OBJECTIVES: We conducted a confirmatory mapping survey to verify previous survey results, which suggested that lymphatic filariasis is endemic in some parts of Lagos State, Nigeria.
METHODS: Finger-prick blood samples were collected from 2787 children aged 7-16 years in six local government areas (LGAs) across 153 targeted schools. Circulating filarial antigen (CFA) levels were measured using the filaria test strip, and LGAs were classified as endemic if there were up to three positive cases (d = 3).
RESULTS: Children who tested positive for CFA were detected in all the six endemic LGAs, sur

Potent inflammatory responses from host-parasite interactions in lymphatic filariasis are driven by macrophage polarization, which criticall … d in the infective L3 larval stage using in-silico analysis approach. Among all, Wb123, a serpin of Wuchereria bancrofti, the most predominantly found filarial worm, was cloned and purified to establish its role in alternative activation. We observed elevated markers of alternative activation; namely CD163, arginase-1, IL-6 and pSTAT3 expression, following r …

BACKGROUND: The lymphatic filariasis (LF) elimination program in all sixty-three endemic districts of Nepal was based on annual mass drug administration (MDA) using a combination of diethylcarbamazine (DEC) and albendazole for at least 5 years. The MDA program was started in the Parsa district of the Terai region and at least six rounds of MDA were completed between 2003 and 2017 in all filariasis endemic districts of Central Nepal. Transmission Assessment Survey (TAS) report indicated that circulating filarial antigen (CFA) prevalence was below the critical value i.e., ≤ 2% in selected LF end

BACKGROUND: Lymphatic filariasis (LF) elimination program in Tanzania started in 2000 in response to the Global program for the elimination of LF by 2020. Evidence shows a persistent LF transmission despite more than a decade of mass drug administration (MDA). It is advocated that, regular monitoring should be conducted in endemic areas to evaluate the progress towards elimination and detect resurgence of the disease timely. This study was therefore designed to assess the status of Wuchereria bancrofti infection in Culex quinqefasciatus and Anopheles species after six rounds of MDA in Masasi D…

Benzimidazole anthelmintics have long been employed for the control of soil-transmitted helminth infections. Flubendazole (FBZ) was approved in 1980 for the treatment of gastrointestinal nematode infections in both veterinary and human medicine. It has also long been known that parenteral administration of FBZ can lead to high macrofilaricidal efficacy in a variety of preclinical models and in humans. As part of an effort to stimulate the discovery and development of new macrofilaricides, particularly for onchocerciasis, research has recently been devoted to the development of new formulations

A long-standing gap in lymphatic filariasis epidemiology is quantifying the potential effect that heterogeneous infection processes occurring in the major mosquito vector genera may have on parasite transmission and control. Although previous studies have focussed on examining the forms of the density dependent mechanisms regulating larval infection in various mosquito genera, there has been little work done thus far in investigating how such differential processes might interact with density-dependent processes occurring in other stages of the parasite life cycle to influence overall transmis

BACKGROUND: Filariasis and its consequences are a major health problem in tropical countries, including the Indian subcontinent. Despite its high incidence, it is unusual to find microfilaria in fine needle aspiration cytology (FNAC) smears. We present a case of subcutaneous firm to cystic swelling, aspiration of which revealed a large number of microfilaria.
CASE: A 30-year-old man presented with a chain of intermittent, firm swellings in both arms. FNAC of the swellings revealed a large number of 4 microfilariae with associated giant cell reaction and inflammatory cell-like eosinophils.

The current global initiative to eliminate lymphatic filariasis is a major renewed commitment to reduce or eliminate the burden of one of the major helminth infections from resource-poor communities of the world. Mathematical models of filariasis transmission can serve as an effective tool for guiding the scientific development and management of successful community-level intervention programmes by acting as analytical frameworks for integrating knowledge regarding parasite transmission dynamics with programmatic factors. However, the power of these tools for supporting control interventions w

The filarial-specific humoral responses (IgG1, IgG2, IgG3, IgG4 and IgE) to a Brugia pahangi antigen was assessed in 9 groups of adult individuals from a Wuchereria bancrofti-endemic area in north-east Tanzania. In 5 of the groups, individuals were negative for microfilariae (mf) and circulating filarial antigen (CFA) and had leg lymphoedema of varying severity ranging from early to more advanced grades. A 6th group had mixed grades of lymphoedema and were actively infected with mf and/or CFA. Three groups of asymptomatic individuals with different infection status (mf+CFA+; mf-CFA+; mf-CFA-) …

