Osteogenic potential of a chalcone in a critical-size defect in rat calvaria bone.

BACKGROUND: This study describes the bone formation stimulated by the application of a type of chalcone to critical-size defects in rat calvarial bone. MATERIAL AND METHODS: Sixty female Wistar rats were divided into 6 groups of 10 animals per group: control (no treatment), vehicle (vaseline) and the chalcone (1-phenyl-3-(4-chlorophenyl)-2-propen-1-one) suspended in vaseline at 10%. A critical-size defect of 5 mm was prepared using a trephine in the calvarial bone, after which the treatment was applied, in a single dose, according to the experimental group. The samples were evaluated macrosco

Stilbenecarboxylate biosynthesis: a new function in the family of chalcone synthase-related proteins.

Chalcone (CHS), stilbene (STS) synthases, and related proteins are key enzymes in the biosynthesis of many secondary plant products. Precursor feeding studies and mechanistic rationalization suggest that stilbenecarboxylates might also be synthesized by plant type III polyketide synthases; however, the enzyme activity leading to retention of the carboxyl moiety in a stilbene backbone has not yet been demonstrated. Hydrangea macrophylla L. (Garden Hortensia) contains stilbenecarboxylates (hydrangeic acid and lunularic acid) that are derived from 4-coumaroyl and dihydro-4-coumaroyl starter resid

Chalcone-pyridine/pyrimidine hybrids: promising candidates for anticancer drug development.

Cancer remains a global health challenge with high morbidity and mortality, and the clinical application of conventional anticancer agents is severely limited by issues such as poor selectivity, systemic toxicity, and multidrug resistance. Chalcones, pyridines, and pyrimidines are privileged heterocyclic scaffolds in medicinal chemistry, with inherent anticancer potential, structural flexibility, and good biocompatibility. Molecular hybridization, a rational drug design strategy, has been widely applied to construct chalcone pyridine/pyrimidine hybrids by covalently conjugating these bioactive…

Structural Dynamics and Susceptibility of Aminonaphthoquinone-Chalcone Hybrids Against Human Tyrosinase.

Tyrosinase is a key enzyme that catalyzes the rate-limiting step in melanogenesis. Although numerous mushroom-derived tyrosinase inhibitors have been identified, many exhibit limited efficacy against human tyrosinase. In this study, in silico approaches were employed to evaluate the inhibitory potential of aminonaphthoquinone-chalcone hybrids against human tyrosinase. Molecular docking analysis revealed that all 10 hybrids showed higher binding affinity than kojic acid. Among them, six hybrids (3, 4, 5, 7, 9, and 10) displayed higher binding affinity than the others and interacted with amino a

Sonochemical synthesis of chalcone-functionalized SnS and SnS2 with improved sonocatalytic activity.

Herein we present a novel method of functionalization of sonochemically obtained SnS and SnS2. For that purpose, a yellow-orange synthetic dye, chalcone 1,5-bis-(4-dimethylamino-phenyl)-penta-1,4-dien-3-one, was obtained via ultrasound-assisted synthesis based on the Claisen-Schmidt condensation. Then it was applied in the sonochemical synthesis of SnS and SnS2 microparticles using two distinct polar protic solvents - methanol and ethanol. In each of syntheses (regardless the addition of chalcone) nanocrystalline SnS2 phase was identified. In two cases (starting from SnCl2 and using methanol a

Identification of the Acetamide-Chalcone Derivative That Selectively Inhibits ABCG2 Transport Activity.

Multidrug resistance (MDR) remains a major obstacle to effective cancer chemotherapy, often driven by the overexpression of ABC transporters such as ABCG2. In this study, we identified the synthetic chalcone derivative, B5, as a selective ABCG2 inhibitor. Among the 20 chalcone analogs evaluated, B5, bearing an acetamide group at R1 and an ethoxy substituent at R2, was the only compound that produced more than 50% inhibition of ABCG2-mediated Hoechst 33342 transport at 10 μM, while showing no activity against ABCB1. B5 exhibited an IC50 of 2.7 μM for ABCG2 inhibition, consistent with the low-mi

Chalcone Analogue as New Candidate for Selective Detection of α-Synuclein Pathology.

