Copper-Mediated Radical Cyclopropanation of Activated Alkenes under Continuous-Flow Conditions.

Polysubstituted cyclopropanes are valuable in complex molecule synthesis but challenging to access due to low atom economy and harsh conditions. Herein, we report a copper-mediated, peroxide-initiated cyclopropanation of activated alkenes with active methylene compounds via a radical pathway. This diazo-free, prefunctionalization-free method, integrated with continuous flow, enhances safety, scalability, and efficiency. Operable under mild conditions with broad substrate scope, it offers a practical and atom-economical route to diverse polysubstituted cyclopropanes.

Cyano-Fluorosulfonylation of Unactivated Alkenes by Photoredox and Copper Dual Catalysis.

Both fluorosulfonyl and cyano groups are important structural motifs in bioactive molecules. Herein, we report a new difunctionalization reaction of alkenes based on fluorosulfonyl radicals, which allows for the introduction of the fluorosulfonyl and cyano groups into unactivated alkenes in one step. This transformation is enabled by merging photoredox and copper catalysis, featuring visible light catalysis, mild conditions, and good functional group tolerance. Further transformation of products via SuFEx reactions is also demonstrated.

Intermolecular Anti-Markovnikov Hydroamination of Alkenes with Sulfonamides, Sulfamides, and Sulfamates.

A general method for the light-driven intermolecular anti-Markovnikov hydroamination of alkenes with primary sulfonamides, sulfamides, and sulfamates is presented. The reaction is mediated by a ternary catalyst system composed of an iridium(III) chromophore, a fluorinated alkoxide base, and a thiol H-atom donor. We hypothesize that the reactions proceed through a proton coupled electron-transfer (PCET) mechanism wherein implementation of the alkoxide base imparts additional thermochemical driving force for the homolytic activation of strong N-H bonds that were previously inaccessible using thi

Visible-Light-Photocatalyzed Dicarbofunctionalization of Conjugated Alkenes with Ketone-Based Dihydroquinazolinones.

A visible-light-photocatalyzed 1,2-arylalkylation of N-(arylsulfonyl)acrylamides with ketone-based dihydroquinazolinones is described. The formal C-C bond cleavage of aliphatic ketones is unified with tandem radical alkylation/1,4-aryl migration/desulfonylation to forge two different types of vicinal C-C bonds and construct an all-carbon quaternary α-stereocenter, thus enhancing the carbogenic complexity and tolerating diverse functionalities. Additional to telescopic synthesis and product diversification, this method features a radical dicarbofunctionalization of conjugated N-(arylsulfonyl)ac

Defluorinative Alkylboration of Alkenes Enabled by Dual Photoredox and Copper Catalysis.

A regioselectivity reversed three-component defluorinative alkylboration of alkenes with trifluoromethyls and bis(pinacolato)diboron via dual photoredox/copper catalysis is reported. The mild conditions are compatible with a wide array of nonactivated trifluoromethyl aromatics bearing electron-donating or electron-neutral substituents, trifluoroacetamides, and various nonactivated terminal and internal alkenes, enabling straightforward access to synthetically valuable γ-gem-difluoroalkyl boronates with high efficiency. Furthermore, this protocol is applicable to alkene-tethered trifluoromethyl

Water-mediated radical C-H tosylation of alkenes with tosyl cyanide.

The water-mediated tosylation of alkenes with tosyl cyanide was discovered. Experimental investigations revealed that the reaction was initiated by the in situ formation of sulfinyl sulfone in the presence of water. The sulfinyl sulfone species decomposed to a sulfonyl radical and a sulfinyl radical through homolytic fission. The vinyl sulfone was afforded via sequential addition of the alkene to the sulfonyl radical and the sulfinyl radical, followed by β-elimination of a sulfinyl moiety.

Electrophilic Sulfur Reagent Design Enables Directed syn-Carbosulfenylation of Unactivated Alkenes.

