Improved constraints on in situ rates and on quantification of complete chloroethene degradation from stable carbon isotope mass balances in groundwater plumes.

Spills of chloroethenes (CEs) at industrial and urban sites can create groundwater plumes in which tetrachloro- and trichloroethene sequentially degrade to dichloroethenes, vinyl chloride (VC) and ethene, or ethane under reducing conditions. For detoxification, degradation must go beyond VC. Assessments based on ethene and ethane, however, are difficult because these products are volatile, may stem from alternative sources, can be further transformed and are not always monitored. To alternatively quantify degradation beyond VC, stable carbon isotope mass balances have been proposed where conce

The carbon isotope ratio of plant organic material reflects temporal and spatial variations in CO2 within tropical forest formations in Trinidad.

A method of monitoring and collecting CO2 samples in the field has been developed which has been used to study both temporal and spatial variations in canopy CO2 isotopic signatures in two contrasting tropical forest formations in Trinidad. These have been related to vertical gradients in the carbon isotope ratio (δ13C) of organic material in conjunction with measurements of other environmental parameters. The δ13C of leaf material from two canopies showed a gradient with respect to height, more negative values being found low in the understorey. The deciduous secondary forest, (Simla) showed

Responsiveness of the Serum Carbon Isotope Ratio to Dietary Added Sugar Reduction in a Randomized Controlled Trial among Youth with Steatotic Liver Disease.

BACKGROUND: The carbon isotope ratio (CIR) has been proposed as a biomarker for added sugar intake, but few studies have been conducted in youth, with data from controlled, feeding studies particularly lacking. OBJECTIVES: This study aims to examine associations of added sugar intake with serum CIR in a randomized, controlled dietary sugar reduction intervention in youth. METHODS: Data were collected from 40 boys (11-16 y) with histologically diagnosed steatotic liver disease who completed a randomized controlled trial and were provided either a low-free sugar diet (<3% of calories from adde

Associations of Food Group Intakes with Serum Carbon Isotope Ratio Values in Youth: Results from 2 Prospective Pediatric Cohort Studies.

BACKGROUND: The carbon isotope ratio (CIR) is a candidate biomarker for sugar-sweetened beverage (SSB) intake in the United States. However, research specific to youth, who differ in their physiology and dietary patterns compared with adults, is lacking. OBJECTIVES: We evaluated longitudinal associations of SSB intakes across childhood/adolescence with serum CIR. We also explored the relationship between other dietary intakes and serum CIR. METHODS: Data were from participants in two longitudinal, pediatric cohorts in Colorado: Exploring Perinatal Outcomes among CHildren (EPOCH) study (visit

Carbon and Acyl Chain Flux during Stress-induced Triglyceride Accumulation by Stable Isotopic Labeling of the Polar Microalga Coccomyxa subellipsoidea C169.

Deriving biofuels and other lipoid products from algae is a promising future technology directly addressing global issues of atmospheric CO2 balance. To better understand the metabolism of triglyceride synthesis in algae, we examined their metabolic origins in the model species, Coccomyxa subellipsoidea C169, using stable isotopic labeling. Labeling patterns arising from [U-13C]glucose, 13CO2, or D2O supplementation were analyzed by GC-MS and/or LC-MS over time courses during nitrogen starvation to address the roles of catabolic carbon recycling, acyl chain redistribution, and de novo fatty ac

A novel high-temperature combustion based system for stable isotope analysis of dissolved organic carbon in aqueous samples. I: development and validation.

RATIONALE: Traditionally, dissolved organic carbon (DOC) stable isotope analysis (SIA) is performed using either offline sample preparation followed by elemental analyzer/isotope ratio mass spectrometry (EA/IRMS) or a wet chemical oxidation (WCO)-based device coupled to an isotope ratio mass spectrometer. The first method is time-consuming and laborious. The second involves the risks of underestimation of DOC concentration and isotopic fractionation due to incomplete oxidation. The development of an analytical method for accurate and sensitive DOC SIA is described in this study. METHODS: A hi

Multi-omics and stable isotopic reveal coordinated carbon-nitrogen metabolic reprogramming sustaining diatom cellular homeostasis under CO2 limitation.

