Transport and catabolism of D-fructose by Spirillum itersomii.

Spirillum itersonii ATCC 12639 utilized d-fructose but neither d-glucose nor d-gluconate as a sole source of carbon and energy. The substrate saturation kinetics for d-fructose and d-glucose uptake by whole cells indicated the presence of a carrier-mediated transport system for d-fructose but not for d-glucose. The d-fructose uptake activity was induced (10- to 12-fold increase) during growth on d-fructose-Casamino Acids (CA) or d-glucose-CA medium, but not CA alone. d-Fructose uptake activity was stimulated by Na(+) or Li(+), but was inhibited by KCN, NaN(3), 2,4-dinitrophenol, and p-chlorome

Properties of alpha-aminoisobutyric acid transport in a thermophilic microorganism.

Uptake of alpha-aminoisobutyric acid (AIB) by a leucine-tyrosine auxotroph of a thermophilic microorganism starved for leucine was studied. AIB was taken up by the cells against a substantial concentration gradient (300:1) and was present there in a free and unchanged form. Various energy inhibitors and sulfhydryl reagents strongly inhibited the accumulation of AIB. AIB uptake obeyed saturation kinetics, and the Lineweaver-Burk plot is characterized by a biphasic curve. AIB most probably shares a common transport system(s) with alanine, serine, and glycine. A mutant defective in l-alanine upta

Morphology and functional cytology of the small intestinal mucosa in malabsorptive disorders and other diseases.

Small intestinal biopsies from 98 patients have been studied macroscopically, histologically, and histochemically, and the results correlated with laboratory and clinical observations. Neither a convoluted nor a flat mucosa can be considered diagnostic for any specific disease. A flat mucosa was found in one adult with intestinal Hodgkin's disease and in another following total gastrectomy. Histochemically all specimens from subjects with coeliac disease and idiopathic steatorrhoea showed a deficiency of succinic dehydrogenase in the epithelial cells, and the histochemical test for this enzyme

Cooling inflammation through metabolism with 4-octyl itaconate.

Itaconate is an immunomodulatory metabolite that links metabolism and inflammation. Li et al. uncover a mechanism by which itaconate and 4-octyl itaconate suppress cytokine signaling through the alkylation of tyrosine kinase 2 and Janus kinase 1. This study reveals a direct metabolic control of inflammation and highlights its therapeutic potential in sepsis.

4-Octyl Itaconate Alleviates Airway Eosinophilic Inflammation by Suppressing Chemokines and Eosinophil Development.

4-Octyl itaconate (4-OI) is a derivative of the Krebs cycle-derived metabolite itaconate and displays an array of antimicrobial and anti-inflammatory properties through modifying cysteine residues within protein targets. We have found that 4-OI significantly reduces the production of eosinophil-targeted chemokines in a variety of cell types, including M1 and M2 macrophages, Th2 cells, and A549 respiratory epithelial cells. Notably, the suppression of these chemokines in M1 macrophages was found to be NRF2-dependent. In addition, 4-OI can interfere with IL-5 signaling and directly affect eosino

Metabolic Signatures of Performance in Elite World Tour Professional Male Cyclists.

BACKGROUND AND OBJECTIVE: Metabolomics studies of recreational and elite athletes have been so far limited to venipuncture-dependent blood sample collection in the setting of controlled training and medical facilities. However, limited to no information is currently available to determine if findings in laboratory settings are translatable to a real-world scenario in elite competitions. The goal of this study was to define molecular signatures of exertion under controlled exercise conditions and use these signatures as a framework for assessing cycling performance in a World Tour competition.

Molecular architecture of OXGR1 reveals an evolutionary conserved mechanisms for metabolite surveillance.

The ability of cells to sense and respond to metabolic signals is fundamental to life, yet the molecular mechanisms underlying metabolite surveillance remain incompletely understood. Here, we elucidate the structural basis of metabolite recognition by OXGR1, a G Protein-Coupled Receptor (GPCR) that senses key intermediates in the tricarboxylic acid (TCA) cycle. Using cryo-electron microscopy, we determined cryo-EM structures of OXGR1 bound to α-ketoglutarate (AKG), itaconate (ITA), and structurally related metabolites succinate (SUC) and maleate (MA). These structures reveal a positively charg

Isolation, structure determination and antibacterial activities of succinamide conjugate diacid from Acinetobacter sp. BJ-L.

Strain BJ-L was isolated from the a water sample taken from Xiao Yue River in Beijing and identified as Acinetobacter sp. BJ-L based on the study of its morphology, physiology, biochemistry and 16S rRNA gene sequence. A new antimicrobial substance was produced after the strain was incubated in potato extract medium at 15°C for 72 h. The antimicrobial substance was sequentially purified by reduced pressure condensation, EtOAc extract, and silica gel column chromatography. The structure of the antimicrobial substance was elucidated as succinamide conjugate diacid (SCD) by spectroscopic data inte

Biopsy evidence of left ventricular myocardial abnormality in patients with mitral-leaflet prolapse and chest pain.

