Trans-arterial chemoembolization as a therapy for liver tumours: New clinical developments and suggestions for combination with angiogenesis inhibitors.

The liver is the primary site of metastases for many malignancies. Gastrointestinal cancers are especially prone to spread to the liver and other tumours, as breast cancer and melanoma often spread to the liver. On the other hand, hepatocellular cancer (HCC) is the fifth most common malignancy in the world due to its common etiology from chronic liver damage caused by hepatitis or cirrhosis. Treatments of liver tumours vary according to histology and liver invasion and until now trans-arterial chemoembolization (TACE) has represented a main approach in the therapy of liver tumours. This review

Multitasking Asynchronous Collaborative Nanosystem for Diabetic Wound Healing Based on Hypoglycemic, Antimicrobial, and Angiogenesis-Promoting Effects.

A diabetic foot ulcer (DFU) is a common and serious complication of diabetes. This complication can result in amputation and death because of the several challenges associated with wound healing that can be attributed to the complex wound microenvironment, including biofilm infection, hyperglycemia, and diabetic angiopathy. Existing investigations on the wound-healing rate consider only one or two pathogenic factors, and therefore, despite the extensive research on these pathological microenvironments, there is an urgent need to optimize the wound-healing rate in patients with diabetic foot ul

Role of Hydrogen Sulfide in Chronic Diseases.

In the past, hydrogen sulfide (H2S) was considered as a poisonous gas or waste of the body. Later, researchers found that H2S-producing enzymes exist in mammals. Moreover, their findings indicated that endogenous H2S was associated with the occurrence of many diseases. Therefore, endogenous H2S is able to participate in the regulation of physiological and pathological functions of the body as a gas signaling molecule. In this review, we summarize the regulation mechanism of endogenous H2S on the body, such as proliferation, apoptosis, migration, angiogenesis, as well as vasodilation/vasoconstr

Abnormal angiogenesis in blood outgrowth endothelial cells derived from von Willebrand disease patients.

: Bleeding associated with angiodysplasia is a common, often intractable complication in patients with von Willebrand disease (VWD). von Willebrand factor (VWF), the protein deficient or defective in VWD, is a negative regulator of angiogenesis, which may explain the pathologic blood vessel growth in VWD. This study explores the normal range of angiogenesis in blood outgrowth endothelial cells (BOECs) derived from healthy donors and compares this to angiogenesis in BOECs from VWD patients of all types and subtypes. BOECs were assessed for VWF and angiopoietin-2 (Ang-2) gene expression, secreti

Nanotechnology and nanotoxicology in retinopathy.

Nanoparticles are nanometer-scaled particles, and can be utilized in the form of nanocapsules, nanoconjugates, or nanoparticles themselves for the treatment of retinopathy, including angiogensis-related blindness, retinal degeneration, and uveitis. They are thought to improve the bioavailability in the retina and the permeability of therapeutic molecules across the barriers of the eye, such as the cornea, conjunctiva, and especially, blood-retinal barriers (BRBs). However, consisting of multiple neuronal cells, the retina can be the target of neuronal toxicity of nanoparticles, in common with

The gonadotropins: tissue-specific angiogenic factors?

The gonadotropins, whose members are human chorionic gonadotropin (hCG), lutenizing hormone (LH) and follicle-stimulating hormone (FSH) are a well characterized hormone family known to regulate reproductive functions in both females and males. Recent studies indicate that they can modulate the vascular system of reproductive organs. It was shown that gonadotropins not only influence the expression of vascular endothelial growth factor (VEGF) and both its receptors VEGFR-1 and -2, but also modulate other ubiquitously expressed angiogenic factors like the angiopoietins and their receptor Tie-2,

Kaposi's sarcoma: a result of the interplay among inflammatory cytokines, angiogenic factors and viral agents.

Kaposi's sarcoma (KS) is an angioproliferative disease occurring in 4 clinic-epidemiologic forms. Although the AIDS-associated KS (AIDS-KS) is the most aggressive, all forms of KS share the same immunological and histopathological features suggesting common etiological and pathogenic factors. Recent data indicate that at least in early stage KS is not a real sarcoma but an angiohyperplastic-inflammatory lesion mediated by inflammatory cytokines and angiogenic factors, that is triggered or amplified by infection with human herpesvirus-8. In addition, the human immunodeficiency virus type-1 Tat

Angiogenesis induction by tumors, embryonic tissues, and lymphocytes.

Although it is well established that tumors can induce endothelial cell mitosis and angiogenesis leading to extensive neovascularization, little is known concerning the spectrum of normal cells or tissues that may have similar inductive capabilities. A variety of developmentally active tissues were tested: mouse embryonic organ rudiments, mouse and rabbit trophoblast and placenta, and blastemal fragments obtained from regnerating deer antlers. Assays for angiogenesis included grafts made to the chorioallantoic membrane of chick embryos and grafts placed intracorneally in adult rabbit eyes. Vir

Acidosis significantly alters immune checkpoint expression profiles of T cells from oesophageal adenocarcinoma patients.

Tumour acidosis contributes to cancer progression by inhibiting anti-tumour immunity. However, the effect of acidosis on anti-tumour T cell phenotypes in oesophageal adenocarcinoma (OAC) is unknown. Therefore, this study investigated the effect of acidosis on anti-tumour T cell profiles and if immune checkpoint blockade (ICB) could enhance anti-tumour T cell immunity under acidosis. Acidic conditions substantially altered immune checkpoint expression profiles of OAC patient-derived T cells, upregulating TIM-3, LAG-3 and CTLA-4. Severe acidosis (pH 5.5) significantly decreased the percentage of

Pathogenesis of AIDS-associated Kaposi's sarcoma.

