The selection of novel probiotic candidates requires rigorous evaluation of safety and functional properties. This study aimed to assess safety, bacteriocin production and probiotic potential of four bacterial strains isolated from artisanal cheeses: Enterococcus faecium ST02JL and ST03JL, Lactococcus garvieae ST04JL, and Staphylococcus epidermidis ST0409KOC. Experiments were conducted at 37°C and 39°C to simulate physiological and thermal stress conditions. All strains were phenotypically safe, negative for gelatinase, hemolytic activity, and biogenic amine production, featured absence of vir

We examined Treponema denticola-induced dysregulation of toll-like receptor (TLR) signaling and its downstream effects on matrix metalloproteinase 2 (MMP2) expression and activation in periodontal cells, and whether the antimicrobial peptide nisin could modulate this process. T. denticola and its outer membrane protease dentilisin/PrtP activated MMP2, whereas a dentilisin/PrtP-deficient T. denticola mutant did not, and nisin attenuated this response. Dentilisin/PrtP directly activated recombinant MMP2 in vitro. T. denticola-mediated MMP2 activation was TLR2-dependent, since TLR2 suppression ab

Surgical site infections (SSIs) are among the leading healthcare-associated infections worldwide, and S. aureus is the most prevalent cause. Antimicrobial resistance, dormant cells, and biofilm formation contribute to treatment failure in SSIs. Therefore, new therapeutic approaches are needed to fight SSIs. The bacteriocin AP7121 has previously shown in vitro bactericidal and anti-biofilm activity against multi-resistant S. aureus. This study aimed to advance on the characterization of the in vitro activity of AP7121 against dormant forms of methicillin-resistant S. aureus (MRSA) and its effec

Mycobacterium tuberculosis, the etiological agent of human tuberculosis, is an intracellular pathogen responsible for one of the infectious diseases with highest mortality rates. Its ability to replicate inside alveolar macrophages and trigger the formation of granulomas, alongside the appearance of multidrug-resistant strains, impose the employment of drugs that exacerbate their toxic effects after the long therapies necessary to deal with the infection. As an alternative to conventional drugs, this work proposes the use of bacteriocin AS-48 immobilized on biomimetic magnetic nanoparticles (B

Using chromosomal barcoding, we observed that >97% of the Streptococcus pneumoniae (Spn) population turns over in the lung within 2 days post-inoculation in a murine model. This marked collapse of diversity and bacterial turnover was associated with acute inflammation (severe pneumococcal pneumonia), high bacterial numbers in the lungs, bacteremia, and mortality. Intra-strain competition mediated by the blp locus, which expresses bacteriocins in a quorum-sensing-dependent manner, was required for each of these effects. Bacterial turnover from the activity of Blp-bacteriocins increased the rele…

UNLABELLED: Streptococcus pneumoniae is an important human pathogen that normally resides in the human nasopharynx. Competence-mediated bact … umoniae must compete with other bacterial inhabitants of the nasal surface for space and nutrients. Bacteriocins are small antimicrobial peptides produced by bacteria that typically target neighboring bacteria by disruption of the cell surface. S. pnuemoniae encodes a large number of potential bacteriocin, but, for most, their role in competitive interactions …

The current study addresses the critical issue of Listeria monocytogenes growth in raw sausage/meat products leading to human infections, mo … ng and storage of the tested meat/sausage. This inhibitory effect was found to be attributed to the bacteriocins produced by Leuconostoc carnosum DH25 rather than factors like pH or water activity. The stability of the anti-Listeria substances was examined, revealing their resistance to temperature and pH changes, making Leuconostoc carnosum DH25 a promising p …

Leishmaniasis, a category I neglected tropical disease, is a group of diseases caused by the protozoan parasite Leishmania species with a wide range of clinical manifestations. Current treatment options can be highly toxic and expensive, with drug relapse and the emergence of resistance. Bacteriocins, antimicrobial peptides ribosomally produced by bacteria, are a relatively new avenue for potential antiprotozoal drugs. Particular interest has been focused on enterocin AS-48, with previously proven efficacy against protozoan species, including Leishmania spp. Sequential characterization of ente…

