After more than 50 years of debates, the role of spatial and temporal gradients during cell chemotaxis is still a contentious matter. One major challenge is that when cells move in response to a heterogeneous chemical environment they are exposed to both spatial and temporal concentration changes. The membrane-associated adhesion molecule JAM-A is required for neutrophil infiltration in inflammatory or ischemic tissues. JAM-A expressed in both endothelial cells and neutrophils has such a role, but the mechanism of action remains elusive. A transient neutrophilic chemotactic deficit was confirmed in a 3 1/2-year-old castrated male dog with recurrent staphylococcal pyoderma accompanied by a persistent eosinophilia and intermittent basophilia. Neutrophilic chemotaxis was quantitatively assessed over the next seven months, and the disappearance of the chemotactic deficit correlated with complete clinical remission of the skin lesions. Aberrations of the complement system were not discovered, and the dog's serum showed increased chemoattractiveness for control neutrophil preparations.

Leukocyte chemotaxis was studied in 11 patients with severe childhood onset atopic dermatitis at a time when their disease was relatively quiescent. Some immunological parameters have been examined in eleven children with juvenile rheumatoid arthritis. A quantitative and functional defect in T lymphocytes has been observed associated with defective chemotaxis of neutrophils. High levels of circulating immune-complexes were detected in sera of 5 out 11 patients. 1 patient showed high levels of antinuclear antibodies and positivity for the rheumatoid factor. Immunological pathogenesis of juvenile rheumatoid arthritis is discussed.

An 11-year-old boy with a life-long history of atopic-like dermatitis and recurrent staphylococcal abscesses was found to have defective neutrophil chemotaxis, impaired-T-lymphocyte functions, hyperimmunoglobulinemia E, and delayed neutrophil bactericidal power. This latter defect has never been found in such patients. The patient's mother revealed a panhypogammaglobulinemia, while his sister and maternal grandmother who had repeated infections were immunologically normal.
