Effects of Hot-Air Drying Temperature on Biochemical Quality, Drying Kinetics, and Predictive Modeling of Ziziphus jujuba Fruits.

Jujube (Ziziphus jujuba Mill.) is an important functional fruit with high levels of bioactive compounds. However, its high moisture content limits its postharvest shelf life. The biochemical properties of jujube fruits influenced by hot-air drying at 50, 60, and 70 °C were studied and predictive models were evaluated to optimize drying. The drying kinetics were modeled using the six classical thin-layer models and artificial neural network (ANN) approach. The Midilli-Kucuk model was the best classical model, and the ANN model showed the best predictive performance, with the RMSE values improve

The role of nanosilica in antioxidant production, nutrition and photochemical efficiency to mitigate sulfur deficiency in Capsicum annuum L.

Sulfur deficiency limits the development of bell pepper in various cultivation regions worldwide. The use of nanosilica is a promising alternative to mitigate the nutritional impairments caused by stress; however, its efficacy in bell pepper under sulfur-deficiency and its potential benefits for non-stressed plants remain unclear. This study aimed to investigate the impact of sulfur-deficiency on bell pepper and evaluate the effects of nanosilica in mitigating these effects, as well as to explore its benefits under sulfur-sufficient conditions. The experiment was conducted in a greenhouse usin

Plasma-activated water as a solvent for foliar fertilizer: effects on the growth and biochemical properties of oregano (Origanum vulgare L.).

BACKGROUND: Plasma-activated water (PAW) is a promising innovation in agriculture, with the potential to promote seed germination and to increase plant growth and development. PAW contains reactive oxygen and nitrogen species being considered as substitute for standard fertilizers. It may enhance also plants’ natural defense mechanisms through activating several physiological and metabolic pathways. RESULTS: Oregano was foliar‑sprayed with either distilled water (control) or PAW (prepared by exposing to low temperature glow plasma for 30 min), with and without the addition of commercial fertil

Revealing pathogenesis-associated metabolites in Histoplasma capsulatum through comprehensive metabolic profiling.

During infection, Histoplasma capsulatum yeasts interact with a variety of phagocytic cells, where macrophages represent an important niche for long-term intracellular fungal survival and replication. In the phagosomes of macrophages, a hostile environment where most microorganisms are killed, Histoplasma not only survives but overcomes several biological challenges and proliferates intracellularly. To better understand the characteristics of intracellular Histoplasma and the phagosomal environment, a metabolomic platform was used to analyze Histoplasma yeasts cultured on different carbon sour

Comparative evaluation of red and white aril genotypes of Manila tamarind for fruit physicochemical and bioactive attributes.

The cultivation and trade of underutilized fruits are gaining prominence worldwide, largely on account of their capacity to contribute to a nutritious diet. Manila tamarind [Pithecellobium dulce (Roxb.) Benth] is a fast-growing, nitrogen-fixing tree, with a fairly high tolerance to abiotic stresses. Despite significant potential in terms of food, fodder, timber, and medicine, it has largely remained an orphan crop. There is a dearth of systematic research on the exploration, conservation, and genetic improvement of Manila tamarind. Our study aimed to assess the genetic variability for commerci

Diabetes Leads to Alterations in Normal Metabolic Transitions of Pregnancy as Revealed by Time-Course Metabolomics.

Women with diabetes during pregnancy are at increased risk of poor maternal and neonatal outcomes. Despite this, the effects of pre-gestational (PGDM) or gestational diabetes (GDM) on metabolism during pregnancy are not well understood. In this study, we utilized metabolomics to identify serum metabolic changes in women with and without diabetes during pregnancy and the cord blood at birth. We observed elevations in tricarboxylic acid (TCA) cycle intermediates, carbohydrates, ketones, and lipids, and a decrease in amino acids across gestation in all individuals. In early gestation, PGDM had el

Abscisic Acid Lowers Postharvest Grape Acidity via PP2C-ALMT9-Dependent Malate Transport.

This study uncovers a novel concentration-dependent signaling network through which abscisic acid (ABA) modulates acid metabolism in grape berries. Treatment with 800 μM ABA significantly decreased malate and citrate synthesis and transport while enhancing glucose and fructose accumulation, optimizing the sugar-acid ratio. Multi-omics analysis revealed differential changes in 96 organic acids and identified a key malate transporter, VvALMT9. Through yeast two-hybrid screening, the VvPP2C-VvALMT9-VvSnRK2.3-VvCDPK interaction network was characterized in a dual regulatory mode. Under low-ABA con

Dietary Patterns during Pregnancy Are Associated with Risk of Gestational Diabetes Mellitus.

