Dynamic modulation of ANO1/TMEM16A HCO3(-) permeability by Ca2+/calmodulin.

Anoctamin 1 (ANO1)/transmembrane protein 16A (TMEM16A) is a calcium-activated anion channel that may play a role in HCO(3)(-) secretion in epithelial cells. Here, we report that the anion selectivity of ANO1 is dynamically regulated by the Ca(2+)/calmodulin complex. Whole-cell current measurements in HEK 293T cells indicated that ANO1 becomes highly permeable to HCO(3)(-) at high [Ca(2+)](i). Interestingly, this result was not observed in excised patches, indicating the involvement of cytosolic factors in this process. Further studies revealed that the direct association between ANO1 and calmo

Intrafamilial phenotypic variability in DYT-ANO3: Video documentation of 16 affected members from an Indian family.

DYT-ANO3 is an autosomal dominant dystonia syndrome caused by pathogenic variants in the ANO3 gene, typically presenting as focal or segmental dystonia of the neck and upper limbs, often accompanied by tremor. However, the clinical spectrum has broadened to include a variety of movement phenotypes. Here, we report a large Indian family harbouring a novel ANO3 variant, demonstrating wide phenotypic heterogeneity.

Zafirlukast inhibits the growth of lung adenocarcinoma via inhibiting TMEM16A channel activity.

Lung cancer has the highest mortality among cancers worldwide due to its high incidence and lack of the effective cures. We have previously demonstrated that the membrane ion channel TMEM16A is a potential drug target for the treatment of lung adenocarcinoma and have identified a pocket of inhibitor binding that provides the basis for screening promising new inhibitors. However, conventional drug discovery strategies are lengthy and costly, and the unpredictable side effects lead to a high failure rate in drug development. Therefore, finding new therapeutic directions for already marketed drug

Introduction of a Cys360Tyr Mutation in ANO5 Creates a Mouse Model for Gnathodiaphyseal Dysplasia.

Gnathodiaphyseal dysplasia (GDD) is a rare autosomal dominant genetic disease characterized by the osteosclerosis of tubular bones and the formation of cemento-osseous lesions in mandibles. Although genetic mutations for GDD have been identified in the ANO5/TMEM16E gene, the cellular and molecular mechanisms behind the pathogenesis of GDD remain unclear. Here, we generated the first knock-in mouse model for GDD with the expression of human mutation p.Cys360Tyr in ANO5. Homozygous Ano5 knock-in mice (Ano5KI/KI ) replicated GDD-like skeletal features, including massive jawbones, bowing tibia, bo

Persistent asymptomatic or mild symptomatic hyperCKemia due to mutations in ANO5: the mildest end of the anoctaminopathies spectrum.

BACKGROUND: The ANO5 gene encodes for anoctamin-5, a chloride channel involved in muscle cell membrane repair. Recessive mutations in ANO5 are associated with muscular diseases termed anoctaminopathies, which are characterized by proximal or distal weakness, or isolated hyperCKemia. We present the largest series of patients with asymptomatic/paucisymptomatic anoctaminopathy reported so far, highlighting their clinical and radiological characteristics. METHODS: Twenty subjects were recruited retrospectively from the Neuromuscular Disorders Units database of two national reference centers. All

Orai1 Contributes to Platelet-Activating Factor (PAF)-Induced Contraction in Association with Voltage-Dependent Ca2+ Channel-Mediated Mechanisms in Guinea Pig Urinary Bladder Smooth Muscle.

Platelet-activating factor (PAF) is known to induce smooth muscle contraction; however, its mechanisms in urinary bladder smooth muscle (UBSM) remain unclear. We investigated the involvement of Orai1 in PAF-induced contraction in guinea pig UBSM. PAF (10-6 M)-induced contraction was markedly inhibited by the Orai1 inhibitor Synta66 (10-5 M) and the L-type voltage-dependent Ca2+ channel (VDCC) inhibitor verapamil (10-5 M). Similarly, under Ca2+-free conditions with store depletion induced by cyclopiazonic acid (3 × 10-5 M), Ca2+-induced contraction was suppressed by Synta66 and verapamil. In co

Multiple molecular mimics in Epstein Barr Nuclear Antigen-1, and the pathogenesis of multiple sclerosis.

