Amelioration of Acrolein-Induced Cytotoxicity, Oxidative Stress, DNA Damage, Apoptosis and Mitochondrial Dysfunction in Human Pancreas Cells by Pycnogenol.

Acrolein (AC) is a potential environmental contaminant that poses a serious threat to human health and the environment. Studies on human diseases that can result from AC have rapidly increased in recent years. Pycnogenol (PYC), a natural phenolic compound, has therapeutic and protective potential against some cancers. Currently, there is insufficient data on the potential effects of AC and PYC on pancreatic cells or on their interaction. Because toxicological effects of AC and interactions between AC and PYC in human pancreatic cells have not yet been investigated, we evaluated the toxicologic…

Apiaceous vegetables protect against acrolein-induced pulmonary injuries through modulating hepatic detoxification and inflammation in C57BL/6 male mice.

Acrolein (Acr) is a reactive aldehyde in the environment. Acr causes oxidative stress and a cascade of catalytic events and has, thereby, been associated with increased risk of pulmonary diseases. Whether apiaceous vegetables (API) consumption can prevent Acr-induced pulmonary toxicity has not yet been explored hence, we investigated the effects of API on Acr-induced pulmonary damages in C57BL/6J mice. The mice were assigned into either negative control [NEG group; American Institute of Nutrition (AIN)-93G diet only], positive control (POS group; AIN-93G+Acr) or API intervention group (API gro…

Inflammatory effects of acrolein, crotonaldehyde and hexanal vapors on human primary bronchial epithelial cells cultured at air-liquid interface.

The cytotoxicity of aldehydes was studied using human primary bronchial epithelial cells (PBEC) cultured at the air-liquid interface (ALI) or under submerged conditions. PBEC were exposed for 30min via the air phase to acrolein (0.1-1mg/m3), crotonaldehyde (1.5-15mg/m3) or hexanal (22-221mg/m3) or under submerged conditions to acrolein (0.1 and 0.2mg/L), crotonaldehyde (1 and 2mg/L) or hexanal (10 and 20mg/L). Cell culture medium was collected 8h and 24h post-exposure and analyzed for interleukin-8 (IL-8) and matrix metalloprotein-9 (MMP-9). The gene expression of inflammatory and oxidative st…

Acrolein, a highly toxic aldehyde generated under oxidative stress in vivo, aggravates the mouse liver damage after acetaminophen overdose.

Although acetaminophen-induced liver injury in mice has been extensively studied as a model of human acute drug-induced hepatitis, the mechanism of liver injury remains unclear. Liver injury is believed to be initiated by metabolic conversion of acetaminophen to the highly reactive intermediate N-acetyl p-benzoquinoneimine, and is aggravated by subsequent oxidative stress via reactive oxygen species (ROS), including hydrogen peroxide (H2O2) and the hydroxyl radical (•OH). In this study, we found that a highly toxic unsaturated aldehyde acrolein, a byproduct of oxidative stress, has a major rol…

Notes from the field: Acute pesticide-related illness resulting from occupational exposure to acrolein - Washington and California, 1993-2009.

Acrolein is an aquatic herbicide used in the western United States to prevent impaired water flow in irrigation canals. Despite its toxicity, few cases of acrolein-related illness have been reported in the literature. On August 15, 2012, an irrigation district notified the Washington State Department of Labor & Industries (L&I) of acrolein-related illness in one of its pesticide applicators. L&I inspected the site and interviewed the exposed worker, coworkers, and employer. The Washington State Department of Health assisted by obtaining medical records, interviewing the patient and hospital st…

S-alk(en)yl-cysteine sulfoxides in Allium species are excellent acrolein scavengers: implications for secondary antioxidants in plants.

Reactive carbonyl species (RCS), such as acrolein (Acr), are generated through the degradation of lipid peroxides and exert cytotoxic effects. To identify natural RCS scavengers, we examined 80% ethanol extracts from 46 angiosperm species for Acr-trapping activity using an HPLC-based assay. Strong activities were observed in several taxa, including garlic, spinach, avocado, broccoli, and lotus. In garlic, the active metabolite was identified as S-allyl-L-cysteine sulfoxide (alliin), a characteristic Allium amino acid. Alliin and its S-(1E)-propenyl and S-methyl derivatives (isoalliin and methi…

Acrolein-induced oxidative and nitrosative stress and its impact on respiratory mechanics in mice assessed using the forced oscillation technique.

Acrolein exposure in the lung was studied to determine the effect of acrolein on airway responsiveness and pulmonary mechanics, as measured by the forced oscillation technique (FOT), under conditions of airway activation by methacholine (MCh), as well as with no activation, using the negative pressure-driven forced expiratory (NPFE) maneuvers to assess quasi-static lung compliance. Direct intratracheal acrolein was applied to C57BL/6 J male mice in dosages of 0 (saline vehicle-only) or 4 mg/kg, with FOT and NPFE assessments made 48 hr post-acrolein administration. Our results suggest that lipi…

Co-modification of amylase by acrolein with formaldehyde or methylglyoxal: Impact on enzyme function, structure, and cellular stress in two cell lines.

Reactive carbonyl species (RCS), such as acrolein (Acr), formaldehyde (FA), and methylglyoxal (MGO) are highly reactive aldehydes co-existing in processed foods and food materials. RCS may modify the digestive enzymes individually while whether they act together on the enzymes remains unknown. In this study, the co-action of RCS on α-amylase was studied. The results show that when Acr is present together with MGO or FA, they simultaneously modify amylase to form carbonylated proteins and inhibit the activity of amylase. The co-action of RCS increases the modification sites of amylase, enhances…

Coherent Mixing of Singlet and Triplet States in Acrolein and Ketene: A Computational Strategy for Simulating the Electron-Nuclear Dynamics of Intersystem Crossing.

