Modifies Inflammatory Environment and Gene Expression in the Caudate-Putamen of Aged Female Rat Brain.

Brain aging is characterized by chronic neuroinflammation caused by activation of glial cells, mainly microglia, leading to alterations in homeostasis of the central nervous system. Microglial cells are constantly surveying their environment to detect and respond to diverse signals. During aging, microglia undergoes a process of senescence, characterized by loss of ramifications, spheroid formation, and fragmented processes, among other abnormalities. Therefore, the study of changes in microglia during is of great relevance to understand age-related declines in cognitive and motor function. We

The role of exosomes in ankylosing spondylitis: from biological features and functional cargo to clinical applications.

Exosomes are small extracellular vesicles secreted by nearly all cell types and widely distributed in body fluids. They not only mediate intercellular material transfer but also play an important role in the regulation of immune pathways. Given their diverse biological functions, studies investigating the regulatory roles of exosomes in ankylosing spondylitis (AS) are receiving increasing attention. The functions of exosomes in AS largely depend on their bioactive cargo, including microRNAs (miRNAs), circular RNAs (circRNAs), long non-coding RNAs (lncRNAs), proteins, and other molecules. In ad

Real-time measurements of aminoglycoside effects on protein synthesis in live cells.

The spread of antibiotic resistance is turning many of the currently used antibiotics less effective against common infections. To address this public health challenge, it is critical to enhance our understanding of the mechanisms of action of these compounds. Aminoglycoside drugs bind the bacterial ribosome, and decades of results from in vitro biochemical and structural approaches suggest that these drugs disrupt protein synthesis by inhibiting the ribosome's translocation on the messenger RNA, as well as by inducing miscoding errors. So far, however, we have sparse information about the dyn

Neutrophils Promote Metabolic Dysfunction-Associated Steatotic Liver Disease Through Extracellular Vesicle-mediated Lipid Transfer.

Metabolic dysfunction-associated steatotic liver disease (MASLD) and metabolic dysfunction-associated steatohepatitis are leading causes of chronic liver disease, yet the contribution of neutrophils to early hepatocellular lipid accumulation remains poorly understood. Here, we investigated the role of neutrophils in hepatic lipid deposition during early MASLD development. Neutrophils acquired extracellular fatty acids (FAs) via FATP2 and CD36 and stored them as triglycerides (TGs). These lipid-laden neutrophils (LNs) did not utilize FAs for energy production or lipid mediator synthesis but ins

T cell transfer into immunodeficient mice uncovers a paralytic phenotype beyond colitis.

Adoptive transfer of CD45RBhigh T cells into immunodeficient mice is a well-established model for inducing colitis. Here, we report for the first time that a subset of these mice unexpectedly developed paralysis, a phenotype resembling experimental autoimmune encephalomyelitis (EAE). Paralytic mice exhibit the hallmark features of central nervous system (CNS) pathology, including spinal cord demyelination and immune cell infiltration, with prominent involvement of ex-Th17 (also called Th1-like) cells. The presence of ex-Th17 cells in the CNS of the affected mice suggests their contributory rol

Lipocalin-2 expression identifies an intestinal regulatory neutrophil population during acute graft-versus-host disease.

Acute graft-versus-host disease (aGVHD) is a life-threatening complication of allogeneic hematopoietic cell transplantation (allo-HCT), for which therapeutic options are limited. Strategies to promote intestinal tissue tolerance during aGVHD may improve patient outcomes. Using single-cell RNA sequencing, we identified a lipocalin-2 (LCN2)-expressing neutrophil population in mice with intestinal aGVHD. Transfer of LCN2-overexpressing neutrophils or treatment with recombinant LCN2 reduced aGVHD severity, whereas the lack of epithelial or hematopoietic LCN2 enhanced aGVHD severity and caused micr

Regulatory T cell adoptive transfer alters uterine immune populations, increasing a novel MHC-IIlow macrophage associated with healthy pregnancy.

Intrauterine fetal demise (IUFD) - fetal loss after 20 weeks - affects 6 pregnancies per 1,000 live births in the United States, and the majority are of unknown etiology. Maternal systemic regulatory T cell (Treg) deficits have been implicated in fetal loss, but whether mucosal immune cells at the maternal-fetal interface contribute to fetal loss is under-explored. We hypothesized that the immune cell composition and function of the uterine mucosa would contribute to the pathogenesis of IUFD. To investigate local immune mechanisms of IUFD, we used the CBA mouse strain, which naturally has mid-

Potential roles of extracellular vesicles as a noninvasive tool for prenatal genetic diagnosis.

