Babesia microti in mice. Adoptive transfer of immunity with serum and cells.

Protection against infection with Babesia microti in mice was passively transferred with either serum or cells. Immune spleen cells were more effective than were immune mesenteric lymph node (MLN) cells in reducing parasitaemias in recipient mice, and the level of protection correlated with the donor to recipient ratio rather than with the number of cells transferred. Protection against primary infection in recipient mice was adoptively transferred by nylon wool adherent, B cell enriched subpopulations of spleen cells. In contrast, higher peak parasitaemias were apparent in mice which received

Transmission risk evaluation of transfusion blood containing low-density Babesia microti.

BACKGROUND: Babesia is a unique apicomplexan parasite that specifically invades and proliferates in red blood cells and can be transmitted via blood transfusion, resulting in transfusion-transmitted babesiosis. However, detecting Babesia in blood before transfusion has not received enough attention, and the risk of transfusing blood containing a low density of Babesia microti (B. microti) is unclear, possibly threatening public health and wellness. PURPOSE: This study aimed to determine the lower detection limit of B. microti in blood and to evaluate the transmission risk of blood transfusion

Absence of Anti-Babesia microti antibody in commercial intravenous immunoglobulin (IVIG).

BACKGROUND: Babesiosis is a worldwide emerging protozoan infection that is associated with a spectrum of disease severity from asymptomatic infection to severe organ damage and death. While effective treatment strategies are available, some immunocompromised patients experience severe acute and prolonged/relapsing illness due in part to an impaired host antibody response. Intravenous immunoglobulin (IVIG) has been used as an adjunctive therapy in some immunocompromised babesiosis patients, but its therapeutic effect is uncertain. We evaluated the presence of Babesia microti antibodies in comme

Babesia microti Confers Macrophage-Based Cross-Protective Immunity Against Murine Malaria.

Malaria and babesiosis, the two primary intraerythrocytic protozoan diseases of humans, have been reported in multiple cases of co-infection in endemic regions. As the geographic range and incidence of arthropod-borne infectious diseases is being affected by climate change, co-infection cases with Plasmodium and Babesia are likely to increase. The two parasites have been used in experimental settings, where prior infection with Babesia microti has been shown to protect against fatal malarial infections in mice and primates. However, the immunological mechanisms behind such phenomena of cross-p

Characteristics of transfusion-transmitted Babesia microti, American Red Cross 2010-2017.

BACKGROUND: Babesia microti, a red blood cell (RBC) parasite transmitted naturally to vertebrate hosts by ixodid ticks, is endemic to the northeastern and upper midwestern United States, with the geographic range of infected ticks expanding. B. microti is a blood safety issue with >200 transfusion-transmissions reported. METHODS: The American Red Cross's Hemovigilance program investigated hospital-reported transfusion-transmitted babesiosis (TTB) cases. Follow-up samples from involved donors were tested for B. microti antibodies and parasite DNA, the latter by real-time polymerase chain react

Congenital Babesiosis After Maternal Infection With Borrelia burgdorferi and Babesia microti.

We describe the cases of 2 infants with congenital babesiosis born to mothers with prepartum Lyme disease and subclinical Babesia microti infection. The infants both developed anemia, neutropenia, and thrombocytopenia, and 1 infant required red blood cell transfusion. Both infants recovered with treatment. Additional studies are warranted to define the optimal management strategy for pregnant women with early Lyme disease in geographic areas in which B microti infection is endemic.

Vertical Transmission of Babesia microti in BALB/c Mice: Preliminary Report.

Babesia spp. (Apicomplexa, Piroplasmida) are obligate parasites of many species of mammals, causing a malaria-like infection- babesiosis. Three routes of Babesia infection have been recognized to date. The main route is by a tick bite, the second is via blood transfusion. The third, vertical route of infection is poorly recognized and understood. Our study focused on vertical transmission of B. microti in a well-established mouse model. We assessed the success of this route of infection in BALB/c mice with acute and chronic infections of B. microti. In experimental groups, females were mated o

A polymorphic multigene family encoding an immunodominant protein from Babesia microti.

Human babesiosis in the United States is caused predominantly by Babesia microti, a tick-transmitted blood parasite. Improved testing methods for the detection of infection with this parasite are needed, since asymptomatic B. microti infection represents a potential threat to the blood supply in areas where B. microti is endemic. We performed immunoscreening of an expression library of genomic DNA from a human isolate of B. microti (strain MN1). Among 17 unique immunoreactive clones, we identified 9 which represent a related family of genes with little sequence homology to other known sequence

Effect of carrier erythrocytes containing inositol hexaphosphate on Babesia microti infection.

Erythrocytes containing inositol hexaphosphate (IHP) were administered to mice. Mice were then challenged with Babesia microti. Mice receiving IHP carrier erythrocytes had significantly lower percentages of parasitaemias on days 3, 5 and 7 after infection. Carrier erythrocytes containing IHP have altered P50 oxygen values. Thus, carrier erythrocytes containing IHP may be useful in treating naive animals before transporting into areas endemic for babesiosis.

Macrophage mediated resistance to Babesia microti in Nematospiroides dubius--infected mice.

Infection with the gastrointestinal nematode, Nematospiroides dubius, induced resistance to a concurrent infection with Babesia microti in mice. This enhanced resistance to B. microti occurred during infection with the larval, rather than the adult stages of N. dubius. Splenectomy or the injection of carrageenan or silica abrogated the protective effect against B. microti induced by infection with N. dubius. Peritoneal macrophages harvested from infected mice produced inhibition of the development of B. microti in vitro. This inhibition was greatest using macrophages harvested from mice recove

Identification of Babesia microti immunoreactive antigens by phage display cDNA screen.

Human babesiosis is a malaria-like illness caused by protozoan parasites of the genus Babesia. Babesia microti is responsible for most cases of human babesiosis in the United States, particularly in the Northeast and the Upper Midwest. Babesia microti is primarily transmitted to humans through the bite of infected deer ticks but also through the transfusion of blood components, particularly red blood cells. There is a high risk of severe and even fatal disease in immunocompromised patients. To date, serology testing relies on an indirect immunofluorescence assay that uses the whole Babesia mic

Babesia microti Infection Inhibits Melanoma Growth by Activating Macrophages in Mice.

Babesia microti is an obligate intraerythrocytic protozoan transmitted by an Ixodes tick. Infections caused by protozoa, including Plasmodium yoelii and Toxoplasma gondii, are shown to inhibit tumor development by activating immune responses. Th1 immune response and macrophages not only are essential key factors in Babesia infection control but also play an important role in regulating tumor development. In this study, we investigated the effects of B. microti infection on melanoma in tumor-bearing mice. The results showed that B. microti infection could inhibit the growth of melanoma, signifi

Identification of a thioredoxin reductase from Babesia microti during mammalian infection.

Babesia microti is the primary causative agent of human babesiosis worldwide and associated with increased human health risks and the safety of blood supply. The parasite replicates in the host's red blood cells, thus, in order to counteract the oxidative stress and toxic effects, parasites employ a thioredoxin (Trx) system to maintain a redox balance. Since thioredoxin reductase (TrxR) plays a critical role in the system, in this study, we report the cloning, expression, and functional characterization of a novel TrxR from B. microti (BmiTrxR). The complete gene BmiTrxR was obtained by amplif