A multi-component approach to structurally complex organosulfur products is described via the nickel-catalyzed 1,2-carbosulfenylation of unactivated alkenes with organoboron nucleophiles and tailored organosulfur electrophiles. The key to the development of this transformation is the identification of a modular N-alkyl-N-(arylsulfenyl)arenesulfonamide family of sulfur electrophiles. Tuning the electronic and steric properties of the leaving group in these reagents controls pathway selectivity, favoring three-component coupling and suppressing side reactions, as examined via computational studi

Cobalt-Catalyzed Markovnikov-Selective Radical Hydroacylation of Unactivated Alkenes with Acylphosphonates.

Acylphosphonates having the 5,5-dimethyl-1,3,2-dioxophosphinanyl skeleton are developed as efficient intermolecular radical acylation reagents, which enable the cobalt-catalyzed Markovnikov hydroacylation of unactivated alkenes at room temperature under mild conditions. The protocol exhibits broad substrate scope and wide functional group compatibility, providing branched ketones in satisfactory yields. A mechanism involving the Co-H mediated hydrogen atom transfer and subsequent trapping of alkyl radicals by acylphosphonates is proposed.

Phosphonodifluoromethyl and phosphonothiodifluoromethyl radicals. Generation and addition onto alkenes and alkynes.

[reaction: see text] Selanylated difluoromethylphosphonates and difluoromethylphosphonothioates are good precursors to phosphonodifluoromethyl and phosphonothiodifluoromethyl radicals, respectively. When generated in the presence of alkenes and a hydrogen donor, the corresponding alpha,alpha-difluorinated alkylphosphonates or alkylphosphonothioates are produced in fair to good yields. The use of alkynes results in the formation of alpha,alpha-difluorinated allyl derivatives in useful yields. The presence of the sulfur atom in phosphonothiodifluoromethyl radicals usually translates into higher

Electrochemical Oxidation-Induced Difunctionalization of the Sulindac Alkene Moiety with Diselenides.

The structural modification of drug molecules is a fundamental approach to optimizing lead compounds designed to achieve concurrent improvements in efficacy, pharmacokinetics, and safety. Herein, we disclose an efficient and eco-benign electrochemical oxidative protocol for regioselective and chemoselective difunctionalization of the sulindac alkene scaffold under transition-metal-free, exogenous oxidant-free, and base-free conditions. This methodology delivers diverse sulindac derivatives in moderate to excellent yields (up to 92%) with a broad functional group tolerance. Mechanistic studies

A catalytic alkene insertion approach to bicyclo[2.1.1]hexane bioisosteres.

C(sp3)-rich bicyclic hydrocarbon scaffolds, as exemplified by bicyclo[1.1.1]pentanes, play an increasingly high-profile role as saturated bioisosteres of benzenoids in medicinal chemistry and crop science. Substituted bicyclo[2.1.1]hexanes (BCHs) are emerging bicyclic hydrocarbon bioisosteres for ortho- and meta-substituted benzenes, but are difficult to access. Therefore, a general synthetic route to BCHs is needed if their potential as bioisosteres is to be realized. Here we describe a broadly applicable catalytic approach that delivers substituted BCHs by intermolecular coupling between ole

Copper-Catalyzed Bifunctionalization/Annulation of Unactivated Alkene with Alkyl Bromides.

β-Lactam is a ubiquitous scaffold in bioactive compounds and pharmaceuticals. Herein, we disclose a streamlined method for the construction of β-lactams starting from unactivated alkenes and alkyl bromides via a Cu-catalyzed inter-/intramolecular carboamidation. This reaction proceeded smoothly under mild reaction conditions and exhibited a broad substrate scope and various functional groups. This protocol is not only compatible with 1, 2, and 3° alkyl bromides but also suitable for α-bromo nitrile as well as various benzyl bromides. The mechanism exploration indicated that sequential radical

Oxidative Cleavage of Alkenes by O2 with a Non-Heme Manganese Catalyst.

The oxidative cleavage of C═C double bonds with molecular oxygen to produce carbonyl compounds is an important transformation in chemical and pharmaceutical synthesis. In nature, enzymes containing the first-row transition metals, particularly heme and non-heme iron-dependent enzymes, readily activate O2 and oxidatively cleave C═C bonds with exquisite precision under ambient conditions. The reaction remains challenging for synthetic chemists, however. There are only a small number of known synthetic metal catalysts that allow for the oxidative cleavage of alkenes at an atmospheric pressure of

Reductive Rearrangement of Tetraphenyldiphosphine Disulfide To Trigger the Bisthiophosphinylation of Alkenes and Alkynes.