The rate of infertility is increasing owing to genetic and environmental factors. Consequently, assisted reproductive technology has been introduced as an alternative. Bearing in mind the global trend toward the transfer of only one embryo, there is an increasing trend for assessing embryo quality before transfer through prenatal genetic diagnosis (PGD) tests. This ensures that the best-quality embryos are implanted into the uterus. In the in vitro fertilization cycle, PGD is not only used for diseases or quality checks before embryo freezing but also for evaluating unfortunate risks, such as

Non-antibiotic pharmaceuticals promote the transmission of multidrug resistance plasmids through intra- and intergenera conjugation.

Antibiotic resistance is a global threat to public health. The use of antibiotics at sub-inhibitory concentrations has been recognized as an important factor in disseminating antibiotic resistance via horizontal gene transfer. Although non-antibiotic, human-targeted pharmaceuticals are widely used by society (95% of the pharmaceuticals market), the potential contribution to the spread of antibiotic resistance is not clear. Here, we report that commonly consumed, non-antibiotic pharmaceuticals, including nonsteroidal anti-inflammatories (ibuprofen, naproxen, diclofenac), a lipid-lowering drug (

Mapping the DNA-Binding Motif of Scabin Toxin, a Guanine Modifying Enzyme from Streptomyces scabies.

Scabin is a mono-ADP-ribosyltransferase toxin/enzyme and possible virulence factor produced by the agriculture pathogen, Streptomyces scabies. Recently, molecular dynamic approaches and MD simulations revealed its interaction with both NAD+ and DNA substrates. An Essential Dynamics Analysis identified a crab-claw-like mechanism, including coupled changes in the exposed motifs, and the Rβ1-RLa-NLc-STTβ2-WPN-WARTT-(QxE)ARTT sequence motif was proposed as a catalytic signature of the Pierisin family of DNA-acting toxins. A new fluorescence assay was devised to measure the kinetics for both RNA an

Care of Patients With HIV Infection: Antiretroviral Drug Regimens.

The advent of combination antiretroviral drug regimens has transformed HIV infection from a fatal illness into a manageable chronic condition. All patients with HIV infection should be considered for antiretroviral therapy, regardless of CD4 count or HIV viral load, for individual benefit and to prevent HIV transmission. Antiretroviral drugs affect HIV in several ways: entry inhibitors block HIV entry into CD4 T cells; nucleotide and nucleoside reverse transcriptase inhibitors prevent reverse transcription from RNA to DNA via chain-terminating proteins; nonnucleoside reverse transcriptase inhi

Individual fish tank arrays in studies of Lepeophtheirus salmonis and lice loss variability.

In studies of the salmon louse Lepeophtheirus salmonis (Krøyer, 1837), experimental design is complicated by a highly variable and unpredictable lice loss among common experimental tanks and a substantial rate of host transfer within tanks. When fish hosting L. salmonis are maintained in individual tanks, unspecific effects such as host transfer, louse predation by cohabitant hosts and agonistic host interactions are excluded. This study suggests that it is possible to maintain Atlantic salmon Salmo salar infected with L. salmonis in an array of small, single fish tanks and, by doing so, provi

Vertical transmission of tick-borne encephalitis virus between generations of adapted reservoir small rodents.

Vertical transmission of tick-borne encephalitis virus (TBEV) between generations of the small rodents-red voles Myodes rutilus Pallas (previously known as Clethrionomys rutilus Pallas) was shown for naturally infected reservoir hosts and after experimental infection with different sublethal doses of the viral strains. For wild red voles and for their progeny born in 240-280 days after experimental infection of their parents the TBEV was detected in up to 90% of samples by RT-PCR, ELISA and bioassays. Small amounts of the TBEV RNA found in embryos, placenta and blood cells could serve as evide

Delivery of a retroviral vector expressing human beta-glucuronidase to the liver and spleen decreases lysosomal storage in mucopolysaccharidosis VII mice.

Mucopolysaccharidosis VII (MPS VII) is caused by beta-glucuronidase (beta-gluc) deficiency and results in lysosomal storage due to the inability to degrade glycosaminoglycans. Transfer of a beta-gluc gene into the liver reduces hepatic pathology as well as storage in other organs via uptake of secreted protein. A Moloney murine leukemia-based retroviral vector expressing the human beta-gluc cDNA was injected intravascularly into MPS VII mice during hepatocyte replication, which was induced with im injection of an adenoviral vector that transiently expressed hepatocyte growth factor (Ad.CMV.
