Antimicrobial effects of clindamycin-loaded platelet-rich fibrin (PRF).

OBJECTIVES: Recent research has demonstrated that platelet-rich fibrin (PRF) is an appropriate carrier for ampicillin/sulbactam. The aim of the study was to investigate whether PRF is also a suitable bio-carrier for clindamycin (CLI). METHODS: PRF membranes were produced from 36 patients receiving intravenous therapy with CLI (e.g. due to the diagnosis of an osteonecrosis of the jaw or infections). Concentrations of CLI in PRF membranes were measured with liquid chromatography-tandem mass spectrometry, and the antimicrobial effects were investigated in vitro in agar diffusion tests with fresh…

Clindamycin Efficacy for Cutibacterium acnes Shoulder Device-Related Infections.

UNLABELLED: Clindamycin is an antibiotic with high bioavailability and appropriate bone diffusion, often proposed as an alternative in guidelines for C. acnes prosthetic joint infections. We aimed to evaluate the efficacy of clindamycin in the treatment of C. acnes shoulder implant joint infections (SIJI). METHODS: A retrospective analysis was conducted at the University Hospital of Nice (France) between 2010 and 2019. We included patients with one shoulder implant surgical procedure and at least one C. acnes positive sample. We selected the C. acnes SIJI according to French and international…

Teratogenicity of Artemether-Clindamycin Nanostructured Lipid Carriers in Rats.

Currently, artemisinin-based combination therapy is considered the best option in the treatment of malaria. However, toxicity of artemisinin … ant reduction in the dose required to cure animals. Thus, 20% of the therapeutic dose of artemether-clindamycin (ARM-CP) loaded in nanostructured lipid carriers (NLCs; mean particle size 55 ± 10 nm) resulted in complete parasite clearance and 100% survival of infected mice. Here, we investigate the teratogenicity of this formulation in rodents (dosing on alte …

Clindamycin-induced acute kidney injury: large biopsy case series.

BACKGROUND: While clindamycin-induced acute kidney injury (AKI) is uncommon, it has occurred more frequently in recent years. SUMMARY: We investigated 24 patients diagnosed with clindamycin-induced AKI retrospectively. The dosage of clindamycin was 1.0-1.5 g/day. Fifteen patients had episodes of gross hematuria, but fever, skin rash and eosinophilia were rare. Urine analysis revealed mild proteinuria and severe tubular dysfunction. Twenty-three patients were diagnosed with AKI stage 3 upon admission. The clindamycin lymphocyte transformation assay was positive for 63.2% of the patients. Acute…

Suspected clindamycin-associated oesophageal injury in cats: five cases.

The clinical findings, treatment and outcome of suspected clindamycin-associated oesophageal injury in five cats are reported. All cats were treated with one 75 mg clindamycin capsule twice daily (dose range 12-19 mg/kg). Capsules were administered without food or a water bolus. Dysphagia, regurgitation, choking or gagging were seen 3-9 days after starting clindamycin. On oesophagoscopy, three cats had oesophagitis, one of which progressed to stricture formation. Two cats had an oesophageal stricture at first presentation. This is the first report of suspected clindamycin-associated oesophagea…

Clindamycin-resistant Fusobacterium varium bacteremia and decubitus ulcer infection.

Bacteremia due to Fusobacterium spp. is unusual (<10% of cases of anaerobic bacteremia), and the isolation of Fusobacterium varium is especially uncommon. The most probable sources of Fusobacterium bacteremia are the respiratory, the gastrointestinal, and the genitourinary tracts. A.-M. Bourgault et al. (Clin. Infect. Dis. 25[Suppl. 2]:181-183) described 40 patients with Fusobacterium bacteremia; only 3 had Fusobacterium varium, and no one had decubitus scars as the portal of entry. In another published series (S. Henry, A. De Maria, and W. R. McCabe, Am. J. Med. 75:225-231, 1983) of 26 cases,

[Use of clindamycin-2-phosphate in ophthalmology (author's transl)].

Clindamycin-2-phosphate was studied to evaluate its possible use in ophthalmology. 1. Sensitivity of organisms isolated from human clinical materials: Of 44 staphylococcal strains in vitro studied 13 were highly sensitive to 0.1 mug/ml and 28 were resistant to 100 mug/ml or more of clindamycin-2-phosphate. 2. In experiments with rabbits, clindamycin-2-phosphate showed superior penetration into the ocular tissues after intravenous injection than after intramuscular injection. 3. Clindamycin-2-phosphate was administered intramuscularly at the dose of 300 mg to 6 healthy volunteers. The blood con…

[Therapy for severe anaerobic infections with clindamycin (author's transl)].

