The tumor microenvironment is more than a backdrop for malignant cells: stromal fibroblasts, extracellular matrix, abnormal vasculature, hypoxia, and immunosuppressive myeloid and lymphoid populations act together to limit both drug delivery and treatment response. Here, we examine nanomedicine along three linked axes: stromal remodeling, hypoxia control, and photodynamic immune activation. CAF-directed and matrix-responsive carriers can improve perfusion and intratumoral transport; however, because fibroblasts are heterogeneous, subtype-selective reprogramming is safer than wholesale depletio…

Prostate cancer (PCa) remains a significant clinical challenge, particularly in localized therapy, where unmet medical needs persist for effective and targeted treatments. This study introduces an innovative solution by developing the first responsive spermine (Spm) hydrogel with an autonomous sensing function, designed to advance local therapy for PCa. Our hydrogel effectively loads Spm and releases Spm in the acidic tumor microenvironment (TME), providing a responsive therapeutic platform. Incorporating the di-lysine-7-nitrobenzofurazan (NBD-KK) dye, which is initially quenched by aggregate …

Oncolytic virotherapy has evolved from a tumor-selective cytolytic strategy into a programmable immunotherapeutic platform that reshapes the tumor microenvironment (TME). Oncolytic viruses (OVs) selectively infect malignant cells, induce immunogenic cell death, release tumor antigens, and activate innate and adaptive immunity, potentially converting cold tumors into inflamed states. However, clinical translation remains limited by antiviral clearance, heterogeneous delivery, stromal barriers, immunosuppressive cells, and incomplete integration with cellular immunotherapy. This review synthesiz…

BACKGROUND: Limited therapeutic options are available for patients with advanced-stage mycosis fungoides (MF), and the 5-year survival rate is 25%. Due to its critical role in MF pathogenesis, the IL4/IL13 pathway presents a promising target for treatment. Here, we analyzed blocking of IL4Rα, the common subunit of the IL4 and IL13 receptors, within the advanced-stage MF cutaneous tumor microenvironment (TME).
METHODS: Cellular Indexing of Transcriptomes and Epitopes by sequencing was employed to assess the transcriptional profile of the cells expressing the IL4 and IL13 receptors within the a…

Human papillomavirus (HPV) significantly influences cervical cancer progression and treatment, yet its interactions with the tumor microenvironment remain incompletely understood. We performed single-cell and spatial transcriptomic sequencing on cervical cancer samples to explore these interactions. By aligning sequencing reads to a merged HPV16-human reference genome, we characterized HPV16 heterogeneity and its association with host states at the single-cell and single-gene levels. E5 transcriptional activity was negatively associated with the host interferon response, indicating a role in i…

Neutrophils are the most abundant immune cells in bone marrow (BM); their differentiation, maturation, and clearance are tightly regulated by the BM microenvironment. Multiple myeloma (MM) is a malignant plasma cell disorder that leads to multi-organ damage, including BM disruption, which may subsequently affect resident neutrophils. The present study was designed to systematically characterize BM neutrophils in MM, to advance the understanding of their potential biological roles in disease initiation and progression, and to identify potential neutrophil-based biomarkers of clinical relevance.

Patient-derived organoids (PDOs) have emerged as promising models for predicting personalized drug responses in cancer therapy. However, the absence of essential immune and stromal components limits their ability to recapitulate the tumor microenvironment. Here, we established a total of 30 patient-derived organoids (PDOs) from 79 patients with locally advanced (LA) and recurrent/metastatic (R/M) head and neck squamous cell carcinoma (HNSCC). These PDOs maintained sustained expansion capacity and preserved the histopathological characteristics and genomic heterogeneity of their parental tumors…

Gastric cancer (GC) remains a leading cause of cancer-related mortality, with chemoresistance posing a critical barrier to effective treatment. Tumor-associated macrophages (TAMs), particularly the immunosuppressive M2-polarized subset, are emerging as pivotal mediators of chemoresistance within the tumor microenvironment (TME). TAMs promote resistance through multifaceted mechanisms, including activation of pro-survival signaling pathways, induction of epithelial-mesenchymal transition (EMT), and enhancement of angiogenesis. For instance, M2-like TAMs secrete CXCL5, which activates the PI3K/A…

Metabolic reprogramming within the tumor microenvironment (TME) plays a central role in cancer progression and immune evasion, with L-arginine metabolism emerging as a key regulatory axis. Arginase overexpression depletes intratumoral L-arginine, thus suppressing T-cell proliferation while fuelling tumor growth through polyamine biosynthesis. OATD-02, a novel dual arginase (ARG1/ARG2) inhibitor, reprograms tumor metabolism by restoring L-arginine availability and reducing the levels of polyamines, thereby shifting the TME toward a more immunostimulatory state. Unlike ARG1-selective inhibitors …

BACKGROUND/AIMS: Pancreatic ductal adenocarcinoma is an aggressive neoplasm with poor prognosis. The metabolic properties of its desmoplastic stroma are hypothesized to contribute to disease progression. Aberrant metabolism in tumor cells and desmoplastic stroma results in the accumulation of metabolic by-products, with lactate being the most important, released into the extracellular space by monocarboxylate transporters (MCTs). This study aimed to determine the prognostic value of these markers and identify potential therapeutic targets.

RAC1A159V is a hotspot mutation associated with poor prognosis in several cancers. By gene editing, we generated endogenous homozygous and heterozygous RAC1A159V mutations, which result in up-regulated RAC1 activity and mammalian target of rapamycin (mTOR) signaling. RAC1A159V tumors grow faster than RAC1WT tumors in immune-proficient mice and are resistant to anti-programmed death protein 1 (PD1). Flow cytometry and scRNA-seq analyses reveal that RAC1A159V cells form "cold" tumors with an immunosuppressive microenvironment and reduced tumor-immune cell interactions. Mechanistically, RAC1A159V