Kaposi's sarcoma presents the oncologist with a myriad of unanswered questions. What accounts for the genesis, distribution, and natural history of this tumor? How could a tumor with such a singular histologic appearance occur in such diverse clinical circumstances? What accounts for the unusual geographic, ethnic, and demographic features of Kaposi's sarcoma? Clearly, the answers to these questions will involve a multifactorial etiology, and may only be arrived at by methodical, piecemeal dissection of each question.

Angiogenic activity of placental tissues of cows.

In Exp. 1, maternal (caruncle) and fetal (cotyledon) portions of the placenta as well as uterine endometrium were obtained from cows at mid-gestation and evaluated for angiogenic activity by placing tissue samples on chick chorioallantoic membranes (CAM). Only caruncular tissues exhibited angiogenic activity in the CAM assay. In Exp. 2, lyophilized homogenates of caruncular tissues obtained from cows at mid-gestation were evaluated for angiogenic activity on CAM and for their ability to stimulate mitosis of bovine aortic endothelial cells in vitro. Homogenates of caruncular tissues again were

Reprogramming T cell fate through antibody-mediated galectin-1 blockade.

Galectins, a family of highly conserved β-galactoside-binding proteins, favor tumor progression by modulating multiple hallmarks of cancer. Galectin-1 (GAL1), a prototype member of this family, plays essential roles in both tumor immunosuppression and angiogenesis through interactions with glycosylated receptors on the surface of immune and vascular cells. Targeting GAL1 inhibits tumor growth and metastasis by enhancing antitumor immunity and preventing aberrant angiogenesis in several experimental models. Therefore, strategies aimed at blocking this multifunctional glycan-binding protein (GBP

Bovine serum albumin-stabilized nano-delivery system potentiates targeted anti-angiogenic therapy and synergistic photo-immunotherapy to restrict lung cancer metastasis.

Tumor angiogenesis plays a crucial role in tumor metastasis as it provides uninterrupted supply of oxygen and nutrients to the rapidly growing tumor cells. The Food and Drug Administration (FDA) recommends that anti-angiogenesis should be used as a supplement along with other tumor treatment modalities, and therefore, anti-angiogenic therapy has recently become an attractive strategy to control tumor metastasis. In our previous work, we reported that glioma-associated oncogene 1 (Gli1) is involved in the angiogenesis of non-small cell lung cancer (NSCLC), and thus could serve as a novel target

Upregulation of RASSF1A in Colon Cancer by Suppression of Angiogenesis Signaling and Akt Activation.

BACKGROUND/AIMS: Silencing of tumor suppressor genes (TSGs) and promotion of angiogenesis are associated with tumor development and metastasis. However, little is known if angiogenic molecules directly control TSGs and vice versa. METHODS: A regulatory link between angiogenesis and down regulation of TSGs was evaluated using an anti-cancer agent, andrographolide (AGP) in cancer cells, mouse xenograft tissues and patient derived organoids through gene/protein expression, gene silencing, and immunohistochemical analyses. RESULTS: AGP treatment demonstrated significant expression of RASSF1A and

A short peptide derived from pigment epithelial-derived factor exhibits an angioinhibitory effect.

BACKGROUND: Pigment epithelial-derived factor (PEDF), a 50 kDa secreted glycoprotein, exhibits distinct effects on a range of cell types. PEDF has been shown to inhibit vascular endothelial growth factor (VEGF)-mediated angiogenesis and widely accepted as a promising agent for treatment eye diseases related to neovascularization. A pool of short peptide fragments derived from PEDF reportedly manifests angioinhibitory activity. This study aims to determine the minimal PEDF fragment which can exert the anti-VEGF effect. METHODS: A series of shorter synthetic peptides, derived from the 34-mer (P

Research progress on the hallmarks of radiation-induced intestinal injury: Mechanisms, biomarkers and therapeutic targets.

Radiation-induced intestinal injury (RIII) is a major factor limiting the dose in abdominal and pelvic radiotherapy. This review summarizes our current understanding of RIII, focusing on its mechanisms, biomarkers, and potential treatment targets. Radiation directly induces DNA damage in intestinal epithelial cells and contributes to excessive reactive oxygen species (ROS), which together lead to increased apoptosis and decreased regeneration of intestinal stem cells (ISCs). An impaired antioxidant system also contributes to excessive ROS levels. Damage to ISCs results in decreased mucus produ

Targeting neural precursors in the adult brain rescues injured dopamine neurons.

In Parkinson's disease, multiple cell types in many brain regions are afflicted. As a consequence, a therapeutic strategy that activates a general neuroprotective response may be valuable. We have previously shown that Notch ligands support neural precursor cells in vitro and in vivo. Here we show that neural precursors express the angiopoietin receptor Tie2 and that injections of angiopoietin2 activate precursors in the adult brain. Signaling downstream of Tie2 and the Notch receptor regulate blood vessel formation. In the adult brain, angiopoietin2 and the Notch ligand Dll4 activate neural p

[Neovascularisation of the cornea].

UNLABELLED: The penetration of blood vessels in the cornea causes a reduction of transparency and as a result reduced vision or even blindness. MATERIALS AND METHODS: Experiments were conducted on 120 New Zealand albino rabbits, separated into different groups. Doses of vasculogenic alkalis provoked burning of the corneas. The following methods were used: Split-lamp investigation of the cornea with fluorescein, fluorescein angiography of the corneal neovessels, light and electron microscopy. Left eyes served as a control. Right eyes were treated with heparin and methylprednisolone, cobalt dic
