The AIDS is a chronic disease and the researchers still exert their high efforts to reach the cure of HIV infection. The most common treatment is the antiretroviral therapy (cART) and the virus can be more effected if the patients stop using cART. The other problem is that the CD8+ T cells might be exhausted by persistent immune activation by cART. This paper introduces fractional-order into a mathematical model of HIV infection combining with stem cell therapy and control the infection by the immune system cells (CD8+ T cells). The paper introduced the numerical solutions for the mathematical

Improvement of culture system and increasing the proliferation of spermatogonia stem cells under in vitro condition are the essential treatment options for infertility before autologous transplantation. Therefore, the present study aimed to evaluate the proliferation of human spermatogonia stem cells on the electrospun polycaprolactone/gelatin nanocomposite. Therefore, for this purpose, nanofiber porous scaffolds were prepared using the electrospinning method and their structures were then confirmed by SEM. After performing swelling, biodegradability and cell adhesion tests, human spermatogoni

Three-dimensional (3D) culture models, particularly multi-spheroid models, are becoming increasingly essential in cancer drug discovery, particularly in stem cell and cancer stem cell (CSC) research. However, analytical methods for 3D multi-spheroid models, especially for single-cell and single-spheroid analysis in CSC research, remain limited. To address this gap we developed 3D multi-spheroid cholangiocarcinoma models that incorporate a CSC live-cell biosensor and a novel analysis method, 3D Surface Integrative Spheroid Profiling (3D-SiSP), utilizing high-content confocal imaging. 3D-SiSP qu

This study investigates the role of Annexin A1 (ANXA1) in regulating T cell function and its implications in bone marrow adiposity in aplastic anaemia (AA). Utilizing single-cell sequencing analysis, we compared bone marrow tissues from AA patients and healthy individuals, focusing on T cell subgroups and their impact on bone marrow pathology. Our findings reveal a significant activation of CD8+ T cells in AA, driven by reduced ANXA1 expression. This heightened T cell activity promotes adipogenesis in bone marrow-derived mesenchymal stem cells via IFN-γ secretion. Overexpression of ANXA1 was f

BACKGROUND: Alopecia is a prevalent condition that significantly impacts appearance, mental health, and overall quality of life. Current clinical treatments for alopecia include medication, hair transplantation, low-level light therapy, and stem cell therapy, among others. However, the effectiveness of these treatments remains far from ideal. Extracellular vesicles (EVs) have emerged as a promising therapeutic strategy, yet challenges, such as mass production and clinical application persist. This review aims to explore recent advancements in the use of EVs for the treatment of alopecia.

The study aimed to investigate the fanconi anemia (FA)-related and stemness-related genes in lung cancer (LC) patients. Firstly, we identified stemness-related genes through weighted gene co-expression network analysis combined with TCGA database. Further combined stemness-related genes with FA-related genes to screen for prognostic-related genes. Risk score was constructed from the screened genes and comprehensive bioinformatics analyses were performed. Finally, single-cell data and in vitro experiment were used to validate our results. We screened a total of eight genes to construct a risk s

Autosomal dominant mutations in transcription factor 4 (TCF4) are associated with a rare syndromic form of Autism Spectrum Disorder (ASD) called Pitt-Hopkins Syndrome (PTHS). Here, we report the generation of a collection of induced pluripotent stem cells (iPSCs) from 5 patients diagnosed with PTHS and 5 familial controls. These patient-derived iPSCs contain a variety of mutations within the TCF4 gene, possess a normal karyotype and express all the appropriate pluripotent stem cell markers. These novel patient lines will be a useful resource for the research community to study PTHS and the fun

Alzheimer's disease (AD) is a heterogeneous clinicobiological syndrome involving impaired protein clearance, neuroinflammation, synaptic and cellular injury, network dysfunction, and progressive functional decline. Amyloid-directed antibodies provide an evidence-based treatment backbone for selected patients with early symptomatic AD, but adjunctive strategies may be needed to address other disease mechanisms. This Perspective proposes a clearance-repair-activation framework for hypothesis-driven research. Clearance includes established pharmacological treatment and risk management as well as

Model organisms are vital for biomedical research and drug testing but face high costs, complexity, and ethical issues. While newer techniques like organoids and assembloids have shown improvements, they still remain inadequate in addressing all research needs. In this study, we present a new method for maintaining the prostate gland of the earthworm, Eudrilus eugeniae ex vivo and examine its potential for regeneration and drug screening. We successfully maintained the earthworm prostate gland in cell culture media for over 200 days, with observed beating behavior confirming its viability. Apo

With the rapid development of society and the economy, population aging has become a common challenge faced by many countries in the world today. Structural and functional changes in the cardiovascular system can occur with age, increasing the incidence and severity of cardiovascular diseases in older adults. Due to the limited regenerative capacity of myocardial cells, myocardial infarction and its resulting heart failure and congenital heart disease have become the number one killer of human health. At present, the treatment of cardiovascular diseases includes drug therapy and nondrug therap

INTRODUCTION: Human induced pluripotent stem cell (iPSC)-derived cardiomyocytes is one of the most widely used cell-based models that resulted from the discovery of how non-embryonic stem cells can be differentiated into multiple cell types. In just one decade, iPSC-derived cardiomyocytes went from a research lab to widespread use in biomedical research and preclinical safety evaluation for drugs and other chemicals.
AREAS COVERED: This manuscript reviews data on toxicology applications of human iPSC-derived cardiomyocytes. We detail the outcome of a systematic literature search on their use

Chemotherapeutic agents used in cancer treatment cause damage to both oocytes and granulosa cells, resulting in follicle loss and consequently premature ovarian failure. In this study the animals were randomly assigned to three groups: control (n = 6), chemotherapy (n = 6), and stem cell treatment (n = 6). Ovarian failure was induced in the chemotherapy and stem cell groups by intraperitoneal administration of cyclophosphamide (200 mg/kg) on days 1 and 8. Ovarian stromal stem cells (OSSCs) were isolated from the ovaries of 4-week-old donor rats (n = 2) using the explant culture method. On day

Neurogenesis is principally restricted to the subventricular zone of the lateral ventricle wall and the subgranular zone of the hippocampal dentate gyrus in physiological situations. However, neuronal stem cells are known to be mobilized into the post- and peristroke area and we have demonstrated that appropriate support of these stem cells, achieved by therapeutic angiogenesis, enhances neuroregeneration followed by neuronal functional recovery in an experimental stroke model. We also found that neural stem cells are mobilized in patients after stroke, as well as in animal models. Based on th

Minimal residual disease (MRD), both before and after transplantation, is a clinically important yet relatively poorly defined aspect of allogeneic hematopoietic stem cell transplantation (alloHSCT). The clinical relevance of MRD in the context of alloHSCT has been demonstrated by its association with the development of clinical relapse. However, with the possible exception of chronic myeloid leukemia (CML), the specific techniques, timing, frequency, and clinical utility, relative to improvement in patient outcomes, for monitoring MRD in the setting of alloHSCT has yet to be clearly defined.