Ten adult patients with severe Bacteroides infections were treated with 0.9 approximately 1.8 g/day of parenteral or oral clindamycin, and a child was treated with 0.3 g/day orally. Remarkable responses and cures were obtained in all the patients, who had no underlying diseases and pure anaerobic infections; a case of sepsis, two cases of liver abscess, a case of subcutaneous abscess and a case of spinal epidural abscess. The other six patients who had ultimately fatal underlying diseases or mixed infections did not respond well to the combination of clindamycin and the other antibiotics thera…

Intravitreal clindamycin as a therapeutic alternative in severe ocular toxoplasmosis.

Ocular toxoplasmosis is a disease than have severe consequences on the eyesight. The aim of this study article is to present a bilateral ocular toxoplasmosis in a sulfamide allergic patient with unilateral activation and her favourable progression with intravitreal Clindamycin and oral steroids treatment. Weekly intravitreal Clindamycin treatment is shown to be a suitable therapeutic alternative in cases of severe ocular toxoplasmosis and/or in patients with a contraindication to classical treatment. Intravitreal Clindamycin treatment is a safe alternative with favourable clinical results.

Paradoxical effect of clindamycin in experimental, acute toxoplasmosis in cats.

Cats were experimentally inoculated parenterally with the ME49 strain of Toxoplasma gondii to characterize the efficacies of two different dosages of orally administered clindamycin hydrochloride in the treatment of ocular toxoplasmosis. Concentrations of clindamycin hydrochloride at levels previously suggested to be inhibitory to T. gondii replication in vitro were achieved in the serum and aqueous humor but not in the cerebrospinal fluid. Antibiotic therapy, initiated 7 days after inoculation, resulted in no significant difference in the morphometric severity of ocular posterior segment lesi…

A multicentre comparison of clindamycin and metronidazole in the treatment of anaerobic infections.

One hundred and seventy patients with intra-abdominal infection with non-sporing anaerobes were prospectively studied in an international multicentre study. Patients were randomly allocated to treatment with clindamycin or metronidazole, for a minimum of 48 h to a maximum of 7 days. Other antimicrobial therapy was permitted if indicated by in vitro susceptibility testing. The commonest infections were peritonitis, intra-abdominal abscesses and appendicitis (72 cases), colorectal carcinoma (23 cases), intestinal perforation (16 cases) and diverticulitis (13 cases). Thirty patients received no o…

Clindamycin-Induced Dermatitis and the Sparing Phenomenon: A Case Report.

Drug-induced dermatitis is a type IV, T-cell-mediated, hypersensitivity reaction characterized by the development of a rash in response to drug exposure. The sparing phenomenon, where drug-induced rash spares areas of recent skin disease, is a recognized but relatively rare clinical observation. In this report, a 62-year-old woman presented to the emergency department with drug-induced dermatitis secondary to clindamycin that spared a recent site of Pasteurella multocida cellulitis. This case visually highlights the complex interplay between T-cell activation and the local immune environment.

Clindamycin: A Comprehensive Status Report with Emphasis on Use in Dermatology.

Clindamycin is a lincosamide antibiotic that has been used as a topical, oral, or injectable formulation for over five decades. It exhibits a narrow spectrum of microbiologic activity, primarily against gram-positive and anaerobic bacteria. In dermatology, clindamycin has been used primarily as a topical agent, usually for the treatment of acne vulgaris. Despite questions surrounding antibiotic resistance and/or its relative contribution to antibiotic treatment efficacy, a large body of data support the therapeutic value of topical clindamycin for acne vulgaris. As a systemic agent, clindamyci…

Mechanistic Basis for the Translation Inhibition of Cutibacterium acnes by Clindamycin.

Inflammation and the Gram-positive anaerobic bacterium Cutibacterium acnes, which is implicated in acne pathogenesis and pilosebaceous-unit inflammation, are the main targets of antibiotic-based therapy against acne vulgaris (acne). The most widely used antibiotics in acne therapy are tetracyclines, macrolides, and lincosamides. Unfortunately, C. acnes bacteria over the past several decades have demonstrated increased resistance to these antibiotics, particularly to clindamycin. The precise knowledge of how antibiotics interact with their clinical target is needed to overcome this problem. Tow…
