Expression patterns of sterol transporters NPC1 and NPC2 in the cnidarian-dinoflagellate symbiosis.

The symbiotic interaction between cnidarians (e.g., corals and sea anemones) and photosynthetic dinoflagellates of the genus Symbiodinium is triggered by both host-symbiont recognition processes and metabolic exchange between the 2 partners. The molecular communication is crucial for homeostatic regulation of the symbiosis, both under normal conditions and during stresses that further lead to symbiosis collapse. It is therefore important to identify and fully characterise the key players of this intimate interaction at the symbiotic interface. In this study, we determined the cellular and subc

Differences in the binding sites of two site-3 sodium channel toxins.

Site-3 toxins from scorpion and sea anemone bind to Na channels and selectively inhibit current decay. Anthopleurins A and B (ApA and ApB, respectively), toxins found in the venom of the sea anemone Anthopleura xanthogrammica, bind to closed states of mammalian skeletal and cardiac Na channels with differing affinities which arise from differences in first-order toxin/channel dissociation rate constants, koff. Using chimera comprising domain interchanges between channel isoforms, we examined the structural basis of this differential affinity. Toxin/channel association rates, kon, were similar

Putting Warburg to work: how imaging of tumour acidosis could help predict metastatic potential in breast cancer.

Solid tumours are often highly acidic compared to normal tissue, and tumour extracellular acidosis contributes to multiple aspects of cancer progression. Now, Anemone et al. in this issue of the British Journal of Cancer provide in vivo evidence that the degree to which various breast cancer cell lines acidify their environment correlates with their ability to metastasise to the lungs. This indicates that measurements of tumour extracellular acidosis have the potential to become a clinical tool for assessing the risk of metastasis.

Nematostella vectensis exhibits an enhanced molecular stress response upon co-exposure to highly weathered oil and surface UV radiation.

Crude oil released into the environment undergoes weathering processes that gradually change its composition and toxicity. Co-exposure to petroleum mixtures and other stressors, including ultraviolet (UV) radiation, may lead to synergistic effects and increased toxicity. Laboratory studies should consider these factors when testing the effects of oil exposure on aquatic organisms. Here, we study transcriptomic responses of the estuarine sea anemone Nematostella vectensis to naturally weathered oil, with or without co-exposure to environmental levels of UV radiation. We find that co-exposure gr

Computational Studies of Venom Peptides Targeting Potassium Channels.

Small peptides isolated from the venom of animals are potential scaffolds for ion channel drug discovery. This review article mainly focuses on the computational studies that have advanced our understanding of how various toxins interfere with the function of K⁺ channels. We introduce the computational tools available for the study of toxin-channel interactions. We then discuss how these computational tools have been fruitfully applied to elucidate the mechanisms of action of a wide range of venom peptides from scorpions, spiders, and sea anemone.

Associations between fishes (Actinopterygii: Teleostei) and anthozoans (Anthozoa: Hexacorallia) in epipelagic waters based on in situ records.

We formally describe the association of fishes and anthozoans in epipelagic waters, extending this relationship to beyond the benthos. In situ observations and photographs of Aluterus schoepfii, Ariomma regulus, Caranx cf. latus and Brama spp. swimming alongside or holding larval tube anemones (Cerianthidae and Arachnactidae) and larval zoanthids (Sphenopidae) were made during blackwater SCUBA dives off Palm Beach, Florida, USA, and off Punaauia, Tahiti, French Polynesia. We report and illustrate the behaviour of these interactions, and suggest an advantage for the anthozoans.

Interactions between benthic predators and zooplanktonic prey are affected by turbulent waves.

Predators capture prey in complex and variable environments. In the ocean, bottom-dwelling (benthic) organisms are subjected to water currents, waves, and turbulent eddies. For benthic predators that feed on small animals carried in the water (zooplankton), flow not only delivers prey, but can also shape predator-prey interactions. Benthic passive suspension feeders collect prey delivered by movement of ambient water onto capture-surfaces, whereas motile benthic predators, such as burrow-dwelling fish, dart out to catch passing zooplankton. How does the flow of ambient water affect these contr

Cnidarian Venoms in the Omics Era: Toxin Discovery and Bioactive Potential.

Cnidaria is a phylum of aquatic invertebrates that includes jellyfish, sea anemones and corals. Their venoms are among the oldest in the animal kingdom and an important source of bioactive molecules. Advances in omics technologies have revolutionized toxin discovery, yet knowledge of cnidarian toxin diversity remains fragmented across databases and individual studies. To address this gap, this review integrates information from 342 toxins curated in the ToxProt database, 76 additional toxins reported in the literature, and evidence from 81 proteomic and 31 transcriptomic studies. Toxins have b

Toxin III from Leiurus quinquestriatus quinquestriatus: a specific probe for receptor site 3 on insect sodium channels.

