A comparative immunoelectron microscopic study of typical and atypical cases of pemphigoid.

Using an immunoelectron microscopic technique we have compared the ultrastructural localization of IgG and complement (C3) in the basement membrane zone of three typical cases of bullous pemphigoid and three atypical variant cases (nodular, polymorphic and localized pemphigoid, respectively). There was no significant difference in the in vivo ultrastructural localization of these immunoreactants between the typical and the atypical cases of pemphigoid. In all cases IgG and C3 were detected between the basilar surface of the basal keratinocytes and the basal lamina. Moreover, the in vitro bindi

Anatomical basis of a congenital hearing impairment: basilar papilla dysplasia in the Belgian Waterslager canary.

Recent investigations into the anatomy of the inner ear of Belgian Waterslager canaries (BWC) have demonstrated myriad malformations associated with dysgenesis of the pars inferior of the otocyst. In those studies, the surface anatomy of BWC's basilar papilla and sacculus was examined utilizing scanning electron microscopy. In the present investigation, we utilized both light microscopy and transmission electron microscopy to describe the cross sectional anatomical pathology of the BWC's basilar papilla. Examination of the BWC's organ of Corti revealed numerous dysmorphologies: 1) hair cells f

The Spectral Extent of Phasic Suppression of Loudness and Distortion-Product Otoacoustic Emissions by Infrasound and Low-Frequency Tones.

We investigated the effect of a biasing tone close to 5, 15, or 30 Hz on the response to higher-frequency probe tones, behaviorally, and by … e-tone frequency by 8-9 dB/octave, consistent with an apex-to-base gradient of biasing-tone-induced basilar membrane displacement, as we verified by computational simulation. In contrast, the biasing-tone level for criterion loudness suppression increased with probe frequency by only 1-3 dB/octave, reminiscent of previously published data on low-side suppression o …

Melatonin and other serotonin derivatives in the guinea pig membranous cochlea.

In mammals, the indolealkylamine melatonin is synthesized mainly in the pineal gland and to a lesser degree in a number of extrapineal sites. To obtain more information on its distribution, melatonin was measured in the membranous cochlea of the guinea pig. Moreover, melatonin and other serotonin derivatives were determined in organ cultures after incubation with 14C-labelled serotonin. The results show that melatonin is detectable in the organ of Corti and the basilar membrane, and to a lesser degree also in the cochlear nerve and stria vascularis, including the spiral ligament. In vitro stud…

Micro-magnetic resonance imaging of the inner ear in the guinea pig.

We applied a magnetic resonance microscopy at 7.05 T with a gradient coil unit to image the fine structure of the guinea pig cochlea. First, … hlear duct could be clearly distinguished. This allowed a determination of the location of both the basilar membrane and Reissner's membrane. We raise the possibility that MRM may become useful in the diagnosis of endolymphatic hydrops (Meniere's disease).

Mechanically evoked tinnitus after cochlear implantation with preservation of residual hearing.

In the recent past, inclusion criteria for cochlear implant surgery expanded to patients with ever more residual acoustic hearing in the low … al ear is believed to be due to the direct transmission of movements via the electrode array to the basilar membrane of the inner ear. In both cases, the mechanically evoked tinnitus led to revision surgery with immobilization of the array in the mastoid cavity. Despite eliminating the tinnitus, the revision surgery led to a loss of residual hearing in one patient …

Posterior cerebral artery P1 segment at the stoma during endoscopic third ventriculostomy in adults.

OBJECTIVES: A serious catastrophic complication of endoscopic third ventriculostomy (ETV) is basilar artery (BA) damage. Although the BA has been discussed as the cause of haemorrhage and even pseudoaneurysm, variations of the posterior cerebral artery proximal segment (P1) and its protrusion into the third ventricle floor have not been emphasised. A series of cases are reported in which the P1 segment was located at the stoma during an ETV and was at risk of perforation. Precautionary techniques for complication avoidance are described. METHODS: A retrospective review was performed of all ET

Frequency-specific Electrocochleography and Traveling Wave Time as a Clinical Test for Menière's Disease.

