Density gradient ultracentrifugation shows that two molecular forms of acetylcholinesterase (4S and 10S) can be distinguished in the bowels of both normal subjects and Hirschsprung's disease patients. The physicochemical properties of Venezuelan equine encephalomyelitis (VEE) virus and of its ribonucleoprotein (RNP) were studied. Upon purification in a discontinuous or linear sucrose gradient, the losses of infectivity were small (25%), and 96% of cellular proteins were removed. Bovine chromaffin cells were separated by density gradient centrifugation into subfractions. Extracellular vesicles (EVs) serve as key communication signals among embryos. They are secreted into the embryo culture medium and hold potential as biomarkers for assessing embryo quality. However, the limited volume of embryo-conditioned medium presents challenges for EV enrichment. This chapter details an effective method for purifying and characterizing EVs from in vitro embryo cultures by using the OptiPrep™ density gradient ultracentrifugation (ODG) method.