In recent years, biocontrol of foodborne pathogens has become a concern in the food industry, owing to safety issues. Listeria monocytogenes is one of the foodborne pathogens that causes listeriosis. The major concern in the control of L. monocytogenes is its viability as it can survive in a wide range of environments. The purpose of this study was to isolate lactic acid bacteria with antimicrobial activity, evaluate their applicability as a cheese starter, and evaluate their inhibitory effects on L. monocytogenes. Lactococcus lactis strain with antibacterial activity was isolated from raw mil

Bacteriocins are emerging as a viable alternative to antibiotics due to their ability to inhibit growth or kill antibiotic resistant pathogens. Herein, we evaluated the ability of the bacteriocin Garvicin KS (GarKS) produced by Lactococcus garvieae KS1546 isolated from cow milk to inhibit the growth of fish and foodborne bacterial pathogens. We found that GarKS inhibited the growth of five fish L. garvieae strains isolated from infected trout and eels. Among fish pathogens, GarKS inhibited the growth of Streptococcus agalactiae serotypes Ia and Ib, and Aeromonas hydrophila but did not inhibit …

Listeriolysin S (LLS) is a thiazole/oxazole-modified microcin (TOMM) produced by hypervirulent clones of Listeria monocytogenes LLS targets specific gram-positive bacteria and modulates the host intestinal microbiota composition. To characterize the mechanism of LLS transfer to target bacteria and its bactericidal function, we first investigated its subcellular distribution in LLS-producer bacteria. Using subcellular fractionation assays, transmission electron microscopy, and single-molecule superresolution microscopy, we identified that LLS remains associated with the bacterial cell membrane

As potential probiotic traits of human milk-isolated bacteria have increasingly been recognized, this study aimed to evaluate the probiotic properties of bacteriocin-producing Enterococcus faecium strains isolated from human milk and colostrum. Among 118 human milk- and colostrum-isolated lactic cocci, only 29 were identified as Enterococcus. Of these, only four Enterococcus faecium isolates exhibited bacteriocigenic activity against several pathogenic Gram-positive bacteria, including Listeria monocytogenes. These isolates exhibited high acid (up to pH 3.0) and bile tolerance (0.5% oxgall) in

Food safety has been an important issue globally due to increasing foodborne diseases and change in food habits. To inactivate foodborne pathogens, various novel technologies such as biopreservation systems have been studied. Bacteriocins are ribosomally synthesized peptides or proteins with antimicrobial activity produced by different groups of bacteria, but the bacteriocins produced by many lactic acid bacteria offer potential applications in food preservation. The use of bacteriocins in the food industry can help reduce the addition of chemical preservatives as well as the intensity of heat…

Many Clostridium species are pathogenic for humans and animals, and most of the resulting diseases, such as tetanus, botulism, gas gangrene and pseudomembranous colitis, are due to the production of potent extracellular toxins. The biochemical mechanisms of action of Clostridium toxins have been extensively studied in the past ten years. However, detailed information about the regulation of toxin gene expression has only recently emerged. TcdR, BotR, TetR and UviA are now known to be related alternative RNA polymerase sigma factors that drive transcription of toxin A and toxin B genes in C. di

Fusobacterium nucleatum is considered for its role in colonization of initial and late microorganisms in dental plaque and for its coaggregation with other bacterial species. It is known that action of different antimicrobial substances may interfere with either virulence factors or with host-bacteria interaction. The goal of this study was to examine the influence of subinhibitory concentrations of chlorhexidine, triclosan, penicillin G and metronidazole on hemolytic activity and bacteriocin-like substance production of oral F. nucleatum. A high resistance to penicillin G was observed and 63%

The monkeypox virus (MPXV) has recently risen to be a significant global health threat and currently there are no approved antiviral agents, which makes it necessary to develop new antiviral strategies. In this study, we employed in silico techniques to investigate whether bacteriocins could be potent inhibitors of the MPXV DNA polymerase (MPDP). At first, the MPXV DNA polymerase enzyme chain was extracted from the Cryo-EM structure of MPXV DNA replication complex and was then assessed using SWISS-MODEL/QMEAN for structural quality. The structures of selected bacteriocins were either retrieved…
