Hypoxia, Inflammation and Necrosis as Determinants of Glioblastoma Cancer Stem Cells Progression.

Tumor hypoxic microenvironment causes hypoxia inducible factor 1 alpha (HIF-1α) activation and necrosis with alarmins release. Importantly, HIF-1α also controls the expression of alarmin receptors in tumor cells that can bind to and be activated by alarmins. Human tumor tissues possess 1-2% of cancer stem cells (CSCs) residing in hypoxic niches and responsible for the metastatic potential of tumors. Our hypothesis is that hypoxic CSCs express alarmin receptors that can bind alarmins released during necrosis, an event favoring CSCs migration. To investigate this aspect, glioblastoma stem-like c…

Damage-associated molecular patterns--emerging targets for biologic therapy of childhood arthritides.

Juvenile idiopathic arthritis (JIA) is a group of chronic childhood arthritides of unknown origin. Although the use of glucocorticoids and immunosuppressants brought a substantial improvement in treatment, the present therapeutic regime could not be considered satisfactory. As inflammation seems to be an essential part of pathogenesis of JIA, efforts have been made to develop pharmaceutical means to mitigate the innate immune system. Emerging targets for treatment are alarmins, a family of multifunctional intracellular proteins with strong pro-inflammatory activity. In the context of JIA, part…

An Optimized Feeder-Free Method for Isolation and Expansion of Functional ILC2s Isolated From Human Peripheral Blood.

Innate lymphoid cells (ILCs) are tissue-resident innate counterparts of T cells. Group 2 ILCs (ILC2s) produce cytokines such as IL-5 and IL-13 in response to alarmins, including IL-25 and IL-33 and play a key role in type 2 immunity. Lately, ILC2s have become a major focus across several research areas, including asthma, allergic lung inflammation, atopic dermatitis, and liver fibrosis. However, ILC2s are rare in human peripheral blood, making their isolation and expansion for functional studies challenging. Existing methods commonly rely on flow-cytometry-based cell sorting, which requires la…

Emerging biological treatments for asthma.

INTRODUCTION: Severe asthma is a chronic airway disease characterized by many pathomechanisms known as endotypes. Biological therapies targe … ard targeting the upstream pathways of asthma pathogenesis, coordinated by innate cytokines such as alarmins. These key proinflammatory mediators orchestrate the activation of complex cellular networks including both innate and adaptive immune responses. Alarmins include thymic stromal lymphopoietin (TSLP), interleukin-25 (IL-25), and interleukin-33 (IL-33 …

Role of Thymic Stromal Lymphopoietin in the Pathophysiology of Asthma and Clinical and Biological Effects of Blockade With Tezepelumab.

The airway epithelium is the first line of defense of the respiratory system against the external environment. It plays an active role in the initiation of immune and allergic responses against potential hazards. Among the various specialized cells and cytokines that participate in epithelium-induced responses, alarmins are particularly interesting, given their ample role in mediating T2 and non-T2 inflammatory mechanisms involved in the pathogenesis of asthma. Thymic stromal lymphopoietin (TSLP) is an alarmin with broad effects in asthma that result from its widespread action on multiple cell…

Pathogenic Role of the Damage-Associated Molecular Patterns S100A8 and S100A9 in Coxsackievirus B3-Induced Myocarditis.

BACKGROUND: The alarmins S100A8 and S100A9 are damage-associated molecular patterns, which play a pivotal role in cardiovascular diseases, inflammation, and viral infections. We aimed to investigate their role in Coxsackievirus B3 (CVB3)-induced myocarditis. METHODS AND RESULTS: S100A8 and S100A9 mRNA expression was 13.0-fold (P=0.012) and 5.1-fold (P=0.038) higher in endomyocardial biopsies from patients with CVB3-positive myocarditis compared with controls, respectively. Elimination of CVB3 led to a downregulation of these alarmins. CVB3-infected mice developed an impaired left ventricular …

Reframing paucigranulocytic asthma through genetic endotyping: a hypothesis-generating focus on the 17q21 rs7216389 locus.

PURPOSE OF REVIEW: Non-T2 asthma is currently defined by missing parameters, such as low blood eosinophils and FeNO, rather than positively identifiable mechanistic features. This definition overlaps with paucigranulocytic asthma (PGA). However, T2-biomarkers fluctuate over time, especially during glucocorticoid therapy, leading to potential over-diagnosis of T2-low asthma. Advancing beyond traditional endotyping is required for precision medicine. RECENT FINDINGS: Alarmin-driven asthma [interleukin (IL)-33, thymic stromal lymphopoietin (TSLP), IL-25] can drive inflammation even without high

Retinoid X receptor gamma dictates the activation threshold of group 2 innate lymphoid cells and limits type 2 inflammation in the small intestine.

Group 2 innate lymphoid cells (ILC2s) are crucial in promoting type 2 inflammation that contributes to both anti-parasite immunity and allergic diseases. However, the molecular checkpoints in ILC2s that determine whether to immediately launch a proinflammatory response are unknown. Here, we found that retinoid X receptor gamma (Rxrg) was highly expressed in small intestinal ILC2s and rapidly suppressed by alarmin cytokines. Genetic deletion of Rxrg did not impact ILC2 development but facilitated ILC2 responses and the tissue inflammation induced by alarmins. Mechanistically, RXRγ maintained th…

HMGB1-dependent signaling in the regulation of mast cell activity during inflammation.