The Epstein-Barr virus (EBV) infects greater than 95% of humans and is associated with initiating and perpetuating multiple sclerosis (MS). Antibody to Epstein-Barr Nuclear Antigen-1 (EBNA1) is present in nearly 100% of patients with MS before the development of clinical symptoms. Infection with EBV is necessary, but not sufficient, for causation of disease. Within the EBNA1 transcription factor is a stretch of 47 amino acids containing three regions with shared linear sequences of portions of three molecules, Glialcell adhesion molecule (CAM), alpha Crystallin-B, and Anoctamin-2. These cross-

Inhibition of mucus secretion by niclosamide and benzbromarone in airways and intestine.

The Ca2+ activated Cl- channel TMEM16A (anoctamin 1; ANO1) is expressed in secretory epithelial cells of airways and intestine. Previous studies provided evidence for a role of ANO1 in mucus secretion. In the present study we investigated the effects of the two ANO1-inhibitors niclosamide (Niclo) and benzbromarone (Benz) in vitro and in vivo in mouse models for cystic fibrosis (CF) and asthma. In human CF airway epithelial cells (CFBE), Ca2+ increase and activation of ANO1 by adenosine triphosphate (ATP) or ionomycin was strongly inhibited by 200 nM Niclo and 1 µM Benz. In asthmatic mice airwa

A rare case of sudden death due to hemorrhage in a gastrointestinal stromal tumor.

Sudden and unexpected natural death presents a diagnostic challenge in forensic practice, as the underlying cause often remains unclear before death. Acute gastrointestinal hemorrhage due to gastric tumors, such as gastrointestinal stromal tumors (GISTs), is an uncommon cause of sudden death. GISTs are primary mesenchymal tumors of the gastrointestinal tract and most frequently arise in the stomach. To the best of our knowledge, this is the first reported case of sudden and unexpected death due to acute hemorrhage from a GIST. This case report describes a medico-legal autopsy of a 62-year-old

[Transmembrane protein 16A--a new target for the treatment of airway inflammatory diseases].

One of the main pathological features of airway inflammatory diseases is hypersecretion of airway mucus, which is manifested by goblet cell hyperplasia and mucociliary clearance dysfunction. In recent years, it has been found that the molecular structure of calcium activated chloride ion channels, transmenbrane protein 16A(TMEM16A), is closely related to airway mucus hypersecretion.TMEM16A not only mediates ion transepithelial transport and hydration, but also participates in the regulation of mucin secretion. TMEM16A is highly expressed in airway epithelium of a variety of inflammatory diseas

Neuronal regulation of myenteric interstitial cells of Cajal (ICC-MY) in the proximal colon.

Interstitial cells of Cajal (ICC) generate contractile patterns of colonic motility. We investigated innervation of ICC within the plane of the myenteric plexus (ICC-MY) in proximal colon using mice expressing GCaMP6f in ICC. ICC-MY generated localized Ca2+ transients that couple to activation of ANO1 channels, a Ca2+-activated Cl- conductance. ICC are electrically coupled to SMCs, so activation or suppression of currents in ICC affects excitability of SMCs. ICC-MY displayed tonic inhibition, as the neurotoxin, TTX, increased the frequency of Ca2+ transients. Tonic inhibition was mimicked by a

The Tmem16a chloride channel is required for mucin maturation after secretion from goblet-like cells in the Xenopus tropicalis tadpole skin.

The TMEM16A chloride channel is proposed as a therapeutic target in cystic fibrosis, where activation of this ion channel might restore airway surface hydration and mitigate respiratory symptoms. While TMEM16A is associated with increased mucin production under stimulated or pro-inflammatory conditions, its role in baseline mucin production, secretion and/or maturation is less well understood. Here, we use the Xenopus tadpole skin mucociliary surface as a model of human upper airway epithelium to study Tmem16a function in mucus production. We found that Xenopus tropicalis Tmem16a is present at

Genetics and Pathogenesis of Dystonia.

Dystonia is a clinically and genetically highly heterogeneous neurological disorder characterized by abnormal movements and postures caused by involuntary sustained or intermittent muscle contractions. A number of groundbreaking genetic and molecular insights have recently been gained. While they enable genetic testing and counseling, their translation into new therapies is still limited. However, we are beginning to understand shared pathophysiological pathways and molecular mechanisms. It has become clear that dystonia results from a dysfunctional network involving the basal ganglia, cerebel

Pan-Cancer Analysis of ANO6 and Experimental Validation in Metastatic Melanoma.