Deposition of α-synuclein (α-syn) aggregates is one of the neuropathological hallmarks of synucleinopathies including Parkinson's disease, dementia with Lewy bodies, and multiple-system atrophy. In vivo detection of α-syn aggregates with SPECT or PET may be an effective tool for medical intervention against synucleinopathy. In the present study, we designed and synthesized a series of chalcone analogues with different aryl groups to evaluate their potential as α-syn imaging probes. In competitive inhibition assays, aryl groups markedly affected binding affinity and selectivity for recombinant

Molecular targets and anticancer activity of quinoline-chalcone hybrids: literature review.

α,β-Unsaturated chalcone moieties and quinoline scaffolds play an important role in medicinal chemistry, especially in the identification and development of potential anticancer agents. The multi-target approach or hybridization is considered as a promising strategy in drug design and discovery. Hybridization may improve the affinity and potency while simultaneously decreasing the resistance and/or side effects. The conjugation of quinolines with chalcones has been a promising approach to the identification of potential anticancer agents. Most of these hybrids showed anticancer activities thro…

Flavonoids and chalcones as selective α-glucosidase inhibitors: from enzymatic screening to validation using the human enzyme.

α-Glucosidase is a key therapeutic target for postprandial glucose control, but most clinically available inhibitors also inhibit α-amylase, leading to gastrointestinal side effects. Natural polyphenols have been widely reported as potent and selective α-glucosidase inhibitors, primarily based on screening assays using yeast enzyme and synthetic substrates. In this study, thirteen flavonoids and five chalcones were initially evaluated using this conventional approach to elucidate the structural determinants of selective α-glucosidase inhibition. Among them, genistein and myricetin emerged as t…

Chalcones and their derivatives as inhibitors of key ABC transporters (ABCB1, ABCG2) to overcome tumor multidrug resistance.

Tumor multidrug resistance (MDR) remains a major cause of chemotherapy failure, primarily resulting from the overexpression of ATP-binding cassette (ABC) transporters, including ABCB1 and ABCG2, which play critical roles in drug resistance. In recent years, chalcones and their derivatives have attracted considerable research interest for reversing MDR due to their distinctive 1,3-diphenyl-2-propen-1-one scaffold, high structural modifiability, and multi-target activities. Numerous studies have demonstrated that chalcone-based compounds could reverse MDR through diverse mechanisms, including di…

Chalcones as Anti-Glioblastoma Stem Cell Agent Alone or as Nanoparticle Formulation Using Carbon Dots as Nanocarrier.

The current prognosis for glioblastoma is dismal. Treatment-resistant glioblastoma stem cells (GSCs) and the failure of most drugs to reach therapeutic levels within the tumor remain formidable obstacles to successful treatment. Chalcones are aromatic ketones demonstrated to reduce malignant properties in cancers including glioblastoma. Nanomedicines can increase drug accumulation and tumor cell death. Carbon-dots are promising nanocarriers that can be easily functionalized with tumor-targeting ligands and anti-cancer drugs. Therefore, we synthesized a series of 4'-amino chalcones with the rat…

O(2)-dependent efficacy of novel piperidine- and piperazine-based chalcones against the human parasite Giardia intestinalis.

Giardia intestinalis is the most frequent protozoan agent of intestinal diseases worldwide. Though commonly regarded as an anaerobic pathogen, it preferentially colonizes the fairly oxygen-rich mucosa of the proximal small intestine. Therefore, when testing new potential antigiardial drugs, O2 should be taken into account, since it also reduces the efficacy of metronidazole, the gold standard drug against giardiasis. In this study, 46 novel chalcones were synthesized by microwave-assisted Claisen-Schmidt condensation, purified, characterized by high-resolution mass spectrometry, (1)H and (13)C…