BACKGROUND: Damaged cells release endogenous molecules known as alarmins into the extracellular space following cellular injury. Alarmins may function as adjuvants by interacting with PRRs to indicate danger and initiate a localized sterile inflammatory response, which facilitates tissue regeneration. A pivotal alarmin is HMGB1, which is internalized through the RAGE to notify adjacent cells about compromised homeostasis. Given the significant role of mast cells (MCs) in inflammatory processes and the critical nature of alarmins as indicators of danger, this study evaluates the hypothesis that…

The Respiratory Epithelial Barrier as an Immunopharmacological Rheostat: From Homeostatic Sentinel to Therapeutic Target in Chronic Lung Disease.

The respiratory epithelium is a dynamic immunopharmacological interface that transcends its traditional role as a passive barrier to become a central coordinator of pulmonary immunity. This review introduces and elaborates on the paradigm of the "Epithelial Immunopharmacological Rheostat," a conceptual framework that posits the epithelium as a dynamic, multidimensional signal processor that continuously calibrates the threshold of immune activation. Distinct from static models of barrier dysfunction or isolated alarmin biology, this rheostat operates through four hierarchically interacting axe

ILC2 require cell-intrinsic ST2 signals to promote type 2 immune responses.

The initiation of type 2 immune responses at mucosal barriers is regulated by rapidly secreted cytokines called alarmins. The alarmins IL-33, IL-25 and TSLP are mainly secreted by stromal and epithelial cells in tissues and were linked to chronic inflammatory diseases, such as allergic lung inflammation, or to resistance against worm infections. Receptors for alarmins are expressed by a variety of immune cells, including group 2 innate lymphoid cells (ILC2s), an early source of the type 2 cytokines, such as IL-5 and IL-13, which have been linked to atopic diseases and anti-worm immunity as wel…

NOX2 mediates NLRP3/ROS facilitating nasal mucosal epithelial inflammation in chronic rhinosinusitis with nasal polyps.

BACKGROUND: Previous investigations have provided limited insight into the role of oxidative stress in nasal mucosa inflammation. The aim of … ed siRNA inhibited these effects in vitro. Moreover, TMAO reversed the downregulation of epithelial alarmins without impacting ROS levels. CONCLUSION: This study highlights the crucial role of NOX2 as a key regulator of ROS accumulation and NLRP3 inflammasome activation in CRSwNP, underscoring its potential as a valuable therapeutic target for CRSwNP.

Emerging immune networks and targeted strategies in T2 asthma.

Asthma is a highly heterogeneous chronic inflammatory disease of the airways, among which Th2-high asthma represents the most prevalent endotype. The pathogenesis of Type 2 (T2) asthma (driven by type 2 inflammation) involves a complex immune network orchestrated by the coordinated actions of multiple effector cell populations, including Th2 cells, group 2 innate lymphoid cells (ILC2s), and type 2 cytotoxic T (Tc2) cells. Epithelial-derived alarmins, particularly interleukin-33 (IL-33) and thymic stromal lymphopoietin (TSLP), function as key upstream initiators that bridge innate and adaptive …

Granulocyte development, tissue recruitment, and function during allergic inflammation.

Granulocytes provide a fast innate response to pathogens and allergens. In allergy and anti-helminth immunity, epithelial cells of damaged barriers release alarmins like IL-25, IL-33, and thymic stromal lymphopoietin (TSLP) but also chemokines like CXCL1 or CCL11 to promote cell recruitment and inflammation. In addition, mast cells positioned at barrier tissue sites also quickly release mediators upon specifically sensing antigens through IgE bound to FcεR1 on their surface. Released mediators induce the recruitment of different granulocytes in a timely ordered manner. First, neutrophils extra…

Polyethylene tube as an attachment to universal serial bus (USB) dermatoscope for preventing cross-infection.

Type 2 immunity has evolved to protect against worms but becomes harmful when activated during allergic inflammation. Group 2 innate lymphoid cells (ILC2s) drive type 2 responses by rapidly secreting IL-5 and IL-13. The alarmins, IL-25, IL-33, and TSLP activate ILC2s and are linked to allergic diseases. However, how alarmins connect to the transcriptional networks driving type 2 effector functions remains elusive. Here, we performed RNA sequencing of ILC2s deficient in IL-25, IL-33, or TSLP pathway and identified the transcription factor Blimp-1 as an IL-33-regulated gene in ILC2s. While Blimp…

Synergistic TCR-independent action of IL-33 and IL-12 drive a potent IFNγ secretory program in human circulating MAIT cells with immunomodulatory properties.

INTRODUCTION: Mucosal-Associated Invariant T (MAIT) cells are a subset of unconventional T cells that rapidly respond to early signs of inflammation, infection, and tissue damage. While MAIT cells have been typically associated with microbial infections, given their ability to respond to both microbial-derived riboflavin metabolites and proinflammatory cytokines such as IL-12, IL-15, or IL-18, their role in other inflammatory conditions remains mostly unknown. Alarmins, including IL-25, IL-33, and thymic stromal lymphopoietin (TSLP), are crucial effectors for early inflammatory responses, bein…
