1. Cats were anaesthetized with chloralose and urethane, and ventilated by an artificial intermittent negative pressure applied to the thorax. The carotid body chemoreceptors were isolated and perfused with oxygenated blood. They were stimulated by substituting hypoxic blood obtained from a donor animal.2. Stimulation of the carotid bodies during constant ventilation caused a bradycardia. When an artificial hyperventilation was induced during carotid body stimulation the heart rate increased.3. The increase in heart rate during hyperventilation, and while the carotid bodies were being stimulat…

Effect of i.v. morphine (1-2 mg/kg) on blood pressure changes evoked by sciatic nerve stimulation (SNS) were studied in chloralose anaesthetised cats. SNS gave a depressor, pressor or a biphasic BP response generally linked to the parameters of stimulation. Morphine produced marked attenuation of depressor and some facilitation of pressor response, suggesting a possible reciprocal relationship between depressor and pressor responses. Depressor response has been correlated with deep tissue or visceral pain mediated through A delta fibres and pressor response to cutaneous nociception involving C…

Stimulation of caudal and cranial ends of vagi in vagotomised chloralose-urethane anaesthetised dogs with 7.5V, 30Cps pulses of 2 msec width after 0, 25, 50, 75 and 100 ml/kg body weight infusion dose of Ringer-Locke solution caused well known effects on heart rate, BP, amplitude of peripheral blood pressure waves, amplitude and frequency of respiration and amplitude of ECG waves. The vagal stimulation effects did not appear to be related to volume of fluid infused into circulation except that caudal end stimulation effect on QRS amplitude was prominent during higher infusion doses while its e…

A constant intravenous infusion of digoxin (2 micrograms/kg/min) to alpha-chloralose-anesthetized cats produced a progressive decrease in intraocular pressure with increasing doses of digoxin between 60 micrograms/kg and drug-induced ventricular arrhythmia which occurred at a mean dose of 172 micrograms/kg. Digoxin elicited a 40% decrease in intraocular pressure just prior to ventricular arrhythmia compared to a decrease of only 10% with a control infusion of diluent in the same animal (P less than 0.05). There was no significant difference (P less than 0.1) between changes in blood pressure a…

1. In cats anaesthetized with chloralose whose spinal cords were transected 6-7 weeks previously, spontaneous contractions of the bladder had no effect on mono- or polysynaptic reflexes recorded electrically from the cut lumbar ventral spinal roots.2. In the same cats contractions of the bladder decreased hind-limb extensor and flexor reflexes recorded myographically.3. The effects of (a) pelvic nerve stimulation and distension of the bladder on mono- and polysynaptic reflexes and (b) the effects of bladder contractions on hind-limb reflexes recorded myographically in acute and chronic spinal,…

Nitric oxide (NO)-releasing NSAIDs have been shown to be safer and more potent as antinociceptive and anti-inflammatory agents than their parent compounds. NCX-701 (nitroparacetamol), in contrast to paracetamol, is an effective antinociceptive drug in normal animals but their effectiveness in monoarthritis has not been compared. We have now investigated this question by comparing the antinociceptive effects of i.v. NCX-701 and paracetamol in monoarthritic rats under alpha-chloralose anesthesia. The influence of spinalization on the effects of NCX-701 was also studied. NCX-701 and paracetamol w…

Distension of the descending colon elicits reflex cardiovascular responses, including increases in heart rate and arterial blood pressure. To study the relative contribution of vasoconstriction in individual vascular beds to this reflex response, experiments were performed on seven dogs anaesthetised with chloralose and instrumented with electromagnetic flowmeters around the superior mesenteric, the left renal and the left external iliac arteries. The colorectal portion of the intestine was distended at constant pressure (36.6 mm Hg, 4.9 kPa mean; range 25-50 mm Hg, 3.3-6.7 kPa) with warm Ring…

The inhibitory influence on respiratory functions is clinically an important attribute of the diazepine drugs. This unwanted side effect is … s, using intravenous administration. For all three drugs, a marked toxic interaction was found with chloralose/urethane anesthesia, and noticeably less distinctive with pentobarbital anesthesia. This toxicity increased in quantity with the sequence brotizolam less than triazolam less than flunitrazepam. After repeated injections of equal or increasing doses, …

Small doses of doxapram (less than 0.5 mg . kg-1 iv) were used to study its effect on carotid chemoreceptor activity and ventilation in cats anesthesized with alpha-chloralose. The effects were studied at several levels of partial pressures of O2 and CO2 in arterial blood (PaO2 and PaCO2). It was found that doxapram stimulated discharge rate of the carotid chemoreceptor afferents by the same magnitude at all levels of PaO2 and PaCO2 studied. However, the ventilatory effect of doxapram was more than additive to the concomitant stimulation by hypoxia or hypercapnia. This ventilatory effect was e…

In cats under chloralose-urethan anaesthesia, tachyphylaxis developed to the hypotensive effect of clonidine. Development of this tachyphylaxis was slow in the case of intravenous low doses (10--20 micrograms/kg) and rapid after high doses (50 micrograms/kg). No tachyphylaxis developed to contractions of the nictitating membrane. The phenomenon resembles the tachyphyaxis seen after the application of indirect sympathomimetics. A tachyphylaxis developed to the latter agents inhibited the hypotensive action of clonidine which, after such an intervention, invariably turned into a vasopressor effe…

Volume loading tends to increase blood pressure. Resultant arterial baroreflexes, especially the heart rate component, would counteract renal compensation of volume expansion. In this study, we investigated effect of volume loading on baroreflex vagal bradycardia (BVB) induced by electrical stimulation of the aortic depressor nerves in chloralose/urethane-anesthetized, succinylcholine-immobilized, artificially ventilated rats. As a result, we confirmed that volume loading inhibits BVB. The inhibition was abolished following spinal cord transection at C2. In contrast, volume loading did not aff…

The parameters of the acid-base-state of the arterial blood were measured in spontaneously hypertensive rats of the Okamoto-strain (SHR) and in normotensive Wistar rats (NWR) of a random-bred strain. The animals were anaesthetized with chloralose-urethane and breathed normal air under sea-level conditions. Structure and size of their carotid bodies were studied by light-microscopic methods. When compared with the NWR, the SHR showed a respiratory alkalosis and enlarged carotid bodies. In the SHR, never in the NWR, the lumen of the branches of the glomic arteries was narrowed by pad-like struct…

In cats anesthetized with chloralose-urethane, the central respiratory chemoreceptors were exposed to mock CSF of pH 7.02, 7.02, or 7.57. The right carotid body was simultaneously stimulated by intracarotid injections of 40, 80, or 160 microgram sodium cyanide in 200 microliter Ringer solution. The left carotid nerve and, in some animals, both vagosympathetic truncs were dissected. It could be demonstrated that the increase in ventilation produced by application of NaCN to the peripheral chemoreceptors is significantly larger at high than at low mock CSF pH (i.e. at low than at high central st…

Pharmacological agents (strychnine, picrotoxin, pentobarbital, chloralose, GABA, penicillin, morphine) were used to investigate the nature of the slow potential recorded in the frog olfactory bulb in response to natural stimulation. Three possible hypotheses were tested: 1) The slow potential is neuroglial in nature; 2) it is the analog of the dorsal-root potential of the spinal cord and reflects depolarization of primary afferents arising in the terminals of the olfactory nerve and responsible for presynaptic inhibition in the frog olfactory bulb; 3) the slow potential reflects postsynaptic p…