Maternal dietary patterns before and during pregnancy play important roles in the development of gestational diabetes mellitus (GDM). We aimed to identify dietary patterns during pregnancy that are associated with GDM risk in pregnant U.S. women. From a 24 h dietary recall of 253 pregnant women (16-41 years) included in the National Health and Nutrition Examination Survey (NHANES) 2003-2012, food items were aggregated into 28 food groups based on Food Patterns Equivalents Database. Three dietary patterns were identified by reduced rank regression with responses including prepregnancy body mass

Collection and Analysis of Phloem Lipids.

The plant phloem is a long-distance conduit for the transport of assimilates but also of mobile developmental and stress signals. These signals can be sugars, metabolites, amino acids, peptides, proteins, microRNA, or mRNA. Yet small lipophilic molecules such as oxylipins and, more recently, phospholipids have emerged as possible long-distance signals as well. Analysis of phloem (phospho)lipids, however, requires enrichment, purification, and sensitive analysis. This chapter describes the EDTA-facilitated approach of phloem exudate collection, phase partitioning against chloroform-methanol for

The Chiron Approach to (3R,3aS,6aR)-Hexahydrofuro[2,3-b]furan-3-ol, a Key Subunit of HIV-1 Protease Inhibitor Drug, Darunavir.

We describe an enantioselective synthesis of (3R,3aS,6aR)-hexahydrofuro[2,3-b]furan-3-ol which is a key subunit of darunavir, a widely used HIV-1 protease inhibitor drug for the treatment of HIV/AIDS patients. The synthesis was achieved in optically pure form utilizing commercially available sugar derivatives as the starting material. The key steps involve a highly stereoselective substrate-controlled hydrogenation, a Lewis acid catalyzed anomeric reduction of a 1,2-O-isopropylidene-protected glycofuranoside, and a Baeyer-Villiger oxidation of a tetrahydrofuranyl-2-aldehyde derivative. [Isoenzyme differentiation of gamma-glutamyltransferase by concanavalin A and Con- A-Sepharose (author's transl)].

In this investigation a new possibility of isoenzyme-differentiation of the gamma-glutamyl-transferase (GGT) (EC Nr.2.3.2.2.) was demonstrated by Concanavalin A and Con A-Sepharose. Because of the different sugar content of the glycoproteins distinction between liver- and kidney-GGT is possible. Furthermore it was possible for the first time to show a different precipitation behaviour of one glycoprotein to Concanavalin A in certain diseases. In cases of alcoholic hepatitis GGT looses its Concanavalin A-affinity because of increased neuraminic acid concentration. The possible reasons of the di

Generation and characterization of four dilutions of diesel engine exhaust for a subchronic inhalation study.

Exposure atmospheres for a rodent inhalation toxicology study were generated from the exhaust of a 2000 Cummins ISB 5.9L diesel engine coupled to a dynamometer and operated on a slightly modified heavy-duty Federal Test Procedure cycle. Exposures were conducted to one clean air control and four diesel exhaust levels maintained at four different dilution rates (300:1, 100:1, 30:1, 10:1) that yielded particulate mass concentrations of 30, 100, 300, and 1000 microg/m3. Exposures at the four dilutions were characterized for particle mass, particle size distribution (reported elsewhere), detailed c

Acetic acid mediates pectin structural remodeling to enhance cellulose binding in potato cell wall.

This study investigates how acetic acid pretreatment modifies pectin structure and its interaction mechanism with cellulose to regulate textural integrity of potato slices (Solanum tuberosum L.) during thermal processing. Potato slices were pretreated with distilled water and acetic acid, and changes in firmness, sugar composition and cell wall architecture were evaluated. Chelate soluble pectin was extracted, purified and characterized by galacturonic acid content, monosaccharide composition and UV-vis analysis. Acetic acid treated samples retained higher firmness and mitigated heat induced d

The changes in Lemna minor metabolomic profile: A response to diclofenac incubation.

Metabolomics is an emerging approach that investigates the changes in the metabolome profile. In the present study, Lemna minor -considered as an experimental aquatic plant model- was incubated with 10 and 100 μM diclofenac (DCF) for 96 h, respectively. Knowing that DCF is internationally often problematic in wastewater effluents and that it might affect particularly the metabolic profiles in aquatic plants, mainly the oxidoreductase, dehydrogenase, peroxidase, and glutathione reductase activities, here it was hypothesized (H) that in the common duckweed, DCF might increase the phenolic and fl

Intact Endogenous Metabolite Analysis of Mice Liver by Probe Electrospray Ionization/Triple Quadrupole Tandem Mass Spectrometry and Its Preliminary Application to in Vivo Real-Time Analysis.

Probe electrospray ionization (PESI) is a recently developed ionization technique that enables the direct detection of endogenous compounds like metabolites without sample preparation. In this study, we have demonstrated the first combination use of PESI with triple quadrupole tandem mass spectrometry (MS/MS), which was then applied to intact endogenous metabolite analysis of mice liver, achieving detection of 26 metabolites including amino acids, organic acids, and sugars. To investigate its practicality, metabolic profiles of control and CCl4-induced acute hepatic injury mouse model were mea
