A novel chemoselective phosphine-mediated tandem reaction between nonsubstituted MBH carbonates and aryl trifluoromethyl ketones is described. The product selectivity of the reaction is easily tunable by changing the ratios of the two reactants, and mono- or bicyclic bistrifluoromethylated vinyl γ-butenolide products can be prepared with good chemoselectivity in modest-to-good yields and diastereoselectivities. The formation of the bicyclic γ-butenolide structures via a one-pot four-step sequence under phosphine catalysis is unprecedented.

A general and direct organocatalytic asymmetric vinylogous Michael reaction of gamma-butenolide with alpha,beta-unsaturated ketones was investigated with a multifunctional primary amine salt as catalyst. The reaction enables straightforward access toward synthetically versatile gamma-substituted butenolides from simple 2(5H)-furanone with satisfactory yields, diastereo- and enantioselectivities (up to 30:1 dr and 95-99% ee).

The first facile and efficient route to pyranose-fused butenolides from furanose scaffolds, convenient for scaling up production, is described. Wittig olefination of 1,2-O-isopropylidene pentofuranos- or hexofuranos-3-uloses with a resonance-stabilized ylide led to the stereoselective formation of the (Z)-alpha,beta-unsaturated ester. In the presence of acid labile 5-O- or 5,6-di-O-protecting groups, acid hydrolysis of the Wittig product resulted in isomerization to the pyranose form and spontaneous lactonization to give the target molecules in good overall yield.

Four new aromatic butenolides, asperimides A-D (1-4), together with a known analogue, butyrolactone I (5), were isolated from solid cultures of a tropical endophytic fungus Aspergillus terreus.