Left ventricular endomyocardial biopsy specimens were obtained at cardiac catheterisation in 11 patients with mitral-leaflet prolapse, chest pain, and normal coronary arteries. Specimens were examined for birefringence before and after contraction induced by adenosine triphosphate and were also subjected to histochemical analysis. Biopsy specimens from a control group of patients with rigorously defined normal left ventricular function and another control group with poor left ventricular function were also studied. Biopsy specimens from patients with mitral-leaflet prolapse showed a markedly s

Isolation of anaerobic respiratory mutants of Shewannella putrefaciens and genetic analysis of mutants deficient in anaerobic growth on Fe3+.

A genetic approach was used to study (dissimilatory) ferric iron (Fe3+) reduction in Shewanella putrefaciens 200. Chemical mutagenesis procedures and two rapid plate assays were developed to facilitate the screening of Fe3+ reduction-deficient mutants. Sixty-two putative Fe3+ reduction-deficient mutants were identified, and each was subsequently tested for its ability to grow anaerobically on various compounds as sole terminal electron acceptors, including Fe3+, nitrate (NO3-), nitrite (NO2-), manganese oxide (Mn4+), sulfite (SO3(2-)), thiosulfate (S2O3(2-)), trimethylamine N-oxide, and fumara

Huntington's Disease: Astrocytes Shift to Fatty Acid Metabolism.

A recent study by Polyzos et al. (Cell Metab., 2019) shows that astrocytes in a Huntington disease (HD) mouse model switch from glycolysis to fatty acid oxidation (FAO), causing increased superoxide radical anion production and loss of succinate dehydrogenase (SD) activity. Blocking mitochondria reactive oxygen species (ROS) with an antioxidant compound called XJB-5-151 reversed lipofuscin formation and protected the mice.

Chronic intestinal pseudoobstruction with myopathy and ophthalmoplegia. A muscular biochemical study of a mitochondrial disorder.

The association of chronic intestinal pseudoobstruction with ophthalmoplegia has been reported previously in visceral myopathies. We report a case of this association in which muscle mitochondria had a crystalline appearance, a dense core, and decreased cytochrome c oxidase and succinate cytochrome c reductase activities. The absence of evident mitochondrial DNA deletion in the skeletal muscle of this patient does not exclude the possibility of localized deletion or mutation of mitochondrial DNA in digestive muscle.

Tracheal obstruction caused by tree needles in a cat.

Tracheal obstruction was suspected in a 1-year-old female domestic shorthair cat. Clinical signs consisted of inspiratory and expiratory dyspnea and cyanosis. The cat did not improve when treated with oxygen, epinephrine, prednisolone sodium succinate, and aminophylline. Bronchoscopy revealed obstruction of the trachea and carina by a 2.5-cm twig and needles from a Douglas fir tree. Needles had migrated into distal airways. The cat was euthanatized at the owner's request.

Mutagenicity of chloroaniline/lignin metabolites in the Salmonella/microsome assay.

Chloroanilines are constituents of many agrochemicals and have been found to be metabolized to succinic acid conjugates, e.g., succinamides and succinimides. The mutagenic potential of five chloroanilines and their succinamides and succinimide derivatives have been tested with two strains of Salmonella typhimurium (TA98 and TA100) with and without rat hepatic microsomal fraction. None of the compounds produced a dose response effect with a two-fold increase in revertants indicating that these compounds are not mutagens or promutagens in these assays.

Adrenocortical insufficiency in the dog.

Primary adrenocortical insufficiency is described in 3 dogs. Clinical signs included marked depression, vomiting, diarrhoea, weakness and bradycardia. Diagnosis was based on clinical signs and biochemical findings of hyperkalaemia, hyponatraemia and a lowered plasma Na+ : K+ ratio. Treatment during the acute illness consisted of intravenous fluids and hydrocortisone sodium succinate. Fludrocortisone acetate has maintained all dogs in good health. One dog was on treatment for 4 years.

INFLUENCE OF IRON PREPARATIONS ON OCCULT BLOOD TESTS.

Ferrous fumarate (Fersamal) and ferrous carbonate (Ferrodic) may produce false-positive results with the orthotolidine tablet tests (Occultest and Hematest) if administered to patients being investigated for alimentary bleeding. Misleading results are also likely if the benzidine test is employed as a filter-paper procedure. Ferrous sulphate, gluconate, succinate, iron and ammonium citrate, and other preparations of iron have no influence on the modern tablet tests for occult alimentary bleeding.

Generation and characterization of six human induced pluripotent stem cell lines (hiPSCs) from three individuals with SSADH Deficiency and CRISPR-corrected isogenic controls.

Succinic Semialdehyde Dehydrogenase Deficiency (SSADHD) is an ultra-rare autosomal recessive neurometabolic disorder caused by ALDH5A1 mutations presenting with autism and epilepsy. Here, we report the generation and characterization of human induced pluripotent stem cells (hiPSCs) derived from fibroblasts of three unrelated SSADHD patients - one female and two males with the CRISPR-corrected isogenic controls. These individuals are clinically diagnosed and are being followed in a longitudinal clinical study.