The facile synthesis of organophosphorus compounds is of great importance for the development of new synthetic methods by using air-stable sources of phosphorus. In this respect, a synthetic method that is based on a reductive rearrangement and is capable of converting air-stable pentavalent phosphorus compounds into reactive trivalent phosphorus compounds is a powerful tool. Tetraphenyldiphosphine disulfide, which is a shelf-stable solid, was the focus of this study, and it was shown to undergo reductive rearrangement to trigger the bisthiophosphinylation of a variety of alkenes, such as term

Phosphine-alkene ligands as mechanistic probes in the Pauson-Khand reaction.

An alkyne tetracarbonyl dicobalt complex with a chelated phosphine-alkene ligand, in which the phosphorus atom and the alkene from the ligand are attached to the same cobalt atom has been prepared, isolated, and characterized by X-ray crystallography. The complex serves as a mechanistic model for an intermediate of the Pauson-Khand (PK) reaction. Although the alkene fragment is located in an equatorial coordination site with an appropriate orientation, and, therefore, should undergo insertion, it failed to give the PK product upon either thermal or N-methylmorpholine N-oxide activation. Chronic toxicity of a mixture of chlorinated alkanes and alkenes in ICR mice.

The aim of this study was to determine the chronic toxicity of a mixture of chlorinated alkanes and alkenes (CA) consisting of chloroform, 1,1-dichloroethane, 1,1-dichloroethylene, 1,1,1-trichloroethane, trichloroethylene, and tetrachloroethylene. These chlorinated organic solvents were present in the underground water near an electronic appliances manufactory in Taoyuan, Taiwan. Male and female weanling ICR mice were treated with low-, medium-, and high-dose CA mixtures in drinking water for 16 and 18 mo, respectively. A significant number of male mice treated with the high-dose CA mixture de

Oxidation of gaseous and volatile hydrocarbons by selected alkene-utilizing bacteria.

Eleven strains of alkene-utilizing bacteria belonging to the genera Mycobacterium, Nocardia, and Xanthobacter were tested for their ability to grow with C(1) to C(6) alkanes, C(2) to C(6) alkenes, alkadienes, and monoterpenes furnished individually as sole sources of carbon and energy in a mineral salts medium. A limited number of alkenes and alkanes supported growth of the bacteria; some bacteria were unable to grow on any of the saturated hydrocarbons tested. Monoterpenes were frequently used as carbon and energy sources by alkene-utilizing bacteria belonging to the genera Mycobacterium and

Solid acids as cocatalysts in the chelation-assisted hydroacylation of alkenes and alkynes.

The use of homogeneous Brønsted acid cocatalysts (such as benzoic acid) in hydroacylation reactions via imine intermediates has been extensively studied. However, the use of heterogeneous cocatalysts has been limited to montmorillonite K10. Thus, we can use other solid acids to increase the efficiency of the reaction. In this study, we describe the effects of sulfated zirconia, Al-MCM-41 or superacid modified montmorillonite on the hydroacylation of alkenes and alkynes with aldehydes via imine intermediates and in the presence of the Wilkinson complex. Furthermore, we addressed the dual role o

Synthesis of Tetrahydrofurans and Pyrrolidines by Copper-Catalyzed Oxy/Aminoarylation of Alkenes.

An efficient copper-catalyzed inter/intramolecular oxy/aminoarylation of γ-hydroxy/aminoalkenes with diaryliodonium triflates is reported. Simple activation of these arylating agents with copper(II) triflate in dichloromethane triggers a smooth activation of the alkene, which is simultaneously trapped by the internal nucleophile, yielding, depending upon its nature, a range of highly substituted tetrahydrofurans and pyrrolidines. The cyclization was moreover found to be stereospecific, with diastereoisomeric alkenes yielding diastereoisomers of the cyclized product, and could be extended to ox