The global lymphatic filariasis elimination programme incorporates disability management along with transmission control, to ensure 'a visible impact' on those who are already affected by the disease. The common manifestations of lymphatic filariasis like lymphoedema; elephantiasis and hydrocele result from irreversible damage caused to the lymphatics by the adult worms. Only palliative treatment in the form of physical methods and surgery is available for lymphoedema and elephantiasis. Hydrocele can be corrected by surgery. The most distressing aspect of lymphatic filariasis is the attacks of

Tea industry is a labor intensive agro-industry and filariasis is mostly localized among the tea garden workers in Assam. The workers live inside the gardens in colonies. Studies conducted in two cosecutive years revealed that among the host seeking Culex quinquefasciatus average infection rate was 4.6% and with 2.1 larvae per mosquito. The overall prevalence of infective mosquitos was 0.8% with average L3 load of 2.0 per mosquito. The probability of infected mosquitos surviving to have complete development of filarial larvae (13 days) was 0.17. The expectation of infective life was 1.416 days

Field studies were carried out to determine the impact of mass human treatment with ivermectin on the survival of anthropophagic mosquitoes of the Anopheles punctulatus complex (Diptera: Culicidae), the vectors of lymphatic filariasis and malaria in Papua New Guinea. In a village where mass treatment had been given, using 400 microg/kg ivermectin plus 6 mg/kg diethylcarbamazine citrate (DEC), we performed pre- and post-treatment collections of freshly blood-engorged mosquitoes from the same nine bedrooms. All blood-fed mosquitoes collected less than 4 days after mass treatment died within 9 da

Four groups of patients were selected: 16 patients with clinical evidence of obstructive filarial lymphangiopathy without microfilaraemia; 12 patients with clinical evidence of obstructive filarial lymphangiopathy with microfilaraemia and 9 patients with microfilaraemia. Two control groups were also included. Blood films, sera and hydrocele fluid samples were collected from all subjects. Polyclonal antibody against Dirofilaria immitis worm homogenate was prepared, fractionated and conjugated with HRP. Both polyclonal antibody and monoclonal antibody (AD12) were used in a sandwich ELISA. Using

One hundred and sixty-two individuals from a community in Kwale District, Kenya, endemic for bancroftian filariasis, were selected for a study on the IgE response to filarial antigen (prepared from adult Brugia pahangi). Following clinical and parasitological examination, the individuals were grouped into different categories, based on the presence/absence of microfilaraemia, the presence/absence of acute or chronic (hydrocele or elephantiasis) clinical manifestations, and age. The total and filarial-specific IgE responses were evaluated in all individuals, and the responses in the various cat

A comparative study was conducted for detecting antibody levels of bancroftian microfilaremia cases by using three serologic tests, IFAT, ELISA and dot-ELISA. A total of 102 serum specimens were collected from bancroftian microfilaremia cases. The positive rate was 89.2% (91/102) with IFAT (serum dilution > or = 1:20), 84.3% (86/102) with ELISA (OD value > or = 0.440) and 89.2% (91/102) with dot-ELISA (serum dilution > or = 1:80), respectively (P > 0.05); while in healthy individuals from non-endemic areas the false positive rate was 0(0/30), 1.8(1/55) and 0(0/54), respectively. The results in

Twenty-one villages from ten provinces and one autonomous region with different mf rates in population and different levels of mean mf density per 60 microliters blood in positive cases and representative species of filaria and vector after DEC treatment were selected as study sites. In these villages a longitudinal investigation for a period of 3-5 years was carried out without adopting any control measure. The main results were: 1. Among 279 residual microfilaremia cases, 235 (84.2%) turned negative spontaneously and successively. The negative conversion rates were 86.6% (46/53) in malayan c

The pathogenesis of lymphoedema in patients infected with Wuchereria bancrofti or Brugia malayi remains unclear. Lymph stasis and local immunological reactions seem to play the main role. In order to discriminate between the obstructive and immunological effects of the parasite, a comparative histological study of skin specimens obtained from two groups of patients, one with filarial and the other with postsurgical lymphedema of lower extremities, was performed. In both groups patients suffered lymph stasis, in the first due to filariasis, in the other due to removal or irradiation of pelvic l…