Algal blooms frequently trigger CO2 limitation, under which diatoms not only survive but dominate. However, the cellular metabolic adaptations underlying this ecological phenomenon remain poorly understood. This study systematically investigated the coordinated carbon-nitrogen metabolic response of the typical diatom Skeletonema costatum (S. costatum) to CO2 limitation (300 ppm, 100 ppm) via stable isotope and multi-omics analysis. CO2 limitation triggered the carbon concentrating mechanism (CCM) and attenuated nitrogen isotope fractionation, redirecting carbon flux towards amino acid biosynth

An Improved EA-IRMS Method Using V2O5-Assisted Combustion for Carbon Isotope Analysis of High-Temperature and High-Pressure Metamorphic Graphite.

An improved method was proposed, which addressed the technical challenge of incomplete combustion and conversion of high-temperature, high-pressure metamorphic graphite-a chemically stable material for conventional EA-IRMS method analysis. The method was based on the addition of an efficient oxidant. Using graphite from a Sri Lankan charnockite (G1) and graphite containing pyrite (G2) as test materials, the effects of oxygen injection modes and solid oxidants (CuO and V2O5) on combustion efficiency were evaluated. It was demonstrated that merely increasing oxygen flow rate or duration failed t

Blood Collection Tube has Minimal Interference on the Carbon and Nitrogen Isotope Composition of Plasma, Red Blood Cells, and Serum in Cow Blood.

RATIONALE: Stable isotopes can be useful for tracking diet composition and other physiological processes in ruminants. These techniques rely on the fact that tissues directly reflect the animal's diet. Blood is a dynamic tissue that can be separated into serum, plasma, and red blood cell (RBC) components. Numerous collection tubes for blood are commercially available. No studies have compared the isotopic composition from cattle blood as affected by collection tube. This study compared commercially available serum and plasma tubes and evaluated how they may affect δ13C, δ15N, and C and N conce

Factors controlling the carbon isotope fractionation of tetra- and trichloroethene during reductive dechlorination by Sulfurospirillum ssp. and Desulfitobacterium sp. strain PCE-S.

Carbon stable isotope fractionation of tetrachloroethene (PCE) and trichloroethene (TCE) was investigated during reductive dechlorination. Growing cells of Sulfurospirillum multivorans, Sulfurospirillum halorespirans, or Desulfitobacterium sp. strain PCE-S, the respective crude extracts and the abiotic reaction with cyanocobalamin (vitamin B(12)) were used. Fractionation of TCE (alphaC=1.0132-1.0187) by S. multivorans was more than one order of magnitude higher than values previously observed for tetrachloroethene (PCE) (alphaC=1.00042-1.0017). Similar differences in fractionation were observe

Advancing Primary Direct Method for Absolute 13C/12C Isotope Amount Ratio Measurement of Carbon Dioxide in Air Using Infrared Absorption Spectroscopy.

We present a comprehensive study aimed at advancing the NIST accurate isotope ratio infrared spectroscopy (AIR-IS) system (Fleisher et al., Nat. Phys. 17, 889-893, 2021), as a primary direct method for determining the absolute 13C/12C isotope amount ratio of CO2 in air. The AIR-IS system employs a dual-wavelength cavity ring-down spectrometer to measure the abundance of the two most prevalent isotopologues of carbon dioxide: 12C16O2 and 13C16O2. This study introduces key methodological improvements, including quantification of surface-associated effects, enhancement of laser-to-cavity coupling

Pacific Ocean-wide profile of CYP1A1 expression, stable carbon and nitrogen isotope ratios, and organic contaminant burden in sperm whale skin biopsies.

BACKGROUND: Ocean pollution affects marine organisms and ecosystems as well as humans. The International Oceanographic Commission recommends ocean health monitoring programs to investigate the presence of marine contaminants and the health of threatened species and the use of multiple and early-warning biomarker approaches. OBJECTIVE: We explored the hypothesis that biomarker and contaminant analyses in skin biopsies of the threatened sperm whale (Physeter macrocephalus) could reveal geographical trends in exposure on an oceanwide scale. METHODS: We analyzed cytochrome P450 1A1 (CYP1A1) expr

What is the influence of centennial evolution from macrophytes to algae on organic matter burial in the upper reaches of Yangtze River from the biomarker carbon isotope fingerprint.

Identification and quantification of sediment organic matter (OM) in the plateau eutrophic lakes, which are strongly influenced by human activities, is noticeable in the study of regional carbon (C) burial and global change. The abundance of n-alkanes and compound-specific carbon isotope (δ13Cn-alk) in sediment cores have been widely utilized as proxies. Here, the n-alkane diagnostic pattern, δ13Cn-alk end-member model, ecological data, and the historical OM input, associated with the ecosystem evolutionary processes were investigated. The results show the long-term ecological evolution under

Optimization of high-performance thin-layer chromatography cleanup for compound-specific carbon and hydrogen stable isotope analysis of polycyclic aromatic hydrocarbons in marine sediments.