INTRODUCTION: Menière's disease is a clinical entity with no definitive objective testing. It has been hypothesized that underlying endolymphatic hydrops stiffens the basilar membrane leading to increased speed of the acoustic stimulus, therefore traveling wave velocity has been proposed as an objective test to aid in the diagnosis. The objective of this study is to compare electrocochleography frequency-specific action potential latency, basilar membrane traveling wave time, and summation to action potential (SP/AP) ratio in Menière's and non-Menière's patients. METHODS: Tympanic electrococh…

Immunohistochemical evaluation of c-erbB-2 oncogene expression in ductal carcinoma in situ and atypical ductal hyperplasia of the breast.

Cases of ductal carcinoma in situ (DCIS) and atypical ductal hyperplasia (ADH) of the breast were examined for expression of the protein product of the c-erbB-2 (neu, HER-2) oncogene using two different polyclonal antibodies via an avidin-biotin immunoperoxidase method on formalin- or Bouin'-fixed, paraffin-embedded tissue. Fifty-five percent (18/33) of DCIS and 10% (2/21) of ADH were positive. Significant c-erbB-2 expression in DCIS was generally divided on histologic grounds: ten of ten comedocarcinomas showed strong membrane staining, while only one of 14 small cell DCIS cases (micropapilla

Sensorineural hearing loss due to cochlear otospongiosis: theoretical considerations of etiology.

Several theories have been advanced to explain the sensorineural hearing loss that occurs in patients with otospongiosis: toxic substances produced by the otospongiotic focus; vascular shunts betwen the inner ear vessels and the otospongiotic focus; and atrophy of the organ of Corti and stria vascularis due to unknown causes. Presented here is yet another theory: impingement upon the cochlear walls by the otospongiotic focus, causing a narrowing of the lumen of the cochlea and distortion of the basilar membrane.

A micromechanical contribution to cochlear tuning and tonotopic organization.

The response properties of hair cells and nerve fibers in the alligator lizard cochlea are frequency selective and tonotopically organized with longitudinal position in the organ. The lengths of the hair-cell hair bundles also vary monotonically with longitudinal position. In this study, quantitative measurements were made of the motion of individual hair bundles in an excised preparation of the cochlea stimulated at auditory frequencies. The angular displacement of hair bundles is frequency selective and tonotopically organized, demonstrating the existence of a micromechanical tuning mechanis

Central nervous system changes associated with noise-induced hearing loss: an electron microscopic and freeze-fracture study of the chick nucleus magnocellularis.

This investigation assessed the effects of noise-induced hearing loss on the avian nucleus magnocellularis, the homologue of the mammalian cochlear nucleus. Fifteen-day-old chicks were exposed to high-intensity broad-band noise (132 dBA) for 24 to 72 hours. Hearing loss was documented with auditory brain-stem response recordings and scanning electron microscopy of the basilar papilla. Nucleus magnocellularis was assessed with thin-section electron microscopy and freeze-fracture techniques. Freeze-fracture preparations of noise-exposed animals showed a decrease in synaptic vesicle fusion sites

Acoustic-reflex activity and behavioral thresholds following exposure to noise.

The relationships between properties of the acoustic reflex and temporary threshold shift (TTS) were examined in eight subjects exposed to a 95 dB SPL, 1.0-kHz octave-band noise for 4 h. The specific pattern of TTS obtained from this exposure was consistent with the expected sensitivity changes along the cochlear partition. Maximum decrease in behavioral thresholds was noted at 1.4 kHz with significant TTS also occurring at 2.0 and 4.0 kHz. Behavioral threshold at 0.5 kHz, one octave below the center frequency of exposure, was not affected. Significant reflex threshold shift (RTS) occurred at

Morphological changes of cerebral arteries in a canine double hemorrhage model.

Cerebral vasospasm is a major cause of morbidity and mortality in patients suffering from subarachnoid hemorrhage (SAH). Despite numerous studies, the pathogenesis of this deadly disorder is not clearly understood. Alterations in endothelial cells are a distinct morphological feature of cerebral vasospasm and some recent studies suggest that apoptosis might play a role in the cells' death. The goal of the present study is to examine the time course of apoptosis in endothelial cells of spastic cerebral arteries following experimental subarachnoid hemorrhage. Fifteen dogs were used in the presen