We present a theoretical study of intersystem crossing (ISC) in acrolein and ketene with the Ehrenfest method that can describe a superposition of singlet and triplet states. Our simulations illustrate a new mechanistic effect of ISC, namely, that a superposition of singlets and triplets yields nonadiabatic dynamics characteristic of that superposition rather than the constituent state potential energy surfaces. This effect is particularly significant in ketene, where mixing of singlet and triplet states along the approach to a singlet/singlet conical intersection occurs, with the spin-orbit c…

The multiplex interactions and molecular mechanism on genotoxicity induced by formaldehyde and acrolein mixtures on human bronchial epithelial BEAS-2B cells.

Aldehydes are common air pollutants with carcinogenicity. Genotoxicity of single aldehyde has been studied well, but the combined genotoxicity is rarely known. Here, we evaluated the combined genotoxicity of formaldehyde and acrolein on BEAS-2B cells in terms of DNA strands breakage, chromosome damage and gene mutation below subcytotoxic concentrations covering smoking-related concentrations. Meanwhile, the molecular mechanism was investigated further based on oxidative stress, DNA-protein crosslinks (DPCs), cell cycle and DNA damage-repair pathway. Co-exposure to formaldehyde and acrolein mix…

A Synthetic Snake-Venom-Based Tripeptide Protects PC12 Cells from the Neurotoxicity of Acrolein by Improving Axonal Plasticity and Bioenergetics.

The synthetic peptide p-BTX-I is based on the native peptide (formed by glutamic acid, valine and tryptophan) isolated from Bothrops atrox venom. We have previously demonstrated its neuroprotective and neurotrophic properties in PC12 cells treated with the dopaminergic neurotoxin 1-methyl-4-phenylpyridinium (MPP+). Now, we have investigated the neuroprotective effects and mechanisms of p-BTX-I against the toxicity of acrolein in PC12 cells. Studies have demonstrated that acrolein might play an important role in the etiology of Alzheimer's disease (AD), which is characterized by neuronal and sy…

Proteomic analysis reveals shared biological pathways linking acrolein to biomolecular changes in the acute phase of rat spinal cord injury.

Spinal cord injury (SCI) pathology is highly difficult to treat due to substantial heterogeneity in injury presentation and spread, along with unclear mechanisms linking damage to pathology. Damages from injury forces (primary injury) are exacerbated by a series of biochemical events that follow the initial damage and injure additional tissue, known as secondary injury. Reactive aldehydes, such as acrolein, play a key role in propagating secondary injury cascades following SCI. Targeting acrolein after SCI has demonstrated therapeutic potential in limiting injury spread and pathology. However,…

Adenosine protects cardiomyocytes against acrolein-induced cardiotoxicity by enhancing mitochondrial homeostasis, antioxidant defense, and autophagic flux via ERK-activated FoxO1 upregulation.

Acrolein is a ubiquitous gaseous air pollutant and endogenous toxicant, which poses strong risk for oxidative stress-related diseases such as cardiovascular disease. Adenosine has been identified as potential therapeutic agent for age-related cardiovascular disease, while the molecular mechanisms underlying its cardioprotection remain elusive. In the present study, we investigated the myocardial protective effects and the mechanism of adenosine on acrolein-induced toxicity in H9c2 cells and primary neonatal rat cardiomyocytes. We found that acrolein caused apoptosis of cardiomyocytes resulting…

P2X7 Receptor Blockade Protects Against Acrolein-Induced Bladder Damage: A Potential New Therapeutic Approach for the Treatment of Bladder Inflammatory Diseases.

Inflammatory conditions of the urinary bladder have been shown to be associated with urothelial damage and loss of function. The purinergic P2X7 receptor has been implicated in several inflammatory conditions. The aim of this study was to investigate the role of the P2X7 receptor in acrolein-induced inflammatory damage using the porcine urinary bladder. For this purpose, an ex-vivo model of porcine urothelial damage induced by direct instillation of acrolein into the whole bladder lumen was used. To determine the role of the P2X7 receptor, the bladders were pre-incubated with a selective P2X7 …

Simultaneous dual-mode detection and removal of acrolein in food samples via a fluorescent PAA-CDs/Sulfhydryl cotton/SA hydrogel composite.

A multifunctional fluorescent carbon dot/sulfhydryl cotton/sodium alginate composite hydrogel (PAA-CDs/Sulfhydryl Cotton/SA hydrogel) had been developed, which could integrate in dual-mode optical responses with fluorescence quenching and colorimetric signals to detect and simultaneous remove acrolein in food samples. The fluorescence detection exhibited a linear range spanning from 0 to 70 μM for acrolein, along with a relatively low LOD of 20 nM. In contrast, smartphone-based RGB colorimetric quantification presented a linear range of 0-70 μM, with an LOD of 350 nM. Meanwhile, the adsorption…

Acrolein but not its metabolite, 3-Hydroxypropylmercapturic acid (3HPMA), activates vascular transient receptor potential Ankyrin-1 (TRPA1): Physiological to toxicological implications.

Acrolein, an electrophilic α,β-unsaturated aldehyde, is present in foods and beverages, and is a product of incomplete combustion, and thus, reaches high ppm levels in tobacco smoke and structural fires. Exposure to acrolein is linked with cardiopulmonary toxicity and cardiovascular disease risk. The hypothesis of this study is the direct effects of acrolein in isolated murine blood vessels (aorta and superior mesenteric artery, SMA) are transient receptor potential ankyrin-1 (TRPA1) dependent. Using isometric myography, isolated aorta and SMA were exposed to increasing levels of acrolein. Acr…
