Adenosine-induced worsening of supraventricular tachycardia.

An approximately 20-year-old to 30-year-old patient presented with a haemodynamically stable supraventricular tachycardia . The patient was managed with intravenous adenosine primarily, with two bolus doses of 6 and 12 mg. This, however, caused a rare paradoxical surge of tachycardia with mild haemodynamic compromise. The patient further required a combination of Metoprolol and Verapamil administration to slow down and reverse the arrhythmia. Following this the patient remained stable with no further episodes till discharge.

Gene therapy for adenosine deaminase deficiency.

PURPOSE OF REVIEW: Gene therapy for severe combined immunodeficiency due to adenosine deaminase deficiency has moved from the early trials of safety and feasibility to recent studies demonstrating efficacy and clinical benefit. This review describes the latest advances in gene therapy trials for this condition using peripheral blood lymphocytes or hematopoietic progenitors. RECENT FINDINGS: In the first patients with severe combined immunodeficiency due to adenosine deaminase deficiency treated with peripheral blood lymphocytes, transduced T cells have been shown to persist for over 10 years,…

Adenosine plasma concentration in pulmonary hypertension.

OBJECTIVE: In this study, we sought to appreciate the role of adenosine in the regulation of pulmonary vascular tone, especially in the case of clinical pulmonary hypertension, by investigating the relationship between endogenous plasma adenosine levels and pulmonary artery vasoconstriction. METHODS: Adenosine plasma concentrations, were measured simultaneously in the distal right pulmonary artery and in the femoral artery, both at steady state (room air) and during pure oxygen inhalation. Three clinical situations were considered: (1) normal hemodynamics [7 control subjects, mean pulmonary a…

Preparation of liposome-encapsulating adenosine triphosphate.

Liposomes encapsulating adenosine triphosphate (ATP) were prepared by sonication, and the liposomes were evaluated for use in a drug delivery system. The liposomes, which were composed of phosphatidylcholine and cholesterol, were about 1.1 microm in size, as observed under a microscope. From their size, the vesicles were thought to be multilamellar. The maximum concentration of ATP in the liposomes was 1.0 mM, when the initial concentrations of lipid and ATP were 20 mM and 300 mM, respectively. The maximum entrapment ratio of ATP in the liposomes was 88%, when the initial concentrations of lip…

[Gene therapy for adenosine deaminase deficiency].

Severe combined immunodeficiency (SCID) due to adenosine deaminase (ADA) deficiency is a fatal recessive disorder caused by mutations in the gene encoding ADA. Based on the first clinical trial of two young girls with ADA-deficient SCID by recombinant retrovirus-mediated gene transfer at the National Institute of Health of USA, we prepared to treat a four-year-old boy with ADA-deficient SCID who had been treated with PEG-ADA for 3 years. Approval to perform the clinical trial of gene therapy by using his peripheral blood T lymphocytes as the target and recombinant retroviral vector (LASN) as t…

[Ventricular fibrillation after administration of adenosine].

In the submitted case-history the authors describe a hitherto unknown proarrhythmic effect of adenosine. Adenosine was administered on account of supraventricular tachycardia and the undesirable proarrhythmic effect was so-called "sine wave-like tachycardia" and ventricular fibrillation. Proarrhythmia was resistant to electric cardioversion and resuscitation was successful probably only after the breakdown of adenosine. The very short half-life of breakdown of this antiarrhythmic is thus useful not only for antiarrhythmic strategy but, because of the possible occurrence of rare proarrhythmias,…

Adenosine deaminase in progressive systemic sclerosis.

The aim of the present study was to confirm the increase of adenosine deaminase (ADA) activity in progressive systemic sclerosis (PSS), described by Sasaki & Nakajima, and to compare plasma ADA activity of patients in different stages of the disease. Enzyme activity was measured with a colorimetric assay. The 48 patients were subdivided into 3 groups: subgroup 1 (n = 10), disease still limited to the skin; subgroup 2 (n = 21), involvement of the skin and oesophagus; and subgroup 3 (n= 17), involvement of the skin and multiple internal organs. ADA levels were highest in subgroup 3. However, the…

Adenosine triphosphate degradation in specific disease.