BACKGROUND: Polycyclic aromatic hydrocarbons (PAHs) are widespread environmental pollutants with carcinogenic potential, commonly accumulating in marine sediments due to their hydrophobic nature. Accurate source identification of PAHs is essential for pollution control but remains challenging in complex matrices because of their low concentrations and the presence of co-eluting substances, such as unresolved complex mixture (UCM). Compound-specific isotope analysis (CSIA) of PAHs offers improved diagnostic resolution; however, it requires highly purified analytes. This study addresses the meth

Dried Blood Spot Sampling of Landlocked Arctic Char (Salvelinus alpinus) for Estimating Mercury Exposure and Stable Carbon Isotope Fingerprinting of Essential Amino Acids.

Dried blood spots (DBS), created by applying and drying a whole blood sample onto filter paper, provide a simple and minimally invasive procedure for collecting, transporting, and storing blood. Because DBS are ideal for use in field and resource-limited settings, we aimed to develop a simple and accurate DBS-based approach for assessing mercury (Hg) exposure and dietary carbon sources for landlocked Arctic char, a sentinel fish species in the Arctic. We collected liquid whole blood (from the caudal vein), muscle, liver, and brains of Arctic char (n = 36) from 8 lakes spanning a Hg gradient in

[Influence of linseed oil and deiterium depleted water on isotopic D/H composition and functional antioxidant defense of the hepatobiliary system in rabbits with carbon tetrachloride intoxication].

The article presents results of studying the influence of linseed oil and drinking diet with a modified isotopic composition with low deuterium content on indicators of prooxidant-antioxidant system during modeling of liver toxicity. The research was performed on 36 rabbits (weighing 3.1-3.5 kg) which were divided into 4 groups. Group 1 consisted of control animals; in group 2, 3 and 4 in rabbits the liver toxicity was modeled by administration of CCl4 (intraperitoneally, in the form of a 50% oil solution, 1 ml per kg bw, 2 times a week for 30 days); nutritional correction using flaxseed oil (

Infant and child diet in Neolithic hunter-fisher-gatherers from Cis-Baikal, Siberia: intra-long bone stable nitrogen and carbon isotope ratios.

Analysis of stable nitrogen and carbon isotopes (δ(15) N and δ(13) C) from subadults and adults allows for assessment of age-related dietary changes, including breastfeeding and weaning, and adoption of an adult diet. In one of the first studies of hunter-fisher-gatherer subadults from Eurasia, three Neolithic (8,800-5,200 calBP) mortuary sites from southwestern Siberia are analyzed to evaluate hypothesized differences in weaning age between Early versus Late Neolithic groups. An intra-individual sampling methodology is used to compare bone formed at different ages. Collagen samples (n = 143) …

Towards elucidation of microbial community metabolic pathways: unravelling the network of carbon sharing in a pollutant-degrading bacterial consortium by immunocapture and isotopic ratio mass spectrometry.

Although much information on metabolic pathways within individual organisms is available, little is known about the pathways operating in natural communities in which extensive sharing of nutritional resources is the rule. In order to analyse such a consortium pathway, we have investigated the flow of 4-chlorosalicylate as carbon substrate within a simple chemostat microbial community using 13C-labelled metabolites and isotopic ratio mass spectrometric analysis of label enrichment in immunocaptured member populations of the community. A complex pathway network of carbon sharing was thereby rev

Foliar stable isotope ratios of carbon and nitrogen in boreal forest plants exposed to long-term pollution from the nickel-copper smelter at Monchegorsk, Russia.

Long-term exposure to primary air pollutants, such as sulphur dioxide (SO2) and nitrogen oxides (NOx), alters the structure and functions of forest ecosystems. Many biochemical and biogeochemical processes discriminate against the heavier isotopes in a mixture; thus, the values of δ13C and δ15N (i.e. the ratio of stable isotopes 13C to 12C and that of 15 N to 14 N, respectively) may give insights into changes in ecosystem processes and identify the immediate drivers of these changes. We studied sources of variation in the δ13C and δ15N values in the foliage of eight boreal forest C3 plants at