Scorpion toxin Lqq III binds to a single class of high affinity (Kd = 72 +/- 19 pM) and low capacity (Bmax = 2.5 +/- 0.2 pmol/mg) binding sites in cockroach neuronal membranes. Its binding was inhibited by Lqh alpha IT (IC50 = 80 +/- 30 pM) and sea-anemone toxin ATX II (IC50 = 2.5 +/- 0.3 nM), suggesting that Lqq III is a specific probe for receptor site 3 on cockroach sodium channels. This was confirmed by competitive binding experiments between 125I-Lqq III and scorpion alpha-toxins which have less toxicity in insects.

Ultraviolet radiation significantly enhances the molecular response to dispersant and sweet crude oil exposure in Nematostella vectensis.

Estuarine organisms are subjected to combinations of anthropogenic and natural stressors, which together can reduce an organisms' ability to respond to either stress or can potentiate or synergize the cellular impacts for individual stressors. Nematostella vectensis (starlet sea anemone) is a useful model for investigating novel and evolutionarily conserved cellular and molecular responses to environmental stress. Using RNA-seq, we assessed global changes in gene expression in Nematostella in response to dispersant and/or sweet crude oil exposure alone or combined with ultraviolet radiation (U

A new species of Aspidophryxus G.O. Sars, 1883 (Crustacea, Isopoda, Dajidae) from Caribbean mysid shrimp.

A new species of dajid isopod in the genus Aspidophryxus G.O. Sars, 1883 is described from three females and one male found infesting the lateral carapace of Heteromysis (Olivemysis) actiniae Clarke, 1955 (Mysidae: Heteromysinae) associated with the corkscrew anemone Bartholomea annulata (Lesuer, 1817) (Anthozoa: Aiptasiidae) from the British Virgin Islands. This is the first record of the genus from the western Atlantic; three other species are known from northern European waters, the central Atlantic and Japan, all on mysid hosts. The new species is the first dajid recorded from tropical Car

Serine protease inhibitors containing a Kunitz domain: their role in modulation of host inflammatory responses and parasite survival.

Proteins containing a Kunitz domain have the typical serine protease inhibition function ranging from sea anemone to man. Protease inhibitors play major roles in infection, inflammation disorders and cancer. This review discusses the role of serine proteases containing a Kunitz domain in immunomodulation induced by helminth parasites. Helminth parasites are associated with protection from inflammatory conditions. Therefore, interest has raised whether worm parasites or their products hold potential as drugs for treatment of immunological disorders. Finally, we also propose the use of recombina

Chlamydiae in corals: shared functional potential despite broad taxonomic diversity.

Cnidarians, such as corals and sea anemones, associate with a wide range of bacteria that have essential functions, including nutrient cycling and the production of antimicrobial compounds. Within cnidarians, bacteria can colonize all microhabitats including the tissues. Among them are obligate intracellular bacteria of the phylum Chlamydiota (chlamydiae) whose impact on cnidarian hosts and holobionts, especially corals, remain unknown. Here, we conducted a meta-analysis of previously published 16S rRNA gene metabarcoding data from cnidarians (e.g. coral, jellyfish, and anemones), eight metage

Symbiosis-induced adaptation to oxidative stress.

Cnidarians in symbiosis with photosynthetic protists must withstand daily hyperoxic/anoxic transitions within their host cells. Comparative studies between symbiotic (Anemonia viridis) and non-symbiotic (Actinia schmidti) sea anemones show striking differences in their response to oxidative stress. First, the basal expression of SOD is very different. Symbiotic animal cells have a higher isoform diversity (number and classes) and a higher activity than the non-symbiotic cells. Second, the symbiotic animal cells of A. viridis also maintain unaltered basal values for cellular damage when exposed

The structure of versutoxin (delta-atracotoxin-Hv1) provides insights into the binding of site 3 neurotoxins to the voltage-gated sodium channel.

BACKGROUND: Versutoxin (delta-ACTX-Hv1) is the major component of the venom of the Australian Blue Mountains funnel web spider, Hadronyche versuta. delta-ACTX-Hv1 produces potentially fatal neurotoxic symptoms in primates by slowing the inactivation of voltage-gated sodium channels; delta-ACTX-Hv1 is therefore a useful tool for studying sodium channel function. We have determined the three-dimensional structure of delta-ACTX-Hv1 as the first step towards understanding the molecular basis of its interaction with these channels. RESULTS: The solution structure of delta-ACTX-Hv1, determined usin

Use of venom peptides to probe ion channel structure and function.

Venoms of snakes, scorpions, spiders, insects, sea anemones, and cone snails are complex mixtures of mostly peptides and small proteins that have evolved for prey capture and/or defense. These deadly animals have long fascinated scientists and the public. Early studies isolated lethal components in the search for cures and understanding of their mechanisms of action. Ion channels have emerged as targets for many venom peptides, providing researchers highly selective and potent molecular probes that have proved invaluable in unraveling ion channel structure and function.