Two hypotheses concerning adenosine triphosphate (ATP) degradation are examined: The metabolic basis of specific disorders involves ATP degradation, and uric acid and its precursors are measurable body fluid markers of ATP degradation. These hypotheses are examined by discussing human models of ATP degradation, methods for measurement of ATP degradation, and disorders of ATP degradation.

Adenosine metabolism in human skin fibroblasts.

When normal fibroblasts were incubated in media containing various initial concentrations of [8-14C]adenosine, ranging from 0.25 to 400 microM, under conditions where product formation was linear, greater than 90% of the intracellular label was found in adenine nucleotides, largely in the form of ATP, less than 1% of the intracellular label appeared in the nucleic acids, the remaining intracellular label was found in adenosine, inosine, and hypoxanthine, and the media contained two labeled products, inosine and hypoxanthine. Production of labeled inosine and hypoxanthine from adenosine was con…

Barbiturates and calcium-activated adenosine triphosphatase.

Anaesthetic barbiturates potentiate and convulsant barbiturates inhibit the calcium-activated adenosine triphosphatase (Ca-ATPase) activity in rat brain synaptosomes. Such differential effects and consequent modification of transmitter release may be important in the contrasting actions of these classes of barbiturates in vivo.

Mood and whole blood adenosine triphosphate.

The claim of Hansen (1969) and Hansen and Dimitrakoudi (1974) that whole blood adenosine triphosphate (ATP) levels correlate with depressed affect was examined. The findings from 84 patients failed to confirm any relationship between mood and whole blood ATP. One patient showing the highest correlation (r = 0-85; p less than 0-001) between the ATP levels and mood in the original report was re-examined; no relationship was now found (r = -0-25; p greater than 0-1).

Adenosine triphosphate-induced life-threatening arrhythmia.

UNLABELLED: A 68-year-old woman with idiopathic dilated cardiomyopathy presented with a wide QRS complex regular tachycardia five days after mitral valve replacement. Adenosine triphosphate (ATP) was administered to make the correct diagnosis; however, tachycardia eventually transitioned to ventricular fibrillation, which required cardioversion. Although ATP is considered a relatively safe drug, it can cause unexpected, life-threatening arrhythmias. Careful monitoring and preparation are advised during ATP administration in the event of a regular wide QRS complex tachycardia in patients with i…

Adenosine sulfamates: Next generation of antimalarials.

Tackling the ancient infectious foe of malaria, Xie et al. (2022) uncover a novel class of nucleoside analogs that selectively hijack and inhibit the tyrosine tRNA synthase of the parasite. With high potency, good oral bioavailability, and minimal host cell toxicity, these inhibitors show promise as next-generation antimalarials.

Adenosine Induces Autophagy in Cholangiocarcinoma Cells.

BACKGROUND/AIM: Cholangiocarcinoma is a lethal disease with increasing incidence worldwide. New therapeutic compounds are urgently needed for this disease. Here, the inhibitory effect of adenosine on cholangiocarcinoma cells was studied. MATERIALS AND METHODS: Western blot analysis was used to study autophagy and flow cytometry to analyze cell death and the cell cycle. RESULTS: Cholangiocarcinoma and immortalized cholangiocytes responded to adenosine differently, and adenosine inhibited cholangiocarcinoma cell growth by inducing autophagy. Adenosine failed to activate adenylyl cyclase in cho…

Adenosine Kinase Deficiency: Report and Review.

Adenosine kinase (ADK) deficiency (OMIM [online mendelian inheritance in man]: 614300) is an autosomal recessive disorder of adenosine and methionine metabolism, with a unique clinical phenotype, mainly involving the central nervous system and dysmorphic features. Patients usually present early in life with sepsis-like symptoms, respiratory difficulties, and neonatal jaundice. Subsequently, patients demonstrate hypotonia and global developmental delay. Biochemically, methionine is elevated with normal homocysteine levels and the diagnosis is confirmed through molecular analysis of the ADK gene…