Anoctamin 6 (ANO6) has been implicated in the oncogenicity of malignancies. However, pan-cancer analysis of ANO6 to fully explore its role in tumors has not been performed and little is reported on its role in melanoma. The ANO6 expression levels, clinical correlation, prognostic significance, mutational profiles, immune infiltration pattern, immune checkpoints, immunomodulatory genes, tumor heterogeneity, and molecular function were explored via systematic bioinformatics analysis and multiple public databases. Subsequently, the biological functions of ANO6 in the pulmonary metastasis of B16F1

The CLCA1/TMEM16A/Cl- current axis associates with H2S deficiency in diabetic kidney injury.

The role played by anionic channels in diabetic kidney disease (DKD) is not known. Chloride channel accessory 1 (CLCA1) facilitates the activity of TMEM16A (Anoctamin-1), a Ca2+-dependent Cl- channel. We examined if CLCA1/TMEM16A had a role in DKD. In mice with type 2 diabetes, renal cortical CLCA1 and TMEM16A content was increased. CLCA1 and TMEM16A content was associated with hydrogen sulfide (H2S) deficiency, mTOR complex 1 (mTORC1) activation, albuminuria, and matrix increase. Administering sodium hydrosulfide (NaHS), a source of H2S, mitigated these changes. In proximal tubular epithelial

ANO1 regulates cardiac fibrosis via ATI-mediated MAPK pathway.

Cardiac fibrosis is associated with most of heart diseases, but its molecular mechanism remains unclear. Anoctamin-1 (ANO1), a calcium-activated chloride channels (CaCCs) protein, plays a critical role in various pathophysiological processes. In the current study, we identified ANO1 expression in myocardial infarction (MI) model of rat and verified the role of ANO1 in cardiac fibrosis using transcriptomics combined with RNAi assays. we found that ANO1 expression was increased during the first two weeks, and decreased in the third week after MI. Fluorescence double labeling showed that ANO1 was

ANO5 mutations in the Polish limb girdle muscular dystrophy patients: Effects on the protein structure.

LGMD2L is a subtype of limb-girdle muscular dystrophy (LGMD), caused by recessive mutations in ANO5, encoding anoctamin-5 (ANO5). We present the analysis of five patients with skeletal muscle weakness for whom heterozygous mutations within ANO5 were identified by whole exome sequencing (WES). Patients varied in the age of the disease onset (from 22 to 38 years) and severity of the morphological and clinical phenotypes. Out of the nine detected mutations one was novel (missense p.Lys132Met, accompanied by p.His841Asp) and one was not yet characterized in the literature (nonsense, p.Trp401Ter, a

Pharmacological profiling of small molecule modulators of the TMEM16A channel and their implications for the control of artery and capillary function.

BACKGROUND AND PURPOSE: TMEM16A chloride channels constitute a depolarising mechanism in arterial smooth muscle cells (SMCs) and contractile cerebral pericytes. TMEM16A pharmacology is incompletely defined. We elucidated the mode of action and selectivity of a recently identified positive allosteric modulator of TMEM16A (PAM_16A) and of a range of TMEM16A inhibitors. We also explore the consequences of selective modulation of TMEM16A activity on arterial and capillary function. EXPERIMENTAL APPROACH: Patch-clamp electrophysiology, isometric tension recordings, live imaging of cerebral cortica

Unraveling the Molecular Basis of the Dystrophic Process in Limb-Girdle Muscular Dystrophy LGMD-R12 by Differential Gene Expression Profiles in Diseased and Healthy Muscles.

Limb-girdle muscular dystrophy R12 (LGMD-R12) is caused by two mutations in anoctamin-5 (ANO5). Our aim was to identify genes and pathways that underlie LGMD-R12 and explain differences in the molecular predisposition and susceptibility between three thigh muscles that are severely (semimembranosus), moderately (vastus lateralis) or mildly (rectus femoris) affected in this disease. We performed transcriptomics on these three muscles in 16 male LGMD-R12 patients and 15 age-matched male controls. Our results showed that LGMD-R12 dystrophic muscle is associated with the expression of genes indica
